Glycosyltransferases of the Cellulose Synthase Like D （CS/D） subfamily have been reported to be involved in tip growth and stem development in Arabidopsis. The csld2 and csld3 mutants are root hair defective and the csld5 mutant has reduced stem growth. In this study, we produced double and triple knockout mutants of CSLD2, CSLD3, and CSLD5. Unlike the single mutants and the csld2/csld3 double mutant, the csld2/csld5, csld3/csld5, and csld2/csld3/csld5 mutants were dwarfed and showed severely reduced viability. This demonstrates that the cooperative activities of CSLD2, CSLD3, and CSLD5 are required for normal Arabidopsis development, and that they are involved in important processes besides the specialized role in tip growth. The mutant phenotypes indicate that CSLD2 and CSLD3 have overlapping functions with CSLD5 in early plant development, whereas the CSLD2 and CSLD3 proteins are non-redundant. To determine the biochemical function of CSLD proteins, we used transient expression in tobacco leaves. Microsomes containing heterologously expressed CSLD5 transferred mannose from GDP-mannose onto endogenous acceptors. The same activity was detected when CSLD2 and CSLD3 were coexpressed but not when they were expressed separately. With monosaccharides as exogenous acceptors, microsomal preparations from CSLD5-expressing plants mediated the transfer of mannose from GDP-mannose onto mannose. These results were supported by immunodetection studies that showed reduced levels of a mannan epitope in the cell walls of stem interfascicular fibers and xylem vessels of the csld2/csld3/csld5 mutant.

RNA-Seq Analysis of Developing Nasturtium Seeds （Tropaeolum majus）： Identification and Characterization of an Additional Galactosyltransferase Involved in Xyloglucan Biosynthesis

A deep-sequencing approach was pursued utilizing 454 and Illumina sequencing methods to discover new genes involved in xyloglucan biosynthesis, cDNA sequences were generated from developing nasturtium （Tropaeolum majus） seeds, which produce large amounts of non-fucosylated xyloglucan as a seed storage polymer. In addition to known xyloglucan biosynthetic genes, a previously uncharacterized putative xyloglucan galactosyltransferase was iden- tified. Analysis of an Arabidopsis thaliana mutant line defective in the corresponding ortholog （AT5G62220） revealed that this gene shows no redundancy with the previously characterized xyloglucan galactosyltransferase, MUR3, but is required for galactosyl-substitution of xyloglucan at a different position. The gene was termed XLT2 for Xyloglucan L-side chain galactosylTransferase position 2. It represents an enzyme in the same subclade of glycosyltransferase family 47 as MUR3. A double mutant defective in both MUR3 （mur3.1） and XLT2 led to an Arabidopsis plant with xyloglucan that consists essentially of only xylosylated glucosyl units, with no further substitutions.

This paper presents the power hardware in the loop(PHIL)validation of a feed forward DC voltage control scheme for the fault ride through(FRT)of voltage source converter(VSC)high voltage DC(HVDC)connected offshore wind power plants(WPPs).In the proposed FRT scheme,the WPP collector network AC voltage is actively controlled by considering both the DC voltage error and the AC current from the WPP AC collector system which ensures fast and robust FRT of the VSC HVDC connected offshore *** PHIL tests were carried out in order to verify the efficacy of the proposed feed forward DC voltage control scheme for enhancing the FRT capability of the VSC HVDC connected *** PHIL test results have demonstrated the proper control coordination between the offshore WPP and the WPP side VSC and the efficient FRT of the VSC HVDC connected WPPs.

To achieve active control of the AC voltage magnitude of wind power plant(WPP)collector network and improve the fault ride-through(FRT)capability,an FRT scheme based on feed forward DC voltage control is presented for voltage source converter-high voltage direct current(VSC-HVDC)connected offshore *** steady state operation,an open loop AC voltage control is implemented at the WPP-side VSC of the HVDC system so that any possible control interactions between WPP-side VSC and VSC of wind turbine are *** during any grid fault,a dynamic AC voltage reference is made according to both the DC voltage error and AC active current from the WPP collector system,thus ensuring fast and robust FRT of the VSC-HVDC-connected offshore *** the unbalanced fault condition in the host power system,the resulting oscillatory DC voltage is directly used in the VSC AC voltage controller at the WPP side so that the WPP collector system voltage also reflects the unbalance in the main *** domain simulations are performed to verify the efficacy of the FRT scheme based on the proposed feed forward DC voltage *** results show satisfactory FRT responses of the VSC-HVDC-connected offshore WPP under balanced and unbalanced faults in the host power system,as is shown under a serious fault in the WPP collector network.

ECG is not a reliable predictor of sudden cardiac death in the general population

Objectives: To determine the predictive value of the ECG for sudden death in the general population. Design: In the Copenhagen City Heart Study, a randomly selected population sample in Copenhagen, Denmarkhas been followed prospectively since 1976. From this population sample, we analyzed ECGs of individuals who had suffered sudden cardiac death (SCD) before the age of 50 years and compared them with ECGs of a randomly selected control individuals from the same population sample. Specific ECG signs that could point toward a condition associated with a risk of SCD were noted. Results: From a total of 18,974 individuals in the cohort, 207 had died at an age younger than 50 years. Among these, 24 persons with SCD were identified. The most prevalent ECG abnormality was QRS fragmentation. We found no ECGs with long or short QTc, Brugada sign or WPW. The prevalence of signs of left ventricular hyper-trophy, early repolarization, or fragmentation was not different from the prevalence of these signs in the control group. Conclusion: In the Copenhagen City Heart Study, the ECG failed to predict SCD in persons who died before the age of 50 years.

Melatonin for Pre- and Postoperative Pain and Anxiety: A Cancelled Clinical Trial

Designing and completing clinical intervention trials can be challenging. Many aspects must be considered to ensure that patients who fulfill the inclusion criteria for the intervention are identified and recruited effectively. The aim of this paper was to disseminate the results of a cancelled trial and present unpredictable barriers met underway, so future researchers can learn from these. The trial examined perioperative analgesic and anxiolytic effects of melatonin. It was registered at https://***/ (NCT02386319) and a study protocol was published a priori. Participants were recruited from the plastic surgery ward of a Danish private hospital. The intended sample size of the trial was 72 patients based on power calculations of the outcome measures. During the six-month recruitment period, six patients were included, with only three completing the trial. Unpredictable barriers were poor communication between investigators and facility staff, lack of access to booking and operation schedules at the recruitment facility, the patient group being unwilling to participate, and the timing of recruitment conversations being unsuited as patients often did not have time to talk to the investigators. Too few data were collected to make any meaningful statistical analyses. Our trial was cancelled prematurely because of unpredictable barriers after commencing recruitment. Considering these barriers when designing a clinical trial may help future researchers avoid cancelling trials. Transparency of research is important and even prematurely cancelled trials should publish their findings.
