KoNA:Korean Nucleotide Archive as A New Data Repository for Nucleotide Sequence Data

During the last decade,the generation and accumulation of petabase-scale high-throughput sequencing data have resulted in great challenges,including access to human data,as well as transfer,storage,and sharing of enormous amounts of *** promote data-driven biological research,the Korean government announced that all biological data generated from government-funded research projects should be deposited at the Korea BioData Station(K-BDS),which consists of multiple databases for individual data ***,we introduce the Korean Nucleotide Archive(KoNA),a repository of nucleotide sequence *** of July 2022,the Korean Read Archive in KoNA has collected over 477 TB of raw next-generation sequencing data from national genome *** ensure data quality and prepare for international alignment,a standard operating procedure was adopted,which is similar to that of the International Nucleotide Sequence Database *** standard operating procedure includes quality control processes for submitted data and metadata using an automated pipeline,followed by manual *** ensure fast and stable data transfer,a high-speed transmission system called GBox is used in ***,the data uploaded to or downloaded from KoNA through GBox can be readily processed using a cloud computing service called *** seamless coupling of KoNA,GBox,and Bio-Express enhances the data experience,including submission,access,and analysis of raw nucleotide *** not only satisfies the unmet needs for a national sequence repository in Korea but also provides datasets to researchers globally and contributes to advances in *** KoNA is available at https://***/kona/.

Inflammation is a primary defense process against various extracellular stimuli,such as viruses,pathogens,foods,and environmental *** cells respond to stimuli for short periods of time,it results in acute or physiological ***,if the stimulation is sustained for longer time or a pathological state occurs,it is known as chronic or pathological *** studies have shown that tumorigenesis in the gastrointestinal (GI) tract is closely associated with chronic inflammation,for which abnormal cellular alterations that accompany chronic inflammation such as oxidative stresses,gene mutations,epigenetic changes,and inflammatory cytokines,are shared with carcinogenic processes,which forms a critical cross-link between chronic inflammation and *** growth factor (TGF)-β is a multi-potent cytokine that plays an important role in regulation of cell growth,apoptosis and *** importantly,TGF-β is a strong anti-inflammatory cytokine that regulates the development of effector ***-β has a suppressive effect on carcinogenesis under normal conditions by inhibiting abnormal cell growth,but on the other hand,many GI cancers originate from uncontrolled cell growth and differentiation by genetic loss of TGF-β signaling molecules or perturbation of TGF-β *** a tumor has developed,TGF-β exerts a promoting effect on the tumor itself and stromal cells to enhance cell growth,alter the responsiveness of tumor cells to stimulate invasion and metastasis,and inhibited immune ***,novel development of therapeutic agents to inhibit TGF-β-induced progression of tumor and to retain its growth inhibitory activities,in addition to anti-inflammatory actions,could be useful in *** this review,we discuss the role of TGF-β in inflammation and carcinogenesis of the GI tract related to abnormal TGF-β signaling.

The flow characteristics of aluminum coated boron steel in hot press forming were ***,the effects of aluminum coated layer on press forming were analyzed during deep *** results show that aluminum coated boron steel exhibits a high sensitivity on temperature and strain *** coating layer appears in surface flaking in a temperature range of 700-800 ℃,but smooth surface is formed above 900 ℃.

Inhibitory actions of mibefradil on steroidogenesis in mouse Leydig cells： involvement of Ca^2＋ entry via the T-type Ca^2＋ channel

Intracellular cAMP and Ca^2＋ are involved in the regulation of steroidogenic activity in Leydig cells, which coordinate responses to luteinizing hormone （LH） and human ehorionic gonadotropin （hCG）. however, the identification of Ca^2＋ entry implicated in Leydig cell steroidogenesis is not well defined. The objective of this study was to identify the type of Ca^2＋ channel that affects Leydig cell steroidogenesis. In vitro steroidogenesis in the freshly dissociated Leydig cells of mice was induced by hCG incubation. The effects of mibefradil （a putative T-type Ca^2＋ channel blocker） on steroidogenesis were assessed using reverse transcription （RT）-polymerase chain reaction analysis for the steroidogenic acute regulatory protein （STAR） mRNA expression and testosterone production using radioimmunoassay. In the presence of 1.0 mmol L-1 extracellular Ca^2＋, hCG at 1 to 100 IU noticeably elevated both StAR mRNA level and testosterone secretion （P 〈 0.05）, and the stimulatory effects of hCG were markedly diminished by mibefradil in a dose-dependent manner （P 〈 0.05）. Moreover; the hCG-induced increase in testosterone production was completely removed when external Ca^2＋ was omitted, implying that Ca entry is needed for hCG-induced steroidogenesis. Furthermore, a patch-clamp study revealed the presence of mibefradil-sensitive Ca^24- currents seen at a concentration range that nearly paralleled those inhibiting steroidogenesis. Collectively, Our data provide evidence that hCG-stimulated steroidogenesis is mediated at least in part by Ca^2＋ entry carried out by the T-type Ca^2＋ channel in the Leydig cells of mice.

