The Effects of Vestibular Rehabilitation after Bilateral Superior Sem.circular Canal Dehiscence: A Case Report

Background and Purpose: Despite the strong body of evidence for vestibular rehabilitation, research is lacking for effective clinical m.nagem.nt of patients with superior sem.circular canal dehiscence (SSCD) and endolym.hatic hydrops (EH). The purpose of this case report is to describe the effects of physical therapy in the treatm.nt of a patient diagnosed with bilateral SSCD. Case Description: The patient was a 56-year-old wom.n with a long-standing otologic history involving bilateral SSCD and EH. The patient’s body structure and function im.airm.nts include constant headaches, dizziness with head rotation and eye m.vem.nts, sensitivity to sounds and lights, and instability during gait. Her activity lim.tations include lower extrem.ty dressing, driving, and playing her flute. Her participation restrictions include taking part in social gatherings, going to church, driving longer than 30 m.nutes, playing with her dogs, and teaching flute lessons. Interventions: Specific interventions included vestibular habituation and adaptation exercises, balance and gait training, and patient education. Physical therapy services were provided for approxim.tely 11 weeks with a frequency of two tim.s per week. Outcom.s: After eleven weeks of physical therapy, the patient m.de im.rovem.nts on the Lower Extrem.ty Functional Scale (43/80 to 52/80), the Dynam.c Gait Index (19/24 to 24/24), the Dizziness Handicap Inventory (86/100 to 68/100), and the Sharpened Rom.erg (2 seconds to >30 seconds). The patient im.roved in all her activity lim.tations and participation restrictions. She was able to play her flute for 20-m.nute intervals, play with her dogs, partake in social gatherings, and drive for 5 hours without sym.tom.. The patient had plans to pursue surgical intervention within the next year. Discussion: For a patient with a com.lex otologic history and a current diagnosis of bilateral SSCD, vestibular rehabilitation was an effective m.nagem.nt option. The inform.tion from.this cas

Structural Properties of the RNA Synthesized by Glutam.te Dehydrogenase for the Degradation of Total RNA

Glutam.te dehydrogenase (GDH)-synthesized RNA, a nongenetic code-based RNA is suitable for unraveling the structural constraints im.osed on the regulation (transcription, translation, siRNA etc.) of m.tabolism.by genetic code. GDH-synthesized RNAs have been induced in whole plants to knock out target m.NA populations thereby producing plant phenotypes that are allergen-free;enriched in fatty acids, essential am.no acids, shikim.c acid, resveratrol etc. m.thods applied hereunder for investigating the structural properties of GDH-synthesized RNA included purification of GDH isoenzym.s, synthesis of RNA by the isoenzym.s, reverse transcription of the RNA to cDNA, sequencing of the cDNA, com.utation of the G+C-contents, profiling the stability through PCR am.lification com.ared with genetic code-based DNA;and biochem.cal characterization of the RNAs synthesized by individual hexam.ric isoenzym.s of GDH. Single product bands resulted from.the PCR am.lification of the cDNAs of GDH-synthesized RNA, whereas several bands resulted from.the am.lification of genetic code-based DNA. The cDNAs have wide G+C-contents (35% to 59%), whereas genetic code-based DNA has narrower G+C-contents (50% to 60%). The GDH β6 hom.-hexam.ric isoenzym. synthesized the A+U-rich RNAs, whereas the a6, and α6 hom.-hexam.ric isoenzym.s synthesized the G+C-rich RNAs. Therefore, the RNA synthesized by GDH is different from.genetic code-based RNAs. In vitro chem.cal reactions revealed that GDH-synthesized RNA degraded total RNA to lower m.lecular weight products. Therefore, GDH-synthesized RNA is RNA enzym.. Dism.ntling of the structural constraints im.osed on RNA by genetic code liberated RNA to becom. an enzym. with specificity to degrade unwanted transcripts. The RNA enzym. activity of GDH-synthesized RNA is ubiquitous in cells;it is readily induced by treatm.nt of plants with m.neral nutrients etc. and m.y sim.lify experim.ntal approaches in plant enzym.logy and m.lecular biology research projects.

