High-throughput discovery of highly selective reversible hMAO-B inhibitors based on at-line nanofractionation

Human monoamine oxidase B(hMAO-B)has emerged as a pivotal therapeutic target for Parkinson's *** to adverse effects and shortage of commercial drugs,there is a need for novel,highly selective,and reversible hMAO-B inhibitors with good blood-brain barrier *** this study,a high-throughput at-line nanofractionation screening platform was established with extracts from Chuanxiong rhizoma,which resulted in the discovery of 75 active compounds,including phenolic acids,volatile oils,and phthalides,two of which were highly selective novel natural phthalide hMAO-B inhibitors that were potent,selective,reversible and had good blood‒brain *** docking and molecular dynamics simulations elucidated the inhibition ***(IC_(50)=103 nmol/L;SI=645)and neocnidilide(IC_(50)=131 nmol/L;SI=207)demonstrated their excellent potential as hMAO-B *** offset the limitations of deactivating enzymes associated with irreversible hMAO-B inhibitors such as *** SH-SY5Y cell assays,sedanolide(EC_(50)=0.962μmol/L)and neocnidilide(EC_(50)=1.161μmol/L)exhibited significant neuroprotective effects,comparable to the positive drugs rasagiline(EC_(50)=0.896μmol/L)and safinamide(EC_(50)=1.079μmol/L).These findings underscore the potential of sedanolide as a novel natural hMAO-B inhibitor that warrants further development as a promising drug candidate.

Infant’s Sigh Breathing Matters—The role of the Hering-Breuer Deflation reflex in Bronchial Decongestion

Background: The prolonged slow expiratory (PSE) technique can induce sigh breaths related to the Hering-Breuer reflex in *** is a specific chest physiotherapy technique often used in children to help with the clearance of secretions, reduce pulmonary obstruction and decrease ventilatory effort. Purpose: The main objective of this review was to synthesize and clarify the existing knowledge of the physiological mechanism behind the Hering-Breuer reflex and its utility in the PSE technique. The secondary objective was to provide an overview of PSE which is probably the main mechanical feature for mucus clearance in infants and to identify gaps and any further research required. Method: A comprehensive literature search was performed using PubMed/MEDLINE and Ovid EMBASE, from 1968 to January 2024, using search terms relating to “Interaction of Hering-Breuer reflex and chest physiotherapy therapy, sigh breath in infants and prolonged slow expiration technique” to create an overview of the advances and gaps in current knowledge of PSE. result: The data obtained following treatment with PSE maneuvers in infants with histories of recurrent wheezing shows that the presence of sighs could be the main active principle of treatment. Conclusion: Changes in volume induced by PSE technique and, effects from crying may stimulate the Hering-Breuer deflation reflex and sighs in turn improving alveolar ventilation as well as lung volume in infants.

In this study,Mn_(3)O_(4) spherical particles(SPs)were synthesized by the sol-gel process,after which they were thermally annealed at 400℃,and comprehensively characterized.X-ray Diffraction(XrD)revealed that Mn_(3)O_(4) exhibited a tetragonal spinel structure,and Fourier transformed infrared(FTIr)spectroscopy identified surfaceadsorbed functional *** electron microscopy(SEM)and the specific surface area analyses by Brunauer−Emmett−Teller(BET)revealed a porous,homogeneous surface composed of strongly agglomerated spherical grains with an estimated average particle size of∼35 nm,which corresponded to a large specific surface area of∼81.5 m^(2)/g.X-ray photoelectron spectroscopy(XPS)analysis indicated that Mn_(3)O_(4) was composed of metallic cations(Mn^(4+),Mn_(3)+,and Mn^(2+))and oxygen species(O_(2)−,OH−and CO_(3)2−).The optical bandgap energy is∼2.55 *** of the catalytic performance of the Mn_(3)O_(4) SPs indicated T90 conversion of CH4 to CO_(2) and H2O at 398℃ for gas hourly space velocity(GHSV)of 72000 mL^(3) g^(−1) h^(−1).This observed performance can be attributed to the cooperative effects of the smallest spherical grain size with a mesoporous structure,which is responsible for the larger specific surface area and available surface-active oxygenated *** cooperative effect of the good reducibility,higher ratio of active species(OLat/OAds),and results of density functional theory(DFT)calculations suggested that the total oxidation of CH4 over the mesoporous Mn_(3)O_(4) SPs might take place via a two-term process in which both the Langmuir−Hinshelwood and Mars−van Krevelen mechanisms are cooperatively involved.

