Rigidity(or stiffness)of materials and extracellular matrix has proven to be one of the most significant extracellular physicochemical cues that can control diverse cell behaviors,such as contractility,motility,and spreading,and the resultant pathophysiological *** 2D materials engineered with tunable rigidity have enabled researchers to elucidate the roles of matrix biophysical cues in diverse cellular events,including migration,lineage specification,and mechanical ***,the recent findings accumulated under 3D environments with viscoelastic and remodeling properties pointed to the importance of dynamically changing rigidity in cell fate control,tissue repair,and disease ***,here we aim to highlight the works related with material/matrix-rigidity-mediated cell and tissue behaviors,with a brief outlook into the studies on the effects of material/matrix rigidity on cell behaviors in 2D systems,further discussion of the events and considerations in tissue-mimicking 3D conditions,and then examination of the in vivo findings that concern material/matrix *** current discussion will help understand the material/matrix-rigidity-mediated biological phenomena and further leverage the concepts to find therapeutic targets and to design implantable materials for the treatment of damaged and diseased tissues.

Nephrotic syndrome(NS)is a kidney disease characterized by hypertriglyceridemia,massive proteinuria,hypo-albuminemia and peripheral ***-gamibang(SKHGMB)was recorded in a traditional Chinese medical book named“Bangyakhappyeon(方藥合編)”and its three prescriptions Sinkihwan,Geumgwe-sinkihwan,and Jesaeng-sinkihwan belong to *** study confirmed the effect of SKHGMB on renal dysfunction in an NS model induced by puromycin aminonucleoside(PAN).The experimental NS model was induced in male Sprague Dawley(SD)rats through injection of PAN(50 mg·kg^(-1))via the femoral *** not only reduced the size of the kidneys increased due to PAN-induced NS,but also decreased proteinuria and *** addition,SKHGMB significantly ameliorated creatinine clearance,creatinine,and blood urea *** relieved glomeruli dilation and tubules fibrosis in the glomeruli of the NS *** inhibited the protein and mRNA levels of the NLRP3 inflammasome including NLRP3,ASC,and pro-caspase-1 in NS *** reduced the protein and mRNA levels of fibrosis regulators in NS *** results indicated that SKHGMB exerts protective effects against renal dysfunction by inhibiting of renal inflammation and fibrosis in NS rats.

Background:Germ cell mitotic arrest is conserved in many vertebrates,including birds,although the time of entry or exit into quiescence phase *** arrest is essential for the normal differentiation of male germ cells into spermatogonia and accompanies epigenetic reprogramming and meiosis inhibition from embryonic development to ***,mitotic arrest was not well studied in chickens because of the difficulty in obtaining pure germ cells from relevant developmental ***:We performed single-cell RNA sequencing to investigate transcriptional dynamics of male germ cells during mitotic arrest in DAZL::GFP *** differentially expressed gene analysis and K-means clustering to analyze cells at different developmental stages(E12,E16,and hatch),we found that metabolic and signaling pathways were regulated,and that the epigenome was reprogrammed during mitotic *** particular,we found that histone H3K9 and H3K14 acetylation(by HDAC2)and DNA demethylation(by DNMT3B and HELLS)led to a transcriptionally permissive chromatin ***,we found that global DNA demethylation occurred gradually after the onset of mitotic arrest,indicating that the epigenetic-reprogramming schedule of the chicken genome differs from that of the mammalian *** hypomethylation persisted after hatching,and methylation was slowly re-established 3 weeks ***:We found a unique epigenetic-reprogramming schedule of mitotic-arrested chicken prospermatogonia and prolonged hypomethylation after *** will provide a foundation for understanding the process of germ-cell epigenetic regulation in several species for which this process is not clearly *** findings on the biological processes related to sex-specific differentiation of prospermatogonia could help studying germline development in vitro more elaborately.

