Bovine Herpesvirus-1 (BoHV-1) is a DNA virus belonging to the family Herpesviridae, subfamily Alfaherpesvirinae; it is a worldwide pathogen, causing serious economic losses in livestock. In Colombia there have been multiple isolates of BoHV-1 that have been subjected to molecular characterization, classifying most of the country isolates as BoHV-1.1. In the present study we developed and evaluated an ethyleneimine binary inactivated isolate from the native BoHV-1 strain (Córdoba-2) in a rabbit model of vaccination and infection. The vaccine was evaluated in two phases, one of immunogenicity with vaccination and a booster after 21 days, and an evaluation phase of protection against challenge with a highly virulent reference strain. The results demonstrate optimum serum-conversion, with protective neutralizing antibody titers 28 days post vaccination and optimal protection against challenge with the reference strain with decreased clinical signs of infection, protection against the onset of fever and decrease of virus excretion post challenge. In conclusion, our results show the enormous potential that an immunogenic inactivated vaccine has produced from the native BoHV-1.1 strain, which produces a high antigen mass to the vaccine to induce optimal immunity and protection, and it is a strong candidate for evaluation and possible future use in different cattle populations.

Bovine Herpesvirus-1 (BoHV-1) is distributed worldwide and is a major pathogen in cattle, being the causal agent of a variety of clinical syndromes. The aim of this study was to isolate and to characterize (molecular and biological characterization) BoHV-1 from 29 immunosuppressed animals. It was possible to obtain 18 isolates, each from a different animal, such as from the respiratory and reproductive tracts. In some cases the cytopathic effect was visible 12 hours post-inoculation, and became characteristic after 36-48 hours. Biological characteristics were evaluated and compared with Iowa and Colorado-1 reference strains, and differences were found in plaque size, virus titer measured by TCID50 and PFU/mL, and one step virus curves. These results showed that some isolates had a highly virulent-like behavior in vitro, compared to the reference strains, with shorter eclipse periods, faster release of virus into the supernatants, and higher burst size and viral titer. There were no differences in glycoprotein expression of BoHV-1 isolates, measured by Western blot on monolayers. Moreover, using restriction endonucleases analysis, most of the viruses were confirmed as BoHV-1.1 and just one of them was confirmed as BoHV-1.2a subtype. These findings suggest that some wild-type BoHV-1 isolates could be useful as seeds to develop new monovalent vaccines.

Effect of treatment failure on the Cag A EPIYA motif in Helicobacter pylori strains from Colombian subjects

AIM To evaluate effect of treatment failure on cag A and vac A genotypes in Helicobacter pylori(H. pylori) isolates from *** One hundred and seventy-six participants infected with H. pylori from Colombia were treated during 14 d with the triple-standard therapy. Six weeks later, eradication was evaluated by 13C-Urea breath test. Patients with treatment failure were subjected to endoscopy control; biopsies obtained were used for histopathology and culture. DNA from H. pylori isolates was amplified using primers specific for cag A and vac A genes. The phylogenetic relationships among isolates obtained before and after treatment were established by conglomerate analysis based on random amplified polymorphic DNA(RAPD) *** Treatment effectiveness was at 74.6%. Of the par-ticipants with treatment failure, 25 accepted subjected to a second endoscopy. Prevalence of posttreatment infection was 64%(16/25) and 40%(10/25) by histology and culture, respectively. Upon comparing the cag A and vac A genotypes found before and after therapy, multiple cag A genotypes(cag A-positive and cag A-negative) were found before treatment; in contrast, cag A-negative genotypes decreased after treatment. vac A s1m1 genotype was highly prevalent in patients before and after therapy. The 3'cag A region was successfully amplified in 95.5%(21/22) of the isolates obtained before and in 81.8%(18/22) of the isolates obtained after treatment. In the isolates obtained from patients with treatment failure, it was found that 72.7%(16/22) presented alterations in the number of EPIYA motifs, compared to isolates found before *** Unsuccessful treatment limits colonization by lowvirulence strains resulting in partial and selective eradication in mixed infections, and acts on the cag A-positive strains inducing genetic rearrangements in cag A variable region that produces a loss or gain of EPIYA repetitions.

AIM To compare the genomic variability and the multiple colonization of Helicobacter pylori(H. pylori) in patients with chronic gastritis from two Colombian populations with contrast in the risk of developing gastric cancer(GC): Túquerres-Nari?o(High risk) and Tumaco-Nari?o(Low risk).METHODS Four hundred and nine patients from both genders with dyspeptic symptoms were studied. Seventy-two patients were included in whom H. pylori was isolated from three anatomic regions of the gastric mucosa,(31/206) of the high risk population of GC(Túquerres) and(41/203) of the low risk population of GC(Tumaco). The isolates were genotyped by PCR-RAPD. Genetic diversity between the isolates was evaluated by conglomerates analysis and multiple correspondence analyses. RESULTS The proportion of virulent genotypes of H. pylori was 99% in Túquerres and 94% in Tumaco. The coefficient of similarity of Nei-Li showed greater genetic diversityamong isolates of Túquerres(0.13) than those of Tumaco(0.07). After adjusting by age, gender and type of gastritis, the multiple colonization was 1.7 times more frequent in Túquerres than in Tumaco(P = 0.05).CONCLUSION In Túquerres, high risk of GC there was a greater probability of multiple colonization by H. pylori. From the analysis of the results of the PCR-RAPD, it was found higher genetic variability in the isolates of H. pylori in the population of high risk for the development of GC.

