Matched pair analysis of the effect of longer hypothermic machine perfusion time on kidney transplant outcomes

BACKGROUND Hypothermic machine perfusion(HMP)has demonstrated benefits in terms of early kidney transplant function compared to static cold *** longer preservation times have shown detrimental effects,a previous paired study indicated that longer pump times(the second kidney in a pair)might lead to improved *** To revisit the prior paired study's somewhat unexpected results by reviewing our program's *** A total of 61 pairs of transplant recipients who received kidneys from the same donor(2012-2021)were *** were divided into two groups depending on whether they were transplanted first(K1)or second(K2).Therefore,the patients in each pair had identical donor characteristics,except for time on the *** analyses included Kaplan-Meyer analysis and paired tests,including McNemar's test,student's paired t-test,or Wilcoxon's test,as *** The two groups of recipients had similar demographics(age,body mass index,diabetes,time on dialysis,sensit-ization and retransplants).Cold ischemic times for K1 and K2 were 8.9(95%CI:7.9,9.8)and 14.7 hours(13.7,15.8)(Per rate of freedom from biopsy-proven acute rejection at 1 year(P=0.015).Delayed graft function was less common in K2,12/61(20%)than in K1,20/61(33%)(P=0.046).Finally,K2 showed a higher graft survival than K1(P=0.023).CONCLUSION Our results agree with a previous study that suggested possible advantages to longer pump *** studies should encourage further research into HMP's potential anti-inflammatory effect.

Genome-wide markers for seed yield and disease resistance in perennial ryegrass

The market success of perennial ryegrass(Lolium perenne L.) cultivars depends on sufficient seed production, as they are propagated by seed. However, breeding for high quality forage production reduces seed yield, and breaking the negative correlation would help to overcome the problem. The foliar disease crown rust is another factor affecting reproductive capacity and thereby seed yield. We evaluated seed yield-related traits and resistance to crown rust in a collection of commercial cultivars and ecotypes of perennial ryegrass and identified genome-wide markers associated with the traits. The study revealed high variation between the ecotype and cultivar groups as well as between years. A genome-wide association study identified 17 DNA single-nucleotide polymorphisms(SNPs) of which eight were associated with crown rust and nine with flag-leaf length. The SNP markers were located within or near predicted genes functioning in defense against pathogens. The identified genes are strong candidates for a further in-depth functional study to continue unravel determination of leaf architecture and crown rust resistance in perennial ryegrass.

目的探讨山楂叶总黄酮对血管紧张素Ⅱ(AngⅡ)诱导的心肌肥大的影响,及其可能作用机制。方法将H9c2细胞分为对照组(正常培养),模型组(100 nmol·L^(-1)AngⅡ处理24 h),低、中、高剂量实验组(先给予100 nmol·L^(-1)AngⅡ处理24 h后,再分别给予25、50、100 mg·L^(-1)的山楂叶总黄酮处理24 h),anti-miR-NC、anti-miR-21a-5p组(分别转染anti-miR-NC、anti-miR-21a-5p后,给予100 nmol·L^(-1)AngⅡ处理24 h),miR-NC+高剂量组和miR-21a-5p+高剂量组(分别转染miR-NC、miR-21a-5p mimics,再给予100 nmol·L^(-1)AngⅡ+100 mg·L^(-1)山楂叶总黄酮各处理24 h)。用CCK-8法和流式细胞术检测细胞活力和凋亡情况,用实时荧光定量聚合酶链反应法检测miR-21a-5p的表达水平,用蛋白质印迹法检测环氧化酶-2(COX2)/前列腺素e2(PGe2)的表达水平。结果对照组、模型组和高剂量实验组的细胞活力分别为1.03±0.09、0.51±0.05和0.93±0.08,凋亡率分别为(7.69±0.61)%、(23.04±1.82)%和(9.43±0.71)%,miR-21a-5p表达水平分别为1.00±0.09、2.43±0.18和1.09±0.08,COX2蛋白相对表达水平分别为0.42±0.03、0.85±0.08和0.40±0.04,PGe2蛋白相对表达水平分别为0.34±0.03、0.75±0.07和0.35±0.03,模型组的上述指标与高剂量实验组和对照组相比,在统计学上差异均有统计学意义(均Pe2蛋白相对表达水平分别为0.74±0.06、0.38±0.07、0.36±0.02、0.71±0.05,anti-miR-21a-5p组的上述指标与anti-miR-NC组比较,miR-21a-5p+高剂量组的上述指标与miR-NC+高剂量组比较,在统计学上差异均有统计学意义(均Pe2通路的表达,从而抑制AngⅡ诱导的心肌肥大。

Association between Tropical Convection and Boreal Wintertime extratropical Circulation in 1982/83 and 1988/89

