The combina.ion of Ma.hine Lea.ning (ML), sma.t energy meters, a.d a.a.la.ility of household a.plia.ce energy profile da.a.ha. opened new opportunities for Non-Intrusive Loa. Monitoring (NILM). However, the number of options ma.es it cha.lenging in selecting optima. combina.ions for different energy a.plica.ions, which requires studies to exa.ine their *** pa.er contributes one such study tha. investiga.ed four esta.lished ML a.proa.hes – K Nea.est Neighbour (KNN), Support Vector Ma.hine (SVM), eXtreme Gra.ient Boosting (XGBoost) a.d Convolutiona. Neura. Network (CNN) – a.d their performa.ce in cla.sifying a.plia.ce events from a.terna.ing Current (a.) a.d Root Mea. Squa.e (RMS) energy da.a.where the sa.pling frequency a.d tra.ning da.a.et set size wa. va.ied (10 Hz–1 kHz a.d 50–2000 exa.ples per cla.s, respectively). The computa.iona. expense during tra.ning, testing a.d stora.e wa. a.so a.sessed a.d eva.ua.ed with reference to rea.-world *** CNN cla.sifier tra.ned on a. da.a.a. 500 Hz a.d 11,000 exa.ples ga.e the best F1-score (0.989) followed by the KNN cla.sifier (0.940). The stora.e size required by the CNN models wa. 3̃MB, which is very close to fitting on cost-effective embedded system microcontrollers. This would prevent high-ra.e da.a.needing to be sent to the cloud a. a.a.ysis could be performed on edge computing Internet-of-Things (IoT) devices.

Despite a.va.cements in medica. thera.y of Crohn's disea.e(CD), ma.ority of pa.ients with CD will eventua.ly require surgica. intervention, with a. lea.t a.third of pa.ients requiring multiple surgeries. It is importa.t to understa.d the role a.d timing of surgery, with the goa.s of thera.y to reduce the need for surgery without increa.ing the odds of emergency surgery a.d its a.socia.ed morbidity, a. well a. to limit surgica. recurrence a.d a.oid intestina. fa.lure. The profile of CD pa.ients requiring surgica. intervention ha. cha.ged over the deca.es with improvements in medica. thera.y with immunomodula.ors a.d biologica. a.ents. The most common indica.ion for surgery is obstruction from stricturing disea.e, followed by a.scesses a.d fistula.. The risk of ga.trointestina. bleeding in CD is high but the likelihood of needing surgery for bleeding is low. Most ma.or ga.trointestina. bleeding episodes resolve sponta.eously, a.beit the risk of re-bleeding is high. The risk of colorecta. ca.cer a.socia.ed with CD is low. While current surgica. guidelines recommend a.tota. proctocolectomy for colorecta. ca.cer a.socia.ed with CD, subtota. colectomy or segmenta. colectomy with endoscopic surveilla.ce ma. be a.rea.ona.le option. a.proxima.ely 20%-40% of CD pa.ients will need peria.a. surgery during their lifetime. We prepa.ed the DNa.cetyltrimethyl a.monium complex, a. well a. the sa.e complex interca.a.ed with sta.le orga.ic free ra.ica.s, a.d studied their ma.netic properties by electron ma.netic resona.ce (EMR) spectroscopy a.d by mea.uring the ma.netiza.ion on a.superconducting qua.tum interference device (SQUID). The UV-vis a.d CD spectra.of DNa.qua.erna.y a.kyl a.monium complex (DNa.-Q+) in orga.ic solvent clea.ly demonstra.ed tha. it reta.ned the double helica. B-form conforma.ion. The interhelica. spa.ing of double stra.d DNa.(dsDNa. increa.ed when the counter ions (Na.) of phospha.e groups of the na.ura. DNa.were repla.ed with the long a.kyl qua.erna.y a.monium groups. The inter-helica. dista.ce of DNa.cetyltrimethyl a.monium (CTMa. wa. 39.1  a. confirmed by X-ra. diffra.tometry. In genera., the ma.netiza.ion of the DNa.CTMa.complex solid wa. found to be significa.tly lower tha. tha. of na.ura. DNa. Moreover, interca.a.ion of the complex with sta.le orga.ic free ra.ica.s did not improve ma.netiza.ion, which a.a.n wa. in ma.ked contra.t to na.ura. DNa. EMR spectroscopic beha.ior of the complex in the solid sta.e a.so wa. quite different from tha. of na.ura. DNa. The unique broa. EMR signa. of na.ura. DNa.in the low field region with g-va.ue grea.er tha. 10 disa.pea.ed in the DNa.CTMa.complex.

LRP6, a.co-receptor for the morphogen Wnt, a.ds endocytosis of a.thra. complexes. Here we report tha. Dickkopfl （DKK1） protein, a.secreted LRP6 liga.d a.d a.ta.onist, is a.so a.modula.or of a.thra. toxin sensitivity, shRNa.media.ed gene silencing or Ta.EN-media.ed gene knockout of DKK1 reduced sensitivity of cells to Pa.dependent hybrid toxins. However, unlike the solely inhibitory effect on Wnt signa.ing, the effects of DKK1 overexpression on a.thra. toxicity were bidirectiona., depending on its endogenous expression a.d cell context. Fluorescence microscopy a.d biochemica. a.a.yses showed tha. DKK1 fa.ilita.es interna.iza.ion of a.thra. toxins a.d their receptors, a. event media.ed by DKK1-LRP6-Kremen2 complex. Monoclona. a.tibodies a.a.nst DKK1 provided dose-dependent protection to ma.ropha.es from killing by a.thra. letha. toxin （LT）. Our discovery tha. DKK1 forms terna.y structure with LRP6 a.d Kremen2 in promoting Pa.media.ed toxin interna.iza.ion provides a.pa.a.igm for ba.teria. exploita.ion of mecha.isms tha. host cells use to interna.ize signa.ing proteins.
