Background:Diabetic retinopathy(DR)is a leading cause of vision loss in ***,the standard imaging technique to monitor and prognosticate DR and diabetic maculopathy is dye-based *** the introduction of optical coherence tomography angiography(OCTA),it may serve as a potential rapid,non-invasive imaging modality as an *** text:Recent studies on the role of OCTA in DR include the use of vascular parameters e.g.,vessel density,intercapillary spacing,vessel diameter index,length of vessels based on skeletonised OCTA,the total length of vessels,vascular architecture and area of the foveal avascular *** quantitative measures may be able to detect changes with the severity and progress of DR for clinical *** may also serve as a non-invasive imaging method to detect diabetic macula ischemia,which may help predict visual ***,there are many limitations of OCTA in DR,such as difficulty in segmentation between superficial and deep capillary plexus;and its use in diabetic macula edema where the presence of cystic spaces may affect image *** applications of OCTA in the anterior segment include detection of anterior segment ischemia and iris neovascularisation associated with proliferative DR and risk of neovascular ***:OCTA may potentially serve as a useful non-invasive imaging tool in the diagnosis and monitoring of diabetic retinopathy and maculopathy in the *** studies may demonstrate how quantitative OCTA measures may have a role in detecting early retinal changes in patients with diabetes.

Extending the neurocircuitry of behavioural inhibition:a role for the bed nucleus of the stria terminalis in risk for anxiety disorders

Behavioural inhibition is a biologically based risk factor for anxiety *** with behavioural inhibition are shy,cautious and avoidant of new *** research on behavioural inhibition has focused on the amygdala as an underlying neural substrate and has identified differences in amygdala function and volume;however,amygdala findings have yet to lead to meaningful interventions for prevention or treatment of anxiety *** bed nucleus of the stria terminalis(BNST)is a prime candidate to be a neural substrate of behavioural inhibition,given current evidence of BNST function and development in human research and animal *** with behavioural inhibition have an increased startle response to safety cues and an increased cortisol response to social evaluative situations,both of which are mediated by the *** rodents,activation of the BNST underlies contextual fear responses and responses to uncertain and sustained ***-human primates with anxious temperament(the macaque equivalent of behavioural inhibition)have increased BNST activity to ambiguous social situations,and activity of the BNST in anxious temperament is significantly ***,the BNST is sexually dimorphic and continues to develop into adulthood,paralleling the development of anxiety disorders in ***,these findings suggest that further investigation of the BNST in behavioural inhibition is necessary and may lead to new avenues for the prevention and treatment of anxiety disorders.

Gene expression arrays as a tool to unravel mechanisms of normal tissue radiation injury and prediction of response

Over the past 5 years there has been a rapid increase in the use of microarray technology in the field of cancer research, The majority of studies use microarray analysis of tumor biopsies for profiling of molecular characteristics in an attempt to produce robust classifiers for prognosis. There are now several published gene sets that have been shown to predict for aggressive forms of breast cancer, where patients are most likely to benefit from adjuvant chemotherapy and tumors most likely to develop distant metastases, or be resistant to treatment. The number of publications relating to the use of microarrays for analysis of normal tissue damage, after cancer treatment or genotoxic exposure, is much more limited. A PublVled literature search was conducted using the following keywords and combination of terms： radiation, normal tissue, microarray, gene expression profiling, prediction. With respect to normal tissue radiation injury, microarrays have been used in three ways：（1） to generate gene signatures to identify sensitive and resistant populations （prognosis）; （2） to identify sets of biomarker genes for estimating radiation exposure, either accidental or as a result of terrorist attack （diagnosis）; （3） to identify genes and pathways involved in tissue response to injury （mechanistic）. In this article we will review all （relevant） papers that covered our literature search criteria on microarray technology as it has been applied to normal tissue radiation biology and discuss how successful this has been in defining predisposition markers for radiation sensitivity or how it has helped us to unravel molecular mechanisms leading to acute and late tissue toxicity. We also discuss some of the problems and limitations in application and interpretation of such data.