软件缺陷定位是指找出与软件失效相关的程序元素.当前的缺陷定位技术仅能产生函数级或语句级的定位结果.这种粗粒度的定位结果会影响人工调试程序和软件缺陷自动修复的效率和效果.专注于细粒度地识别导致软件缺陷的具体代码令牌,为代码令牌建立抽象语法树路径,提出基于指针神经网络的细粒度缺陷定位模型来预测出具体的缺陷代码令牌和修复该令牌的具体操作行为.开源项目中的大量缺陷补丁数据集包含大量可供训练的数据,且基于抽象语法树构建的路径可以有效捕获程序结构信息.实验结果表明所训练出的模型能够准确预测缺陷代码令牌并显著优于基于统计的与基于机器学习的基线方法.另外,为了验证细粒度的缺陷定位结果可以贡献于缺陷自动修复,基于细粒度的缺陷定位结果设计两种程序修复流程,即代码补全工具去预测正确令牌的方法和启发式规则寻找合适代码修复元素的方法,结果表明两种方法都能有效解决软件缺陷自动修复中的过拟合问题.

Theoretical and Experimental Quantification of Solar radiation through a Tracking System

This work deals with the estimation of solar radiation through a solar tracker aimed at evaluating the effect of solar tracking on the solar deposit in Burkina Faso. Using a two-axis solar tracking system, we experimentally measured solar radiation at our Joseph KI-ZErBO University site and compared it with that obtained by a numerical simulation run using Fortran programming software based on a mathematical model by Brichambaut. The results obtained from the mathematical and experimental studies show that, with a solar tracker, on a clear-sky day, solar irradiation is between 800 W·m−2 and 1000 W·m−2 between about 8 a.m. and 4 p.m., i.e. a duration of 8 hours of insolation. Analysis of the numerical and experimental results shows very good quantitative and qualitative agreement, with an average relative error of 18%.

Seroprevalence of HBV and HCV among People Living with HIV in Burkina Faso and Diagnostic Performance of HIV/HCV/HBsAg Combined rapid Test in Comparison with Architect Assays

Background: The diagnosis of human immunodeficiency virus (HIV), hepatitis B virus (HBV), and hepatitis C virus (HCV) remains a constraint for some populations in sub-Saharan Africa. This study aimed to determine the prevalence of HBV and HCV in people living with HIV and to evaluate the performance of a combined rapid test for the simultaneous detection of HIV, HBV, and HCV. Methods: This is a cross-sectional study that took place from February 2017 to November 2018 and included 139 HIV-infected individuals followed up at different medical centers in Ouagadougou, Burkina Faso. HBV and HCV serology tests were performed on-site using finger prick whole blood with HIV/HCV/HBsAg combined rapid test and then serum with two reference tests “Architect HBsAg Qualitative” and “Architect HIV Ag/Ab Combo”. results: The mean age of the participants was 57 ± 8 years. Of the 139 participants, 10% (14/139) were HIV-1 positive, 71.9% (100/139) were HIV-2 positive, and 18.0% (25/139) were HIV-1/HIV-2 coinfected. The sensitivity and specificity of the HIV/HCV/HBsAg combined rapid test were 33.33% vs 99.11% and 20% vs 99.25% compared to Architect HBsAg Qualitative and Architect HIV Ag/Ab Combo, respectively. The Kappa and Youden Index values were 0.4262 and 0.3244 and 0.2707 and 0.1925, respectively, compared to each of the two reference tests. Conclusion: The results show that the HIV/HCV/HBsAg combined rapid test has poor diagnostic efficiency and should not be recommended for the diagnosis of these viruses.