In biology, cancer is the most beautiful natural model of a chaotic system, which under uncontrolled proliferations, generates extreme disorder that finally causes intercellular collisions. The authors have described and documented fractal self-assembly of geometric triangular chiral hexagonal crystal-like complex organizations (GTCHC) and interface comet tail effect patterns in cancer processes. According to this novel observation cancer incorporates a real visualization world with a great surprising finding in biology, physics, and geology. This visualization platform literally allows us to see what would otherwise remain completely invisible. From theory to practice this irreducible geometric matrix allowed us to identify in geology, real measurable green infrared-electromagnetic stripe line in interface with hexagonal geomorphic pattern, triangular chiral pyramidal rock structures, geology well defined mirror images, template platform to bio signature characterization of ancestral primitive polar head-tail organization, embryoid and human-like shape pattern embedded as giant fossils in rocks that have never been seen before. Electromagnetic field released in collision-impact events generate in the matrix interphase fractal scalable invariant order of geometric triangular chiral hexagonal structures. The laws of biology and geology can finally be redirected to the laws of physics;specifically magnetic fields create a new kind of classification based on these fractal structural similarities of the relationship. Further interdisciplinary collaboration must be carried out to study these geometric self-assembly geological structures, ancient sediments and rocks that could provide insights into antecedents of life.

The shroud continues to remain one of the most studied and controversial artifacts in human history. Many tests, X-ray fluorescence, reflectance, spectrometry and low energy/high-resolution X ray transmission have shown that the crucified body is not compatible with a painted image. Researchers confirm that the alleged blood is real blood. We documented the self-assembly of geometric triangular chiral hexagon complex (GTCHC) with structural organization of embryoid bodies in cancer tissues. The identification of these structures is not only limited to malignant tumors but also appears in extreme injured tissues. Our interest is to determine if we can predict and identify these patterns in the Shroud of Turin. Based on pattern recognition image was analyzed over 100 shroud images. We identified a central spectral emission line that exhibits a characteristic signature on a plot of residual electromagnetic radiation, head area narrowing and low extremities broadening, indication of decay energy changes in the velocity of the molecules in the traversal trajectory. This Electromagnetic collision event generates in the cloth stagnant blood areas with patterns identical to those identified for us in cancer damage tissues. Inflammatory cytokines activate stem cells and Notch signaling proteins in cascade of interactions to generate real clonal human embryoid template. Can we predict function from structure? These structures evoke life, regeneration, but not death. Our findings suggest the image of a crucified man on the Shroud of Turin is a real physical electromagnetic collision event in response to extreme tissue injury, with the fact that supports our previous findings in cancer tissues as real and predictable. Proteins derived from these emergent damage tissue derivate stem cells could be used to design biologic templates in regenerative medicine and develop novel strategies in cancer therapy.

Porcine respiratory disease complex(PRDC) is a serious health problem that mainly affects growing and finishing pigs. PRDC is caused by a combination of viral and bacterial agents, such as porcine reproductive and respiratory syndrome virus(PRRSV), swine influenza virus(SIV), Mycoplasma hyopneumoniae(Myh), Actinobacillus pleuropneumoniae(APP), Pasteurella multocida and Porcine circovirus 2(PCV2). To characterize the specific role of swine influenza virus in PRDC presentation in Colombia, 11 farms from three major production regions in Colombia were examined in this study. Nasal swabs, bronchial lavage and lung tissue samples were obtained from animals displaying symptoms compatible with SIV. Isolation of SIV was performed in 9-day embryonated chicken eggs or Madin-Darby Canine Kidney(MDCK) cells. Positive isolates, identified via the hemagglutination inhibition test, were further analyzed using PCR. Overall, 7 of the 11 farms were positive for SIV. Notably, sequencing of the gene encoding the hemagglutinin(HA) protein led to grouping of strains into circulating viruses identified during the human outbreak of 2009, classified as pandemic H1N1-2009. Serum samples from 198 gilts and multiparous sows between 2008 and 2009 were obtained to determine antibody presence of APP, Myh, PCV2 and PRRSV in both SIV-H1N1p-negative and-positive farms, but higher levels were recorded for SIV-H1N1p-positive farms. Odds ratio(OR) and P values revealed statistically significant differences(p<0.05) in PRDC presentation in gilts and multiparous sows of farms positive for SIV-H1N1 p. Our findings indicate that positive farms have increased risk of PRDC presentation, in particular, PCV2, APP and Myh.

The formation of polyacids in a glass-ionomer cement was computationally modeled. The polyacid modelled is an oligomer made up of three molecules: acrylic acid, itaconic acid and an additional molecule that acts as a spacer group between the previous two. Acryloyl and meta-acryloyl derivatives of some amino acids and related molecules such as N-vinyl pyrrolidone and N-vinyl caprolactam were used as spacer groups. Some of these molecules have been previously used experimentally as spacer groups. In this work the spacer behaviour of 15 different molecules was investigated. To our knowledge, this is the first computational attempt to model some promising molecules to be incorporated into glass-ionomer dental cements. The results revealed the best structural arrangement for the investigated molecules.

The carbon isotope discrimination （δ^13C） of leaves has been shown to be correlated with the transpiration efficiency of leaves in a wide range of species. This has led to δ^13C being used in breeding programs to select for improved transpiration efficiency. The correlation between δ^13C and transpiration efficiency was determined under well-watered conditions during the vegetative phase in six genotypes of lentil （Lens culinaris Medikus）, six genotypes of chickpea （Cicerarietinum L.） and 10 cultivars of narrow-leafed lupin （Lupinus angustifolius L.）. Biomass （dry matter） accumulation and water use （transpiration） varied among the genotypes in all three species and transpiration efficiency was 40% to 75% higher in the most efficient compared with the least efficient genotypes. However, δ^13C and transpiration efficiency were not significantly correlated in any of the species. This suggests that the δ^13C technique cannot be used in selection for transpiration efficiency in the three grain legumes （pulses） studied.