Production of pectic extracts from sugar beet pulp with antiproliferative activity on a breast cancer cell line

In the last years, sugar beet pectins have been the subject of several investigations involving extraction methodologies, chemical composition and functional properties. The structure of pectins, which depends on the extraction method, is decisive in their capacity to induce apoptosis on several cancer cell lines like colon, prostate and breast. In this work, sugar beet pectin extraction was performed in the following steps： lipid extraction with hexane, removal of soluble complex carbohydrates and proteins, and enzymatic treatment with amyloglucosidase, protease, and pectinase. The enzymatic treatment was carried out with Rohapect DA6L under the following conditions： 50℃, pH 4.0, 2% enzyme/substrate （E/S） ratio, 15 h, and a solid to liquid ratio of 1 ： 10. The pectic extract showed a degree of polymerization （DP） profile of 55.8% with DP≥7; 4.9% with DP6; 5.8% between DP2 and DP6 ; 4.7% with DP2; and 28.8% with DP1. The pectic extract was examined for its antiproliferative activity on the MCF-7 breast cancer cell line. At a concentration range of 12.5-25 mg/mL the pectic extract killed 80.6% of the cells, exhibiting a higher antiproliferative activity than 4-hydroxytamoxifen （4- OHT）, a classical anticancer drug, which killed 56.5% of the cells.

Rationale:Acute caustic ingestion from suicidal intent is not usual in emergency departments in developed *** of the substances commonly ingested by suicidal patients,phosphoric acid,tends to cause multi-system ***’s Concern:A 41-year-old male patient presented with complaints of throat discomfort,severe generalized abdominal pain,and multiple episodes of hematemesis after ingesting a restroom cleaning ***:Poisoning by acute caustic ingestion(containing<30%phosphoric acid and<4%ethylene glycol).Interventions:The patient was administered 50 mL of 8.4%sodium bicarbonate solution followed by an isotonic sodium bicarbonate solution running at 500 mL/h,a hyperkalemia kit,ceftriaxone,metronidazole,omeprazole,and *** patient then underwent urgent ***:The patient suffered gastrointestinal bleeding as a result of local caustic *** addition,his course of illness was complicated by severe acidemia from high anion gap metabolic acidosis and deranged electrolytes(hyperphosphatemia,hyperkalemia,and hypocalcemia).He developed multi-organ failure and eventually ***:The clinician needs to be mindful of the multi-system complications arising from such a caustic *** patients need to be monitored closely for deterioration,and have prompt management of the various arising complications,to reduce the high morbidity and mortality associated with this condition.

Development of new classes of plasmon active nano-structures and their application in bio-sensing and energy guiding

Metal nanostructures exhibit special optical resonance modes originating fronl tile subwavelengt.h confinement of conductive electrons in the material. These resonance modes represent a strong research focus due to their application potential in optics and sensing application, in this short review recent achievements of our group in this field are highlighted. A wet-chemistry approach synthesis of advanced metallic nanostruetures will be introduced and their exact positioning and manipulation by electric field is shown. Next. the application of these nanostructures for a detection of small molecules will be described in several examples. Also, it will be shown that inetal nanostruetures can be used for sub-wavelength light focusing and for efficient energy coupling into polymer chains.

Genomic imbalances in esophageal carcinoma cell lines involve Wnt pathway genes

AIM： To identify molecular markers shared across South African esophageal squamous cell carcinoma （ESCC） cell lines using o/togenetics, fluorescence in situ hybridization （FISH） and single nucleotide polymorphism （SNP） array copy number analysis. METHODS： We used conventional cytogenetics, FISH, and multicolor FISH to characterize the chromosomal rearrangements of five ESCC cell lines established in South Africa. The whole genome copy number profile was established from 250K SNP arrays, and data was analyzed with the CNAT 4.0 and GISTIC software. tions involved the following chromosomal regions and genes： 11q13.3 （CCND1, FGF3, FGF4, FGF19, MYEOV）, 8q24.21（C-MYC, FAM84B）, 11q22.1-q22.3 （B[RC2, BIRC3）, 5p15.2 （CTNND2）, 3qll.2-q12.2 （MINA） and 18p11.32 （TYMS, YES1）. The significant deletions included 1p31.2-p31.1 （CTH, GADD45a, DIRAS3）, 2q22.1 （LRPIB）, 3p12.1-p14.2 （FHIT）, 4q22.1-q32.1 （CASP6, SMAD1）, 8p23.2-q11.1 （BNIP3L） and 18q21.1-q21.2 （SMAD4, DCC）. The 3p11.2 translocation breakpoint was shared across four cell lines, supporting a role for genes involved at this site, in particular, the EPHA3 gene which has previously been reported to be deleted in ***： The finding that a significant number of genes that were amplified （FGF3, FGF4, FGF19, CCND1 and C-MYC） or deleted （SFRP2 gene） are involved in the Wnt and fibroblast growth factor signaling pathways, suggests that these pathways may be activated in these cell lines.