Electrical conductivity(EC)of soil-water extracts is commonly used to assess soil ***,its conversion to the EC of saturated soil paste extracts(ECe),the standard measure of soil salinity,is currently required for practical *** many regression models can be used to obtain ECe from the EC of soil-water extracts,the application of a site-specific model to different sites is not straightforward due to confounding soil factors such as soil *** study was conducted to develop a universal regression model to estimate a conversion factor(CF)for predicting EC_(e) from EC of soil-water extracts at a 1:5 ratio(EC_(1:5)),by employing a site-specific soil texture(i.e.,sand content).A regression model,CF=8.9105e^(0.0106sand)/1.2984(r^(2)=0.97,P<0.001),was developed based on the results of coastal saline soil surveys(n=173)and laboratory experiments using artificial saline soils with different textures(n=6,sand content=10%-65%)and salinity levels(n=7,salinity=1-24 dS m^(-1)).Model performance was validated using an independent dataset and demonstrated that EC_(e) prediction using the developed model is more suitable for highly saline soils than for low saline *** feasibility of the regression model should be tested at other *** soil factors affecting EC conversion factor also need to be explored to revise and improve the model through further studies.

BACKGROUND Patients with Barcelona clinic liver cancer(BCLC)stage B hepatocellular carcinoma(HCC)are considerably heterogeneous in terms of tumor burden,liver function,and performance *** improve the poor survival outcomes of these patients,treatment approaches other than transarterial chemoembolization(TACE),which is recommended by HCC guidelines,have been adopted in realworld clinical *** hypothesize that this non-adherence to treatment guidelines,particularly with respect to the use of liver resection,improves survival in patients with stage B *** To assess guideline adherence in South Korean patients with stage B HCC and study its impact on *** A retrospective analysis was conducted using data from 2008 to 2016 obtained from the Korea Central Cancer *** with stage B HCC were categorized into three treatment groups,guideline-adherent,upward,and downward,based on HCC guidelines recommended by the Asian Pacific Association for the Study of the Liver(APASL),the European Association for the Study of the Liver(EASL),and the American Association for the Study of Liver Diseases(AASLD).The primary outcome was HCC-related deaths;tumor recurrence served as the secondary *** among the groups was compared using the Kaplan-Meier method and the log-rank *** of survival outcomes were identified using multivariable Cox regression *** In South Korea, over the study period from 2008 to 2016, a notable trend was observed in adherence to HCCguidelines. Poly Adenylate Binding Protein Interacting protein 1(PAIP1) plays a critical role in translation initiation and is associated with the several cancer types. However, its function and clinical significance have not yet been described in oral squamous cell carcinoma(OSCC) and its associated features like lymph node metastasis(LNM). Here, we used the data available from Gene Expression Omnibus(GEO), The Cancer Genome Atlas(TCGA), and Clinical Proteomic Tumor Analysis Consortium(CPTAC) to analyze PAIP1 expression in oral cancer. The publicly available data suggests that PAIP1 mRNA and protein levels were increased in OSCC. The high PAIP1 expression was more evident in samples with advanced stage, LNM, and worse pattern of invasion. Moreover, the in vitro experiments revealed that PAIP1 knockdown attenuated colony forming, the aggressiveness of OSCC cell lines, decreasing MMP9 activity and SRC phosphorylation. Importantly, we found a correlation between PAIP1 and pSRC through the analysis of the IHC scores and CPTAC data in patient samples. Our findings suggest that PAIP1 could be an independent prognostic factor in OSCC with LNM and a suitable therapeutic target to improve OSCC patient outcomes.

Nitric oxide(NO)has been shown to promote revascularization and nerve regeneration after peripheral nerve ***,in vivo application of NO remains challenging due to the lack of stable carrier materials capable of storing large amounts of NO molecules and releasing them on a clinically meaningful time ***,a silica nanoparticle system capable of reversible NO storage and release at a controlled and sustained rate was *** this study,NO-releasing silica nanoparticles(NO-SNs)were delivered to the peripheral nerves in rats after acute crush injury,mixed with natural hydrogel,to ensure the effective application of NO to the *** using a polymer dye and immunohistochemical staining for the detection of CD34(a marker for revascularization)results showed that NO-releasing silica nanoparticles increased revascularization at the crush site of the sciatic *** sciatic functional index revealed that there was a significant improvement in sciatic nerve function in NO-treated *** and anatomical analyses showed that the number of myelinated axons in the crushed sciatic nerve and wet muscle weight excised from NO-treated rats were ***,muscle function recovery was improved in rats treated with *** together,our results suggest that NO delivered to the injured sciatic nerve triggers enhanced revascularization at the lesion in the early phase after crushing injury,thereby promoting axonal regeneration and improving functional recovery.