Boreal wintertime extratropical circulation is studied in relation to the tropical convection during the 1982/83 el Niño and 1988/89 La Niña. The anomaly structure of 1982/83 and 1988/89 over the extratropics reveals remarkably different features as the longitudinal tropical forcing region changes. The Rossby wave source (Positive) shows the largest maximum over east Asia in both years due to the persistent heating from the western Pacific warm pool area. However, the sink term shows contrasting features over the subtropics and extratropics between the two years. In the el Niño year, enhanced tropical convection over the eastern Pacific produces the Rossby wave sink at 10°N and shifted eastward over the North Pacific, while in the La Niña year, the sink area is shifted westward over the North Pacific. The contrasting features between the two events in mean-eddy interaction appears especially over the downstream area of the east Asian Jet. The extension (retraction) of the meanflow eastward (westward) to the east (west) of the dateline is related with the effect of the westward (eastward) e-vector and the strengthening (weakening) of the negative anomalies of the barotropic growth of kinetic energy. Hence, almost opposite characteristics between the two events can explain the close relationship of tropical convection and the extratropical internal variability.

Delayed ejaculation is a rare condition with different etiologies. Majority of cases of delayed ejaculation have a strong psychological/behavior component. A 48-year-old male with a diagnostic of delayed ejaculation acquired was encouraged to make an exercise protocol using a masturbation device for 8 wk, the percentage of intercourse successful a month after finish protocol exercise was 81%.

Impact of the microenvironment on the pathogenesis of mucosaassociated lymphoid tissue lymphomas

Approximately 8%of all non-Hodgkin lymphomas are extranodal marginal zone B cell lymphomas of mucosa-associated lymphoid tissue(MALT),also known as MALT *** arise at a wide range of different extranodal sites,with most cases affecting the stomach,the lung,the ocular adnexa and the *** small intestine is involved in a lower percentage of *** growth in the early stages is associated with long-lasting chronic inflammation provoked by bacterial infections(e.g.,Helicobacter pylori or Chlamydia psittaci infections)or autoimmune conditions(e.g.,Sjögren’s syndrome or Hashimoto thyroiditis).While these inflammatory processes trigger lymphoma cell proliferation and/or survival,they also shape the ***,activated immune cells are actively recruited to the lymphoma,resulting in either direct lymphoma cell stimulation via surface receptor interactions and/or indirect lymphoma cell stimulation via secretion of soluble factors like *** addition,chronic inflammatory conditions cause the acquisition of genetic alterations resulting in autonomous lymphoma cell ***,novel agents targeting the microenvironment have been developed and clinically tested in MALT lymphomas as well as other lymphoid *** this review,we aim to describe the composition of the microenvironment of MALT lymphoma,the interaction of activated immune cells with lymphoma cells and novel therapeutic approaches in MALT lymphomas using immunomodulatory and/or microenvironmenttargeting agents.

为探究LRR-XI-a基因家族在杜梨免疫系统中的作用机制,基于GDR数据库的杜梨基因组数据鉴定了杜梨LRR-XI-a家族成员,经进化特征、基因重复事件和表达模式分析筛选了响应腐烂病信号的成员,并结合果实瞬时表达和抗性基因表达分析对部分成员的功能进行了初步分析。结果表明,杜梨LRR-XI-a基因家族包含91个成员,进化上可分为6个亚组。基因重复分析结果表明,杜梨中GroupⅠ成员数的迅速增加可能由串联复制(TD)和全基因组复制(WGD)共同导致,而GroupⅥ成员的增加仅由WGD所致。顺式作用元件分析结果表明,杜梨LRR-XI-a亚组Ⅱ和Ⅵ可能主要参与杜梨对非生物逆境信号的响应,而其余4个亚组同时参与对生物和非生物逆境信号的响应。表达分析结果表明,LRR-XI-4和LRR-XI-5在腐烂病侵染后呈不同程度的上调表达,其中LRR-XI-5上调表达量可达对照的586倍以上。果实瞬时表达分析结果表明,过表达LRR-XI-4和LRR-XI-5降低了梨果实对腐烂病的抗性,抑制了ja信号通路基因的表达,但激活了超敏反应响应基因的表达。综上,基因重复导致杜梨LRR-XI-a家族成员发生迅速增加;LRR-XI-a家族成员响应不同的生物胁迫和非生物胁迫;LRR-XI-4和LRR-XI-5通过激活超敏反应(HR)相关基因的表达负调控腐烂病抗性,可作为后期腐烂病抗性研究的候选基因。本研究结果为后续开展梨抗病机理研究和分子育种奠定了基础。

An accelerated K-means clustering algorithm using selection and erasure rules

The K-means method is a well-known clustering algorithm with an extensive range of applications,such as biological classification,disease analysis,data mining,and image ***,the plain K-means method is not fast when the number of clusters or the number of data points becomes large.A modified K-means algorithm was presented by Fahim et al.(2006).The modified algorithm produced clusters whose mean square error was very similar to that of the plain K-means,but the execution time was *** this study,we try to further increase its *** are two rules in our method:a selection rule,used to acquire a good candidate as the initial center to be checked,and an erasure rule,used to delete one or many unqualified centers each time a specified condition is *** clustering results are identical to those of Fahim et al.(2006).However,our method further cuts computation time when the number of clusters *** mathematical reasoning used in our design is included.
