A Short Amino-Terminal Part of Arabidopsis Phytochrome A Induces Constitutive Photomorphogenic Response

Phytochrome A （phyA） is the dominant photoreceptor of far-red light sensing in Arabidopsis thaliana, phyA accumulates at high levels in the cytoplasm of etiolated seedlings, and light-induced phyA signaling is mediated by a com- plex regulatory network. This includes light- and FHY1/FHL protein-dependent translocation of native phyA into the nucleus in vivo. It has also been shown that a short N-terminal fragment of phyA （PHYA406） is sufficient to phenocopy this highly regulated cellular process in vitro. To test the biological activity of this N-terminal fragment of phyA in planta, we produced transgenic phyA-201 plants expressing the PHYA406-YFP （YELLOW FLUORESCENT PROTEIN）-DD, PHYA406- YFP-DD-NLS （nuclear localization signal）, and PHYA406-YFP-DD-NES （nuclear export signal） fusion proteins. Here, we report that PHYA406-YFP-DD is imported into the nucleus and this process is partially light-dependent whereas PHYA406-YFP-DD-NLS and PHYA406-YFP-DD-NES display the expected constitutive localization patterns. Our results show that these truncated phyA proteins are light-stable, they trigger a constitutive photomorphogenic-like response when localized in the nuclei, and neither of them induces proper phyA signaling. We demonstrate that in vitro and in vivo PHYA406 Pfr and Pr bind COP1, a general repressor of photomorphogenesis, and co-localize with it in nuclear bodies. Thus, we conclude that, in planta, the truncated PHYA406 proteins inactivate COP1 in the nuclei in a light-independent fashion.

Hence, this tutorial review should continue from where it left off and present examples concerning the usage of solar power curves. In a nutshell, this tutorial review,together with the preceding one, should elucidate how surface shortwave radiation data, be they ground-based, satelliteretrieved, or model-output, are bridged to various power system operations via solar power curves.

Impact of Clocking on the Aero-Thermodynamics of a Second Stator tested in a One and a Half Stage HP Turbine

This paper focuses on the experimental investigation of the time-averaged and time-accurate aero- thermodynamics of a second stator tested in a 1.5 stage high-pressure turbine. The effect of clocking on aerodynamic and heat transfer are investigated. Tests are performed under engine representative conditions in the VKI compression tube CT3. The test program includes four different clocking positions, i.e. relative pitch-wise positions between the fh-st and the second stator. Probes located upstream and downstream of the second stator provide the thermodynamic conditions of the flow field. On the second stator airfoil, measurements are taken around the blade profile at 15, 50 and 85% span with pressure sensors and thin-film gauges. Both time-averaged and time-resolved aspects of the flow field are addressed. Regarding the time-averaged results, clocking effects are mainly observed within the leading edge region of the second stator, the largest effects being observed at 15% span. The surface static pressure distribution is changed locally, hence affecting the overall airfoil performance. For one clocking position, the thermal load of the airfoil is noticeably reduced. Pressure fluctuations are attributed to the passage of the up- stream transonic rotor and its associated pressure gradients. The pattern of these fluctuations changes noticeably as a function of docking. The time-resolved variations of heat flux and static pressure are analyzed together showing that the major effect is due to a potential interaction. The time-resolved pressure distribution integrated along the second stator surface yields the unsteady forces on the vane. The magnitude of the unsteady force is very dependent on the clocking position.

Drying of suspensions and solutions on inert particle surface in mechanically spouted bed dryer

To eliminate some disadvantages of the conventional spouted bed dryers the mechanically spouted bed （MSB） system was developed. This dryer type is convenient to use inert particles providing an increased surface area for drying of materials of high-moisture content and heat sensitive materials. On three different drying tasks are demonstrated the experimental optimization of process parameters to obtain products of demanded quality. The main object was at drying of AIO（OH） suspension to preserve the particle size under 2.5μm and to obtain product with a moisture content of about 0.05 kg/kg （d.b.）. For this reason a very thin particle coating and intensive abrasion had to be assured. At drying of tomato concentrates the thermoplasticity makes the process very difficult. To jump over the deliquescent and sticky state developed at the critical temperature-moisture content values a very short drying time （8-10 s） must be provided. The third task was to form powder-like product from bovine serum albumin （BSA） solution having very low solid content （2-4%）. The selected process parameters given in this paper resulted in a mean particle size of less than 20 μm while the soluble oreserved orotein content was higher than 90%.

Start the Earth System Governance that Integrates “Climate-Altering Technologies”

Addressing climate emergencies requires a radical social change, and an “earth system” governance approach that combines different factors(including technologies that affect climate). As the Paris Agreement has been reached for four years and came into force for three years, there is a growing recognition that the global average temperature rise cannot be limited to 1.5–2°C only by emissions reduction or existing carbon removal measures. The reason is that the world has not taken enough actions to deal with the crisis. As reported by IPCC(2018), hundreds of millions of people worldwide are already experiencing the harsh consequences of climate change, from storms to floods, to heatwaves and droughts [IPCC. 2018. “International Panel on Climate Change Special Report on Global Warming of 1.5 Degrees.” https://***/sr15(accessed November 5, 2019)]. According to Spratt and Dunlop(2019), as the climate change intensifies,all sectors of society have realized the need to avoid the risks brought by climate change and to deal with the severe disasters that already exist [Spratt, David, and Ian Dunlop. 2019.“Existential Climate-related Security Risk: A Scenario Approach.” https://***/ugd/***(accessed November 5, 2019)]. This raises some profound questions. For instances, should people consider a new responsive measure that is compatible with the natural system which sustains life on the earth, and within the limits of the earth’s tolerance? What forms of decision-making might we need, to help us make the smart collective choices needed for a world where no risk-free options remain? Are familiar governance and decision-making processes still suitable for the goals? Who will make the decisions to promote this transformation? If people really want to change the way they make decisions, they may need to create new forms of governance and decide how this transformation begins and which authority is subject to.
