Transgenesis and genome editing in birds are based on a unique germline transmission system using primordial germ cells(PGCs), which is quite different from the mammalian transgenic and genome editing system. PGCs are progenitor cells of gametes that can deliver genetic information to the next generation. Since avian PGCs were first discovered in nineteenth century, there have been numerous efforts to reveal their origin, specification, and unique migration pattern, and to improve germline transmission efficiency. Recent advances in the isolation and in vitro culture of avian PGCs with genetic manipulation and genome editing tools enable the development of valuable avian models that were unavailable before. However, many challenges remain in the production of transgenic and genome-edited birds,including the precise control of germline transmission, introduction of exogenous genes, and genome editing in ***, establishing reliable germline-competent PGCs and applying precise genome editing systems are critical current issues in the production of avian models. Here, we introduce a historical overview of avian PGCs and their application, including improved techniques and methodologies in the production of transgenic and genome-edited birds, and we discuss the future potential applications of transgenic and genome-edited birds to provide opportunities and benefits for humans.

This study aimed to develop a pH-responsive folic acid-grafted organic/inorganic hybrid nanocomposite system for site-selective oral delivery of therapeutic antibodies. A folic acid-grafted aminoclay(FA-AC) was prepared via an in situ sol-gel method. Then, a drug-loaded nanocomplex was prepared via the electrostatic interaction of FA-AC with infliximab(IFX), a model antibody, and coated with Eudragit? S100(EFA-AC-IFX). FA-AC exhibited favorable profiles as a drug carrier including low cytotoxicity, good target selectivity, and capability to form a nanocomplex with negatively charged macromolecules. A pH-responsive FA-AC-based nanocomplex containing IFX(EFA-AC-IFX) was also obtained in a narrow size distribution with high entrapment efficiency(>87%). The conformational stability of IFX entrapped in EFA-AC-IFX was well maintained in the presence of proteolytic enzymes. EFA-ACIFX exhibited pH-dependent drug release, minimizing premature drug release in gastric conditions and the upper intestine. Accordingly, oral administration of EFA-AC-IFX to colitis-induced mice was effective in alleviating the progression of ulcerative colitis, while oral IFX solution had no efficacy. These results suggest that a pH-responsive FA-AC-based nanocomposite system can be a new platform for the site-selective oral delivery of therapeutic antibodies.

OBJECTIVE:To assess the efficacy of acupoint catgut embedding(ACE)for simple obesity in preclinical animal ***:We searched the following 14 electronic databases:PubM ed,Cochrane Library,EMBASE,Oriental Medicine Advanced Searching Integrated System,KoreaMed,Korean Studies Information Service System,Science-on,Research Information Sharing Service,Korea Citation Index,Korea Traditional Knowledge Portal,China Network Knowledge Infrastructure Database,Wanfang Database,Chinese Science and Technology Journal Database,and Chinese Biology Medicine Database,from inception to November 2021 without language *** assessment was performed according to the guidelines of Animal Research:Reporting of in vivo experiments;and Metaanalysis was performed using Reviewer Manager 5.4.1 ***:Twenty-four studies involving 813 animals were ***-analysis showed that ACE was beneficial for weight control[n=40,MD=-50.63,95%CI(-57.59,-43.67),P<0.00001,I 2=0%]and reduced the Lee index[n=40,MD=-18.79,95%CI(-20.01,-17.57),P<0.00001,I 2=0%].However,when efficacy of ACE was compared with that of manual acupuncture,electroacupuncture,or oilistat therapy,statistical difference was not observed between the two ***:This systematic review suggests that ACE may be efficacious in treating ***,the analyses highlighted the necessity to perform welldesigned,higher-quality experiments.

Indoor air quality(IAQ) directly affects the health of occupants. Household manufacturing equipment(HME) used for hobbies or educational purposes is a new and unexplored source of air pollution. In this study, we evaluated the characteristics of particulate and gaseous pollutants produced by a household laser processing equipment(HLPE). Various target materials were tested using a commercial HLPE under various operating conditions of laser power and sheath air flow rate. The mode diameters of the emitted particles gradually decreased as laser power increased, while the particle number concentration(PNC) and particle emission rate(PER) increased. In addition, as the sheath air flow rate quadrupled from 10 to 40 L/min, the mode diameter of the emitted particles decreased by nearly 25%, but the effect on the PNC was insignificant. When the laser induced the target materials at 53 m W, the mode diameters of particles were 2.0 × 10^(4) particles/cm^(3). Particularly, analyses of sampled aerosols indicated that harmful substances such as sulfur and barium were present in particles emitted from leather. The carcinogenic gaseous pollutants such as acrylonitrile, acetaldehyde, 1,3-butadiene, benzene, and C 8 aromatics(ethylbenzene) were emitted from all target materials. In an actual indoor environment, the PNC of inhalable ultrafine particles(UFPs) was > 5 × 10^(4) particles/cm^(3) during 30 min of HLPE operation. Our results suggest that more meticulous control methods are needed, including the use of less harmful target materials along with filters or adsorbents that prevent emission of pollutants.

Background:Several studies evaluated the current guideline of the American Society for Gastrointestinal Endoscopy(ASGE)and reported only suboptimal *** study evaluated the diagnostic performance of the ASGE guideline based on computed tomography(CT)and role of endoscopic ultrasonography(EUS)and magnetic resonance cholangiopancreatography(MRCP)in patients with suspected choledocholithiasis but negative CT ***:Patients with suspected choledocholithiasis undergoing ERCP between January 2016 and January 2017 were retrospectively *** patients underwent CT to detect *** or MRCP was performed when the CT scan showed negative *** were classified into the high and intermediate-risk groups,based on predictors from the ASGE ***:Of 583 patients with suspected choledocholithiasis,340(58.3%)had stones on ERCP(65.9%in the high-risk group and 40.6%in the intermediate-risk group).The accuracy of ASGE guideline for CT was 63.98%(79.12%sensitivity,42.80%specificity)and 36.02%(20.88%sensitivity,57.20%specificity)in the high-risk and intermediate-risk groups,*** 103 patients in the high-risk group underwent both CT and US,the accuracy of CT was higher than that of US for detecting choledocholithiasis(78.64%vs.53.40%),with a significant difference in area under the curve(AUC)(0.78 vs.0.59,Phan that of MRCP(90.91%vs.82.76%),but with no significant difference in AUC(0.91 vs.0.83,P=0.347).Conclusions:CT-based ASGE guideline showed superior diagnostic performance than US for predicting *** diagnostic options,EUS or MRCP,with negative CT finding showed comparable ***,the diagnostic modality should be selected based on availability,experience,cost,and contraindications.

Germ cells are the only cell type in the body that can transfer genetic information to the next generation. Germline-competent stem cells can self-renew and contribute to the germ cell lineage giving rise to pluripotent stem cells under specific conditions. Hence far, studies on germline-competent stem cells have contributed to,the generation of avian model systems and the conservation of avian genetic resources. In this review, we focus on previous studies on germline-competent stem cells from avian species, mainly chicken germline-competent stem cells, which have been well established and characterized. We discuss different sources of germline-competent stem cells and recent advances for the future applications in birds.
