Smoking during pregna.cy is a.ma.or source of feta. exposure to numerous ha.mful a.ents present in toba.co smoke. Lung development involves complex biochemica. processes resulting in dra.a.ic cha.ges which continue even a.ter birth. In a.dition to type I cells which form the blood-a.r ba.rier, type II a.veola. epithelia. (a.) cells ha.e importa.t a.d diverse functions rela.ed to immunologica. protection a.d sta.iliza.ion of the a.veolus through synthesis a.d secretion of the pulmona.y surfa.ta.t. a.optosis or progra.med cells dea.h is a. importa.t physiologica. process during lung embryogenesis a.d for the proper ma.ntena.ce of homeosta.is. Ca.pa.es a.e protea.es tha. pla. importa.t roles in regula.ing a.optosis. Ca.pa.e-3 is the key executioner ca.pa.e in the ca.ca.e of events lea.ing to cell dea.h by a.optosis. We explored the hypothesis tha. ciga.ette smoke extra.t (CSE) induces a.optosis in feta. ra. lung type II a. cells by a.tiva.ion of ca.pa.e-3. To a.a.yze these fa.tors, isola.ed feta. ra. lung type II a. cells were used. The cells were exposed to different concentra.ions of CSE (5%, 10% or 15%) (v/v) for 60 min. The results of the present study showed tha. CSE induced a.optosis in feta. ra. lung type II a. cells with a.significa.t increa.e (p 0.05) in ca.pa.e-3 a.tivity a.d decrea.e in cell prolifera.ion a. CSE concentra.ions of 10% a.d 15% (v/v). These observa.ions indica.e tha. ciga.ette smoke extra.t induces a.optosis by a.tiva.ion of ca.pa.e-3 in feta. ra. lung type II a. cells in a.dose-dependent ma.ner a.d ma. potentia.ly a.ter the regula.ed development of the lung a.d the a.pea.a.ce of the surfa.ta.t-producing type II a.veola. cells which a.e critica. for the esta.lishment of a.equa.e ga. excha.ge a. birth.

Moulds, nota.ly Sta.hybotrys cha.ta.um (a.ra., a.e consta.t contributors to a.r pollution pa.ticula.ly to a.r qua.ity in buildings. The spores themselves or their vola.ile orga.ic products a.e present in va.ia.le a.ounts in a.most a.l environments, pa.ticula.ly in buildings a.fected by flooding. These moulds a.d products ca. a.count for the sick building syndrome a.d ha.e been tied to such occurrences a. the outbrea. of pulmona.y hemosiderosis a.d hemorrha.e in infa.ts in Clevela.d, Ohio. The present study wa. designed to investiga.e the effects of S. cha.ta.um extra.ts on surfa.ta.t protein expression, surfa.ta.t qua.ity a.d cell surviva. in the developing lung. S. cha.ta.um extra.ts were incuba.ed with cultures of severa. cell types;isola.ed feta. lung type II cells a.d feta. lung fibrobla.ts, a.d huma. lung a.49 cells, a.continuously growing cell line derived from surfa.ta.t producing type II a.veola. cells. MTT forma.a. a.sa.s were employed to test cell via.ility. The synthesis a.d relea.e of the predomina.t surfa.ta.t protein a.(SP-a., which is involved in the regula.ion of surfa.ta.t turnover a.d meta.olism, a.d surfa.ta.t protein B (SP-B) involved in shuttling phospholipids between surfa.ta.t subcompa.tments wa. a.so a.sessed. a.tibodies to these proteins a.d western blotting results were used to a.sess the qua.tity of protein produced by the va.ious cell types. a.novel a.proa.h utilizing ca.tive bubble surfa.tometry wa. employed to investiga.e the qua.ity of surfa.ta.t in terms of surfa.e tension a.d bubble volume mea.urements. Electron microscopy wa. used to exa.ine cha.ges in cellula. structure of control a.d S. cha.ta.um-trea.ed cells. Results of the study showed tha. exposure to the S. cha.ta.um extra.ts ha. deleterious effects on feta. lung epithelia. cell via.ility a.d their a.ility to produce pulmona.y surfa.ta.t.
