Human sperm quality and lipid content after migration into normal ovulatory human cervical mucus containing low numbers of leukocytes

The aim of this study was to investigate whether a relationship exists between the presence of low numbers of leukocytes in normal ovulatory cervical mucus and sperm quality and lipid content after migration. The percentages of live, motile and morphologically normal spermatozoa, movement parameters assessed by computer-aided sperm analysis （cASA）, and ionophore-induced acrosome reaction measured by flow cytometry were determined before and after migration. High-performance liquid chromatography with ultraviolet detection was used to measure the sperm lipid content, including the various diacyl subspecies. The number of leukocytes found in solubilized mucus samples was counted using a haemocytometric method. Overall, the presence of leukocytes in the cervical mucus samples did not significantly influence sperm motility and morphology, sperm kinematic parameters, or the sperm content in sphingomyelin or cholesterol. In contrast, after migration, the decrease in various sperm diacyls and the level of induced acrosome reaction was significantly less pronounced in mucus samples containing ≥10^4 leukocytes than in mucus samples with no or rare leukocytes whereas the level of induced acrosome reaction was higher. The present data suggest that the low level of leukocytes found in normal ovulatory cervical mucus could influence the process of sperm lipid remodelling/capacitation.

Synergistic Antitumor Activity of Vitamins c and K3 on Human Bladder cancer cell Lines

Exponentially growing cultures of human bladder tumor cells (RT4 and T24) were treated with Vitamin c (Vc) alone, Vitamin K3 (VK3) alone, or with a Vc:VK3 combination continuously for 5 days or treated with vitamins for 1 h, washed with PBS and then incubated in culture medium for 5 days. co-administration of the vitamins enhanced the antitumor activity 12- to 24-fold for the RT-4 cells and 6- to 41-fold for the T24 cells. Flow cytometry of RT4 cells exposed to the vitamins revealed a growth arrested population and a population undergoing cell death. Growth arrested cells were blocked near the G0/G1-S-phase interface, while cell death was due to autoschizis. catalase treatment abrogated both cell cycle arrest and cell death which implicated hydrogen peroxide (H2O2) in these processes. The H2O2 production resulted in a moderate increase in lipid peroxidation and depletion of cell thiol levels. Analysis of cellular ATP levels revealed a transient increase in ATP production for Vc and the Vc:VK3 combination, but decreased ATP levels following VK3 treatment. Lipid peroxidation, thiol depletion and ATP modulation occurred at a 17-fold lower concentration in the vitamin combination than with either vitamin alone. These results suggested that the increased cytotoxicity of the vitamin combination was due to redox cycling and increased oxidative stress.

characterization of Household Solid Waste in the Town of Abomey - calavi in Benin

Identification of waste characteristics is an important step towards improving waste recovery. The aim of this research was to determine the physical and physico-chemical characteristics of waste of Abomey–calavi city and to study the relationship between standard of living and average ratio of daily waste generated by each person. In this study the methodology used French standards to characterize particle size and typology of solid waste generated by the population of calavi city in Benin, West Africa. According to home criteria, the study area was stratified into three distinct levels of standard of living called: high standing, medium standing and low standing;Waste from 60 households was weighed daily. The total waste produced by each household was collected seven (7) days a week, for a period of three weeks. Waste characterization was performed using ratio, size granulometry and typological composition. Physico- chemical analysis including organic mater, pH, Total Organic carbon, total Kjeldahl nitrogen and metal trace element were also performed. To better assess waste compostability, water extractable organic matter was quantified and qualitative identification was made with XAD8 and XAD4 resins. Results show that the amount of waste increases with the standard of living;the average ratio of daily waste generated is 0.89 kg day-1 person-1. Independently of the standard of living, fermentable compounds represent the largest proportion of waste materials (45%). Qualitative difference of waste content in organic matter is shown as a function of the population’s living standards. These results could be explained by a higher consumption of meat in the households with a higher standard of living, reflecting a greater proportion of transphilic (TPI), and hydrophilic (HPI) fractions. Virological response and resistance mutations to NS3/4A inhibitors in hepatitis c virus-human immunodeficiency virus coinfection

AIM: To evaluate virological response to telaprevir or boceprevir in combination with pegylated interferon and ribavirin and resistance mutations to NS3/4A inhibitors in hepatitis c virus-human immunodeficiency virus(HcV-HIV) coinfected patients in a real life setting. METHODS: Patients with HcV genotype 1-HIV coinfection followed in Nice University Hospital internal medicine and infectious diseases departments who initiated treatment including pegylated interferon and ribavirin(Peg IFN/RBV) + telaprevir or boceprevir, according to standard treatment protocols, between August 2011 and October 2013 entered this observational study. Patient data were extracted from an electronic database(Nadis®). Liver fibrosis was measured by elastometry(Fibroscan®) with the following cut-off values: F0-F1: cal response(SVR) twelve weeks after completing treatment, frequency and type of adverse events, and NS3/4A protease inhibitor mutations were described. RESULTS: Forty-one patients were included: 13(31.7%) patients were HcV-treatment na?ve, 22(53.7%) had advanced liver fibrosis or cirrhosis(Fibroscan stage F3 and F4); none had decompensated cirrhosis or hepatocellular carcinoma; all were receiving antiretroviral treatment, consisting for most them(83%) in either a nucleoside reverse-transcriptase inhibitor/protease inhibitor or/integrase inhibitor combination; all patients had undetectable HIV-RNA. One patient was lost to follow-up. SVR was achieved by 52.5% of patients. Five patients experienced virological failure during treatment and four relapsed. Seven discontinued treatment due to adverse events. Main adverse events included severe anemia(88%) and rash(25%). Experimental and Finite Element Analysis of a T-Joint Welding

