OBJECTIVE:To investigate the optimal timing and underlying mechanism of electroacupuncture(EA)at Baihui(GV 20)and Dazhui(GV 14)for improved long-term functional recovery after focal cerebral ischemia in a photothrombotic stroke mouse ***:Totally 50 adult male C57 BL/6 J mice were assigned into 5 groups:(a)the control group,sham-operated mice(n=10);(b)the vehicle group,focal cerebral ischemia induction without EA(n=10);(c)the acute EA group,mice received EA immediately post-ischemia,followed by once-daily treatments for 7 consecutive days(n=10);(d)the subacute EA group,mice received EA 4 days post-ischemia,followed by once-daily treatments for 7 consecutive days(n=10);(e)the delayed EA *** stimulation(2 Hz,2 V for 20 min)was applied to acupuncture points(acupoints),Baihui(GV 20)and Dazhui(GV 14),once a day for 7 consecutive days beginning immediately(acute treatment),4 d(subacute treatment)and 10 d(delayed treatment)after focal cerebral ischemia in C57 BL/6 J *** assessments were conducted 21 and 28 d post-ischemia and histopathological analyses were performed 28 days ***:The subacute EA treatment at Baihui(GV20)and Dazhui(GV 14)significantly improved functional recovery compared to the vehicle group 28 d after ischemic brain injury,although brain atrophy was not *** number of Neu N+and Neu N+/Brd U+cells as well as GFAP intensity in the ipsilateral cortex were significantly increased in the subacute group compared to the vehicle group 28 d *** concluded that EA stimulation 4 d post-ischemia(subacute treatment)enhanced neurogenesis and astrogliosis,likely contributing to long-term functional recovery following focal cerebral ***:Our findings suggest that the timing of the EA therapy at Baihui(GV 20)and Dazhui(GV 14)determines the therapeutic effects in mice with focal cerebral ischemia induced by photothrombotic occlusion.

Gastric cancer(GC)is a pervasive global malignancy with high mortality rates due to distant metastasis[1].GC metastasis can occur via hematogenous route,peri-toneal route,and specifically through ovarian spread in females[2].Among these,peritoneal metastasis is the most prevalent and challenging condition to treat[3].The Cancer Genome Atlas(TCGA)has uncovered four molec-ular subtypes of GC:microsatellite instability,Epstein-Barr virus-related,chromosomal instability,and genomically stable[4].

Thioredoxin Reductase Type C （NTRC） Orchestrates Enhanced Thermotolerance to Arabidopsis by Its Redox-Dependent holdase Chaperone Function

Genevestigator analysis has indicated heat shock induction of transcripts for NADPH-thioredoxin reduc-tase, type C （NTRC） in the light. Here we show overexpression of NTRC in Arabidopsis （NTRC°E） resulting in enhanced tolerance to heat shock, whereas NTRC knockout mutant plants （ntrcl） exhibit a temperature sensitive phenotype. To investigate the underlying mechanism of this phenotype, we analyzed the protein＇s biochemical properties and protein structure. NTRC assembles into homopolymeric structures of varying complexity with functions as a disulfide reductase, a foldase chaperone, and as a holdase chaperone. The multiple functions of NTRC are closely correlated with protein structure.. Complexes of higher molecular weight （HMW） showed stronger activity as a holdase chaperone, while low molecular weight （LMW） species exhibited weaker holdase chaperone activity but stronger disulfide reductase and fol-dase chaperone activities. Heat shock converted LMW proteins into HMW complexes. Mutations of the two active site Cys residues of NTRC into Ser （C217/454S-NTRC） led to a complete inactivation of its disulfide reductase and foldase chaperone functions, but conferred only a slight decrease in its holdase chaperone function. The overexpression of the mutated C217/454S-NTRC provided Arabidopsis with a similar degree of thermotolerance compared with that of NTRC°E plants. however, after prolonged incubation under heat shock, NTRC°E plants tolerated the stress to a higher degree than C217/454S-NTRC°E plants. The results suggest that the heat shock-mediated holdase chaperone function of NTRC is responsible for the increased thermotolerance of Arabidopsis and the activity is significantly supported by NADPH.

Ecological evaluation of marine macroalgal communities on five islands of Korea in the Yellow Sea

Macroalgae have long been used as biological indicators of marine ecosystem health worldwide due to their ecological importance and sensitivity to environmental stress.A number of previous studies have utilized macroalgal communities in monitoring surveys of environmental *** study examined the characteristics and patterns of marine macroalgal communities in the Yellow Sea off the western coast of *** were analyzed for the number of species,biomass,and coverage ratio by macroalgal *** the study period,82 macroalgal species(10 green algae,17 brown algae,and 55 red algae)were identified at the five study sites,with the highest number of species found at Gwanrido and Uido(both containing 41 species)and the lowest at Daeijakdo(27 species).The average biomass(via dry weight)was 98.63 g/m^(2),consisting of green algae(8.39 g/m^(2)),brown algae(35.08 g/m^(2)),and red algae(55.16 g/m^(2)).The dominant macroalgae species in terms of biomass were Corallina pilulifera,Sargassum thunbergii,and Ulva australis in the intertidal zones,and Botryocladia wrightii and Gelidium elegans in the subtidal ***,evenness,and diversity indices based on the biomass of abundant species were 5.08,0.65,and 2.30,respectively,over the entire study *** on the evaluation of the environmental states by the community indices,overall,the Ecological Evaluation Index of macroalgae communities in the study area was marked as“Good-Moderate”,but was determined as“ModerateLow”at several sites during *** results can be a direct approach in the assessment of coastal habitats in which anthropogenic as well as climate change influences persist.

