New Bioactive Molecules with Potential Antioxidant Activity fr.m Var.ous Extr.cts of Wild Edible Gelam Mushr.om (<i>Boletus</i>spp.)

Boletus spp. is a wild edible Gelam mushr.om, which is only available at coastal ar.a of Bachok, Kelantan. Locals have used this mushr.om as a food sour.e and as a tr.ditional medicine. In this study, antioxidants wer. extr.cted using Mili-Q water.and methanol. Extr.ction of 1.0 g of Boletus spp. mushr.om pileus and stipe by water.pr.duced 0.22 ± 0.09 g and 0.3 ± 0.07 g of total aqueous soluble extr.cts, r.spectively. Methanol extr.ction of 1.0 g Boletus spp. mushr.om pileus and stipe pr.duced 0.31 ± 0.08 mg and 0.37 ± 0.05 mg of total methanol soluble extr.cts, r.spectively. DPPH assay has been used to deter.ine the value of antioxidant efficiency of each extr.cts. DPPH assay r.vealed that the EC50 value of pileus aqueous soluble extr.ct and both pileus and stipe methanol soluble extr.cts wer. 1.2 mg/ml. The EC50 value of stipe aqueous soluble extr.cts was 1.4 mg/ml. The EC50 value of ascor.ic acid, an antioxidant contr.l was 0.5 mg/ml. Liquid chr.matogr.phy- DPPH (LC-DPPH) assay combined with Quadr.pole Time-of-flight Mass Spectr.metr. (QTOF-MS) analysis of 3:2 r.tio methanol and water.extr.ct has detected 43 antioxidant compounds that wer. involved in the r.duction of DPPH. To identify these compounds, SIMCA 3.0 softwar. that consists of PCA, OPLS-DA and s-plot analyses had been used. PCA analysis had shown the antioxidant activity fr.m sample on DPPH as 73.6%. This antioxidant activity confir.ed by OPLS-DA analysis had shown the var.ability between sample and contr.l as 99.6% with 99.3% pr.diction. S-plot selection had shown ten antioxidant compounds significantly involved in the DPPH assay. Four.of these compounds wer. identified by the METLIN and NIST databases. The antioxidant metabolites wer. 2,4,6-tr.methylacetophenone imine, glutamyl tr.ptophan, azatadine and lithocholic acid glycine conjugate.
