Background: Neuroblastoma is the most common extracranial solid tumor in children, and treatment options for recurrent neuroblastoma are limited. Using molecular profiling to target the molecular vulnerabilities of neuroblastoma with existing therapeutic agents may result in a rational, data-driven approach with potential to improve clinical outcomes. Methods: The primary objective of this pilot study was to evaluate the feasibility of supporting real-time treatment decisions through predictive modeling of genome-wide mRNA gene expression data from neuroblastoma tumor biopsies. Feasibility was defined as completion of tumor biopsy, histopathological evaluation, RNA extraction and quality control, gene expression profiling within a CLIA-certified laboratory, bioinformatic analysis, generation of a drug predicttion report, molecular tumor board review yielding a formulated treatment plan, and independent medical monitor review within a 2-week period. Results: Five patients with multiply relapsed or refractory neuroblastoma were enrolled between April and June 2010. All biopsies passed histopathology and RNA quality control. Generation of gene expression data and its analysis (3-7 days), reports which linked this data into medically actionable drug candidates (1-5 days), molecular tumor board (1-3 days) and independent medical monitor review (1 day) were all completed in real-time. The average time was 10.5 days for all patients. Conclusion: This study shows that it is feasible to create therapeutic treatment plans based on genomic profiling in less than 12 days. This warrants further testing in a Phase I study to determine safety of predicted treatments and evaluate whether the information obtained in these analyses would result in patient benefit.

Behavioural inhibition is a biologically based risk factor for anxiety *** with behavioural inhibition are shy,cautious and avoidant of new *** research on behavioural inhibition has focused on the amygdala as an underlying neural substrate and has identified differences in amygdala function and volume;however,amygdala findings have yet to lead to meaningful interventions for prevention or treatment of anxiety *** bed nucleus of the stria terminalis(BNST)is a prime candidate to be a neural substrate of behavioural inhibition,given current evidence of BNST function and development in human research and animal *** with behavioural inhibition have an increased startle response to safety cues and an increased cortisol response to social evaluative situations,both of which are mediated by the *** rodents,activation of the BNST underlies contextual fear responses and responses to uncertain and sustained ***-human primates with anxious temperament(the macaque equivalent of behavioural inhibition)have increased BNST activity to ambiguous social situations,and activity of the BNST in anxious temperament is significantly ***,the BNST is sexually dimorphic and continues to develop into adulthood,paralleling the development of anxiety disorders in ***,these findings suggest that further investigation of the BNST in behavioural inhibition is necessary and may lead to new avenues for the prevention and treatment of anxiety disorders.

Production of pectic extracts from sugar beet pulp with antiproliferative activity on a breast cancer cell line

In the last years, sugar beet pectins have been the subject of several investigations involving extraction methodologies, chemical composition and functional properties. The structure of pectins, which depends on the extraction method, is decisive in their capacity to induce apoptosis on several cancer cell lines like colon, prostate and breast. In this work, sugar beet pectin extraction was performed in the following steps： lipid extraction with hexane, removal of soluble complex carbohydrates and proteins, and enzymatic treatment with amyloglucosidase, protease, and pectinase. The enzymatic treatment was carried out with Rohapect DA6L under the following conditions： 50℃, pH 4.0, 2% enzyme/substrate （E/S） ratio, 15 h, and a solid to liquid ratio of 1 ： 10. The pectic extract showed a degree of polymerization （DP） profile of 55.8% with DP≥7; 4.9% with DP6; 5.8% between DP2 and DP6 ; 4.7% with DP2; and 28.8% with DP1. The pectic extract was examined for its antiproliferative activity on the MCF-7 breast cancer cell line. At a concentration range of 12.5-25 mg/mL the pectic extract killed 80.6% of the cells, exhibiting a higher antiproliferative activity than 4-hydroxytamoxifen （4- OHT）, a classical anticancer drug, which killed 56.5% of the cells.

Development of new classes of plasmon active nano-structures and their application in bio-sensing and energy guiding

Metal nanostructures exhibit special optical resonance modes originating fronl tile subwavelengt.h confinement of conductive electrons in the material. These resonance modes represent a strong research focus due to their application potential in optics and sensing application, in this short review recent achievements of our group in this field are highlighted. A wet-chemistry approach synthesis of advanced metallic nanostruetures will be introduced and their exact positioning and manipulation by electric field is shown. Next. the application of these nanostructures for a detection of small molecules will be described in several examples. Also, it will be shown that inetal nanostruetures can be used for sub-wavelength light focusing and for efficient energy coupling into polymer chains.

Parametric instability of a liquid metal sessile drop under the action of low-frequency alternating magnetic fields

A 2-D mathematical model is developed in order to simulate a parametric electromagnetic instability oscillation process of a liquid metal droplet under the action of low frequency magnetic field. The Arbitrary Lagrangian-Eulerian （ALE） method and weak form constraint boundary condition are introduced in this model for implementation of the surface tension and electromagnetic force on liquid droplet free surface. The results of the numerical calculations indicate the appearance of various regimes of oscillation. It is found that according to the magnetic field frequency various types of oscillation modes may be found. The oscillation is originated from an instability phenomenon. The stability diagram of liquid metal droplet in the parameter space of magnetic frequency and magnetic flux density is determined numerically. The diagram is very similar to that found in the so-called parametric instability.

BACKGROUND Paraspinal muscle strength and fatigue are considered important in low back pain(LBP)prevention and *** reliability of paraspinal strength and electromyographic(EMG)-fatigue parameters has not been universally ***,the discriminative validity of these parameters requires further exploration,under the threat of potentially poor reliability of the methods *** To investigate the reliability and discriminative validity of paraspinal strength and EMG-related fatigue in subjects with recurrent LBP and healthy *** Test-retest measurements were performed in 26 healthy and 66 LBP volunteers,for *** isometric maximal and mean strength were determined with a maximum voluntary isometric contraction(MVIC)protocol,performed in a custom-made *** the fatigue test,participants performed a 60% MIVC level continuous isometric contraction of the paraspinals,in conjunction with EMG analysis from 4 muscle sites of the lumbar *** median frequency(IMF),the median frequency slope(MFslope),as well as the root mean square(RMS)slope EMG parameters were used as fatigue *** were analysed with repeated measures ANOVA for test-retest *** reliability,the intraclass correlation coefficient(ICC3,1),standard error of the measurement(SEM)and the smallest detectable difference(SDD)were ***-related differences for fatigue measures were analysed with a Multivariate Analysis of Covariance,with age,weight and strength as *** Isometric strength presented statistically significant between-day differences(P<0.01),however these did not exceed 10%(healthy:7.2%/LBP-patients:9.7%)and ICC reliability values were excellent,yet test-retest error was increased for the patient group(healthy:ICC3,1:0.92-0.96,SEM:5.72-5.94 Hz,SDD:18.51%-18.57%/LBP-patients:ICC3,1:0.91-0.96,SEM:6.49-6.96,SDD:30.75%-31.61%).For the frequency data,IMF reliability was excellent(healthy:ICC3,1:0