Preparation of human decellularized peripheral nerve allograft using amphoteric detergent and nuclease

Animal studies have shown that amphoteric detergent and nuclease(DNase I and ribonuclease A) is the most reliable decellularization method of the peripheral nerve. however, the optimal combination of chemical reagents for decellularization of human nerve allograft needs further investigation. To find the optimal protocol to remove the immunogenic cellular components of the nerve tissue and preserve the basal lamina and extracellular matrix and whether the optimal protocol can be applied to larger-diameter human peripheral nerves, in this study, we decellularized the median and sural nerves from the cadavers with two different methods: nonionic and anionic detergents(Triton X-100 and sodium deoxycholate) and amphoteric detergent and nuclease(3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate(CHAPS), deoxyribonuclease I, and ribonuclease A). All cellular components were successfully removed from the median and sural nerves by amphoteric detergent and nuclease. Not all cellular components were removed from the median nerve by nonionic and anionic detergent. Both median and sural nerves treated with amphoteric detergent and nuclease maintained a completely intact extracellular matrix. Treatment with nonionic and anionic detergent decreased collagen content in both median and sural nerves, while the amphoteric detergent and nuclease treatment did not reduce collagen content. In addition, a contact cytotoxicity assay revealed that the nerves decellularized by amphoteric detergent and nuclease was biocompatible. Strength failure testing demonstrated that the biomechanical properties of nerves decellularized with amphoteric detergent and nuclease were comparable to those of fresh controls. Decellularization with amphoteric detergent and nuclease better remove cellular components and better preserve extracellular matrix than decellularization with nonionic and anionic detergents, even in large-diameter human peripheral nerves. Clinicopathological features of patients with middle third gastric carcinoma

AIM: To compared the prognosis of middle third gastric carcinoma(MGC) patients with those of patients with proximal/distal gastric carcinoma(PGC/DGC).METhoDS: Of 3299 patients diagnosed with gastric carcinoma who underwent surgery at our hospital over a 15-year period, 919(27.9%) were diagnosed with MGC. For each patient, the following information was obtained from hospital records: Age, sex, tumor size, depth of invasion, histologic type, nodal involvement, extent of lymph node dissection, hepatic metastasis, peritoneal dissemination, stage at initial diagnosis, operative type, curability, and survival ***: T1 category tumors were more common in patients with MGC than in patients with PGC(P howed that age, tumor size, serosal invasion, lymph node metastasis, and curability were significant predictors of survival in patients with MGC. The survival rate for MGC patients was similar to that for PGC/DGC patients(52.8% vs 44.4%/51.4%, P = 0.1138). The 5-year survival rate for MGC patients with curative resection was higher than that for MGC patients with non-curative resection(62.9% vs 8.7%, P < 0.001).CONCLUSION: These results indicate that tumor location did not affect the prognosis. Curative resection is important for improving the prognosis of patients with MGC.

Application of direct laser metal tooling for AISI H13 tool stee

In the die industry, it is commonly agreed that residual tool life can be successfully extended by timely repair of damaged surfaces. Traditionally, the main repair process is tungsten inert gas (TIG) welding, but a new process called direct laser metal tooling (DLMT) emerges. DLMT is a manual process, of which results depend on the materials of the powders and tools, the laser process and parameters. This technology is a direct-metal freeform fabrication technique in which a 200 W fiber laser is used. AISI H13 tool steel is a suitable material for die casting tools because of the high resistance to thermal fatigue and dimensional stability. In this research, AISI H13 tool steel was melted with metal powder by fiber laser. Before melting AISI H13, the powders were analyzed with XRF equipment. Then, hardness distribution of laser melted zone was investigated. The microstructure in laser melted zone was discussed. In order to identify the effect of particle size of powder on the melted zone, two types of particle sizes of powders were used. Experimental results show that the mold repair process using DLMT can be applied in the mold repair industry.

Effect of Laser Parameters on Sintered Powder Morphology

Selective laser sintering is a kind of rapid prototyping process whereby a three-dimensional part is built layerwise by laser scanning a powder. This process is highly inffuenced by powder and laser parameters such as laser power, scan rate, spot size and layer thickness. Therefore a study on fabricating a line with Fe-Ni-Cr powder on AISI H13 tool steel has been performed by selective laser sintering. In this study, fabrication was performed by experimental facilities consisting of a 200W fiber laser which can be focused to 0.08 mm and atmospheric chamber which can control atmospheric pressure with argon. The line was fabricated with various laser power, scan rate and layer thickness. Line width and surface quality were investigated. With power increase or layer thickness decrease, line width was decreased and line surface quality was improved with scan rate optimization.

An attempt was made to improve the surface hardness and wear properties of AISI H13 tool steel through solid solution hardening and refinement of microstructures using a 200 W fiber laser as a heat generating *** hardness of laser melted zone was *** order to identify the effect of heat input on the laser melting zone,scanning conditions were *** results show that,the hardness of as-received AISI H13 tool steel is approximately Hv 240,and the hardness after laser surface heat treatment is around Hv *** hardening depth and width are increased with the increase in the heat input *** of experimental results will be considered in tooling industry.