Application of direct laser metal tooling for AISI H13 tool stee

In the die industry, it is commonly agreed that residual tool life can be successfully extended by timely repair of damaged surfaces. Traditionally, the main repair process is tungsten inert gas (TIG) welding, but a new process called direct laser metal tooling (DLMT) emerges. DLMT is a manual process, of which results depend on the materials of the powders and tools, the laser process and parameters. This technology is a direct-metal freeform fabrication technique in which a 200 W fiber laser is used. AISI H13 tool steel is a suitable material for die casting tools because of the high resistance to thermal fatigue and dimensional stability. In this research, AISI H13 tool steel was melted with metal powder by fiber laser. Before melting AISI H13, the powders were analyzed with XRF equipment. Then, hardness distribution of laser melted zone was investigated. The microstructure in laser melted zone was discussed. In order to identify the effect of particle size of powder on the melted zone, two types of particle sizes of powders were used. Experimental results show that the mold repair process using DLMT can be applied in the mold repair industry.

Effect of Laser Parameters on Sintered Powder Morphology

Selective laser sintering is a kind of rapid prototyping process whereby a three-dimensional part is built layerwise by laser scanning a powder. This process is highly inffuenced by powder and laser parameters such as laser power, scan rate, spot size and layer thickness. Therefore a study on fabricating a line with Fe-Ni-Cr powder on AISI H13 tool steel has been performed by selective laser sintering. In this study, fabrication was performed by experimental facilities consisting of a 200W fiber laser which can be focused to 0.08 mm and atmospheric chamber which can control atmospheric pressure with argon. The line was fabricated with various laser power, scan rate and layer thickness. Line width and surface quality were investigated. With power increase or layer thickness decrease, line width was decreased and line surface quality was improved with scan rate optimization.

Laser surface hardening of AISI H13 tool steel

An attempt was made to improve the surface hardness and wear properties of AISI H13 tool steel through solid solution hardening and refinement of microstructures using a 200 W fiber laser as a heat generating *** hardness of laser melted zone was *** order to identify the effect of heat input on the laser melting zone,scanning conditions were *** results show that,the hardness of as-received AISI H13 tool steel is approximately Hv 240,and the hardness after laser surface heat treatment is around Hv *** hardening depth and width are increased with the increase in the heat input *** of experimental results will be considered in tooling industry.

An attempt to fabricate Fe-Ni-Cr coating on AISI H13 tool steel was performed with selective laser ***-Ni-Cr coating was produced by experimental facilities consisting of a 200 W fiber laser which can be focused to 80 μm and atmospheric chamber which can control atmospheric pressure with N2 or *** layer was fabricated with various process parameters such as laser power,scan rate and fill *** quality and coating thickness were measured and *** different surface patterns,such as typeⅠ,typeⅡand type Ⅲ,are shown with various test conditions and smooth regular pattern is obtained under the conditions as 10 μm of fill spacing,50-350 mm/s of scan rate and 40 μm of fill spacing,10-150 mm/s of scan *** maximum coating thickness is increased with power elevation or scan rate drop,and average thickness of 10 μm fill spacing is lower than that of 40 μm fill spacing.

The fiber properties depending on the coating thickness and long-term time aging were investigated. The coating strip forces of optical fiber were strongly increased with secondary coating thickness. But it did not change with time aging up to 90 days. The tensile strength and stress corrosion parameters of coated fiber were increased with time aging also but the increasing rate was quite different depending on the coating

Predictors of poor outcomes in patients with wild mushroom-induced acute liver injury

To identify early predictive markers of poor outcomes in patients with acute liver injury from wild mushroom *** observational, retrospective record review involved adults aged ≥ 18 years admitted to emergency department with mushroom intoxication from January 2005 to December 2015. The diagnosis of mushroom intoxication was based on the following: (1) a positive history of recent wild mushroom intake (either raw or cooked); (2) the onset of gastrointestinal symptoms, such as watery diarrhea, vomiting, and/or abdominal pain, after ingestion; and (3) the exclusion of other possible causes of acute liver injury. Acute liver injury was defined by a > 5-fold elevation of liver enzymes or moderate coagulopathy [international normalized ratio (INR) > 2.0]. Clinical and laboratory findings were compared in survivors and *** 93 patients with mushroom intoxication, 23, 11 men (47.8%) and 12 women (52.2%), of median age 61 years, developed acute liver injury. The overall in-hospital mortality rate was 43.5% (10/23). Among the laboratory variables, mean serum alkaline phosphatase (73.38 ± 10.89 mg/dL vs 180.40 ± 65.39 mg/dL, P howed that total bilirubin concentration (OR = 3.58, 95%CI: 1.25-10.22), indirect/direct bilirubin ratio (OR = 0.14, 95%CI: 0.02-0.94) and aPTT (OR = 1.30, 95%CI: 1.04-1.63) were significantly associated with mortality.