This paper presents the Finite Element （FE） modeling of a two-seam welding process for a T-joint with a V chamfer preparation： The aim of the model is to predict the deformations, distortions and residual stresses resulting from the welding of the plates and experiments have been carried out in order to compare to the FE model. The ＂birth and death＂ method is used in ANSYS to simulate the filler metal deposition and the heat generation and weld pool simulation are conducted accordingly with the double ellipsoid configuration as proposed by Goldak et al. The model takes into consideration the temperature dependent non-linear material properties and uses a new formulation to compute the temperature dependent combined coefficient of heat loss. Improvements in the calculation are achieved by combining two types of meshing. The FE simulation is divided into two consecutive parts： the thermal simulation followed by the structural simulation. The results of the numerical model are compared to experiments.

Implementation of the Semi-Distributed SWAT (Soil and Water Assessment Tool) Model capacity in the Lobo Watershed at Nibéhibé(center-West of côte d’Ivoire)

The Lobo watershed is an agricultural area where the use of fertilizers by farmers is intensive, causing eutrophication problems that deteriorate the quality of drinking water distributed to the population. Since the phenomenon of eutrophication is directly linked to runoff, it is essential to model the flow in order to better control the transfer of nutrients responsible for eutrophication. It is within this framework that this study was conducted. The objective of this study is to assess the ability of the semi-distributed SWAT (Soil and Water Assessment Tool) model to simulate runoff in the Lobo watershed. The methodology adopted was based on the use of the QSWAT graphical interface to manipulate and execute the main functions of the SWAT model from QGIS tools. The hydrological modeling was carried out with the QSWAT interface for SWAT 2012. The results showed good performance for the flow calibration (1982-1984) with the evaluation criteria R2, NSE and PBIAS respectively of 0.64, 0.64 and 3.1. In the validation period (1984-1987), the model also showed good performance in the streamflow simulation for R2 and NSE of 0.84 and 0.76 respectively as values. However, for the PBAIS criterion, the result was less good but still remains satisfactory with a value of 19.6. It emerges from this study that the SWAT model is suitable for simulating water transfer and can therefore be used to study the transfer of pollutants in the fight against eutrophication in the Lobo watershed.

NBO Analysis by ONIOM and DFT (B3LYP) calculations of Intramolecular and Intermolecular Interactions of Artemisinin, Quinine and Thirteen Manzamenones with H₂O or Alanine

This work was undertaken to analyze intramolecular and intermolecular interactions of Manzamenones from natural bond orbitals (NBO method). For their use in the treatment of malaria, the results of these molecules are compared to those of Artemisinin and Quinine. Manzamenones are a class of atypical fatty acids. They are isolated from a marine sponge of the genus Plakortis kenyensis. The analysis of intramolecular interactions compares the results of each molecule (Manzamenones, Artemisinin and Quinine) in the non-complexed state with those of its complex with a water molecule. Thus, for the same electron donors (i) and associated acceptors (j), the electron density (ED), stabilization energy E2 related to the delocalization of i to j, the energies of the NBO orbitals εi and εj of the donor and acceptor, respectively, and element of the Fock matrix Fi,j are determined and compared. The change in E2 is used to deduce whether or not the molecule is stabilized after complex formation. These analyses allowed to match each Manzamenone to one of the two antimalarials. The intermolecular interactions were analyzed, for each molecule (Manzamenones, Artemisinin and Quinine), in two complexes. These complexes are obtained with a water molecule on the one hand and with an alanine molecule on the other hand. For these interactions, the electron donor and its electron density, the electron acceptor and its electron density as well as the donor—acceptor stabilization energy have been calculated. The ONIOM 2 method is used to study Manzamenones. Theoretical calculations were done using density functional theory (B3LYP) by combining one of the two function bases 6-31++G(d,p) and 6-31+G(d,p).

Mapping of Potential Recharge Areas of Fractured Aquifers in the Departments of Yamoussoukro and Toumodi (central côte d’Ivoire)

Groundwater is an important natural resource for all human activity. Today, due to climate change and population growth, the demand has increased considerably, thus requiring their evaluation to ensure sustainability. However, one of the important but difficult to estimate parameters due to its variability is the recharge. This work aims to map the potential recharge of aquifers in the departments of Yamoussoukro and Toumodi. Saaty’s AHP (Analytical Hierarchy Process) multi-criteria analysis technique was used by integrating different thematic layers. First, a reclassification then weighting of these parameters was made according to their influence in the recharge process. Finally, they were integrated into a GIS to produce the map of potential groundwater recharge zones. The results indicate that the potential high recharge areas represent approximately 43.11% of the area of the study area. These areas appear scattered in the sub-prefecture of Yamoussoukro, Kokoumbo Attiegouakro, Toumodi et Kpouèbo.

Dirac concentrations in a chemostat Model of Adaptive Evolution(In honor of the immense scientific influence of Ham Brezis)

This paper deals with a non-local parabolic equation of Lotka-Volterra type that describes the evolution of phenotypically structured populations. Nonlinearities appear in these systems to model interactions and competition phenomena leading to selection. In this paper, the equation on the structured population is coupled with a differential equation on the nutrient concentration that changes as the total population *** methods aimed at showing the convergence of the solutions to a moving Dirac mass are reviewed. Using either weak or strong regularity assumptions, the authors study the concentration of the solution. To this end, BV estimates in time on appropriate quantities are stated, and a constrained Hamilton-Jacobi equation to identify where the solutions concentrates as Dirac masses is derived.