Investigation of Carbonation of Concrete Based on Crushed Sand and Admixtures

Carbonation is a natural aging process that occurs in all types of concrete. One of its primary implications is the acceleration of steel corrosion caused by the phenomena of depassivation. The goal of this research is to investigate the carbonation of quarry sand-based concrete. The concrete is made of 100% crushed sand 0/6.3, gravel 8/15, and 15/25 from the Arab Contractor quarry in Nomayos, Cameroon, with CEM II B-P 42.5 r from CIMENCAM (Cimenteries du Cameroun). The study employed two admixtures: one with a dual superplasticizing and reducing action (Sikamen) and another with a water-repellent effect (Sika liquid). Carbonation was performed on concrete samples at the following dates: 0, 7, 14, 28, 56, 90, 180 days, one year, and six months. Carbonated concrete (CC) and non-carbonated concrete (NCC) samples are compared in terms of their physical attributes and mineralogical characteristics. The results of this investigation reveal that after more than a year and six months of carbonation, porosity decreases and permeability increases. Despite the high fineness modulus of quarry sand, the compressive strength of quarry sand-based concrete is satisfactory. Carbonation depth is relatively high on some dates, exceeding the minimal cover value for concrete reinforcement. Sikament additive increases concrete compactness and durability while decreasing permeability. Sika water repellant mixes with the lime in cement to generate complimentary crystallizations that block the mortar’s capillaries, making it watertight.

Uncovering the Challenges of Faecal Sludge Management in Benin’s Urban Hubs: The Cases of Abomey-Calavi and Natitingou

The issue of faecal sludge management in developing countries remains a significant challenge due to the indiscriminate discharge of sludge into the environment. This practice generates more environmental problems than it solves. This study aims to assess the management of household faecal sludge in the communes of Abomey-Calavi and Natitingou, Benin, in order to optimise the best storage conditions for subsequent more effective treatment. To this end, a sociological survey was conducted among households in Abomey-Calavi and Natitingou, as well as among manual emptiers. The results of our studies revealed that two types of latrines are used in households. 20.55% of households use traditional latrines, while 59.83% use ordinary latrines. 7.97% of households use both types of latrine. Moreover, the depth of the latrines varies from 2.5 m to 7 m in Abomey-Calavi and from 2.5 m to 8 m in Natitingou. Among households with a latrine, 28.26% empty their pit at least once, while 71.74% have never emptied it. The emptying cost varies between 35,000 FCFA and 90,000 FCFA. The mixing of faecal sludge with solid waste, including plastic materials, makes emptying difficult due to the obstruction of the vacuum pump. The results obtained will serve as a reliable database to facilitate decision-making in the context of faecal sludge management.

In the digital age, the data exchanged within a company is a wealth of knowledge. The survival, growth and influence of a company in the short, medium and long term depend on it. Indeed, it is the lifeblood of any modern company. A companys operational and historical data contains strategic and operational knowledge of ever-increasing added value. The emergence of a new paradigm: big data. Today, the value of the data scattered throughout this mother of knowledge is calculated in billions of dollars, depending on its size, scope and area of intervention. With the rise of computer networks and distributed systems, the threats to these sensitive resources have steadily increased, jeopardizing the existence of the company itself by drying up production and losing the interest of customers and suppliers. These threats range from sabotage to bankruptcy. For several decades now, most companies have been using encryption algorithms to protect and secure their information systems against the threats and dangers posed by the inherent vulnerabilities of their infrastructure and the current economic climate. This vulnerability requires companies to make the right choice of algorithms to implement in their management systems. For this reason, the present work aims to carry out a comparative study of the reliability and effectiveness of symmetrical and asymmetrical cryptosystems, in order to identify one or more suitable for securing academic data in the DrC. The analysis of the robustness of commonly used symmetric and asymmetric cryptosystems will be the subject of simulations in this article.