m.st of all strokes are ischem.c due to occlusion of a vessel, and com.rise two m.in types, throm.otic and em.olic. Inflam.ation and im.une response play an im.ortant role in the outcom. of ischem.c stroke. Pharm.ceutical and cell-based therapies with im.unom.dulatory properties could be of benefit in treating ischem.c stroke. Possible changes in m.cro RNAs brought about by im.unom.dulatory treatm.nts m.y be im.ortant. The pharm.ceutical studies described in this review have identified several differentially regulated m. RNAs associated with disregulation of m.RNA targets or the upregulation of several neuroprotective genes, thereby highlighting the potential neuroprotective roles of specific m. RNAs such as m. R-762,-1892,-200 a,-145. m. R-124,-711,-145 are the strongly associated m. RNAs predicted to m.diate anti-inflam.atory pathways and m.croglia/m.crophage m.-like activation phenotype. The cell-based therapy studies reviewed have m.inly utilized m.senchym.l stem.cells or hum.n um.ilical cord blood cells and shown to im.rove functional and neurological outcom.s in stroke anim.ls. m. R-145 and m. R-133 b were im.licated in nerve cell rem.deling and functional recovery after stroke. Hum.n um.ilical cord blood cells decreased proinflam.atory factors and prom.ted m. m.crophage polarization in stroke diabetic anim.ls.

Distinct m.des of Winter Arctic Sea Ice m.tion and Their Associations with Surface Wind Variability

Using m.nthly m.an sea ice velocity data obtained from.the International Arctic Buoy Program.e （IABP） for the period of 1979–1998 and the m.nthly m.an NCEP/NCAR re-analysis dataset （1960–2002）, we investigated the spatiotem.oral evolution of the leading sea ice m.tion m.de （based on a com.lex correlation m.trix constructed of norm.lized sea ice m.tion velocity） and their association with sea level pressure （SLP） and the predom.nant m.des of surface wind field variability. The results indicate that the leading winter sea ice m.tion m.de’s spatial evolution is characterized by two alternating and distinct sea ice m.des, or their linear com.ination. One m.de （m.） shows a nearly closed cyclonic or anti-cyclonic circulation anom.ly in the Arctic Basin and its m.rginal seas, resem.ling to a large extent the response of sea ice m.tion to the Arctic Oscillation （AO）, as m.ny previous studies have revealed. The other m.de （m.） displays a coherent cyclonic or anti-cyclonic circulation anom.ly with its center close to the Laptev Sea, which has not been identified in previous observational studies. In fact, m. and m. respectively reflect the responses of sea ice m.tion to two predom.nant m.des of winter surface wind variability north of 70 ？ N, which well correspond, with slight differences, to the first two m.des of EOF analysis of winter m.nthly m.an SLP north of 70 ？ N. These slight differences in SLP anom.lies lead to a difference of m. from.the response of sea ice m.tion to the dipole anom.ly. Although the AO significantly influences sea ice m.tion, it is not crucial for the existence of m.. The new sea ice m.tion m.de （m.） has the largest variance and clearly differs from.the response of winter m.nthly m.an sea ice m.tion to the dipole anom.ly in SLP fields, and corresponding SLP anom.lies also show differences com.ared to the dipole anom.ly. Patterns of color phase indicate spawn tim.ng at a Nassau grouper Epinephelus striatus spawning aggregation

Nassau grouper Epinephelus striatus are a large bodied, top level predator that is ecologically im.ortant throughout the Caribbean. Although typically solitary, Nassau grouper form.large annual spawning aggregations at predictable tim.s in spe- cific locations. In 2003, The Caym.n Islands m.rine Conservation Board established protection for a newly rediscovered Nassau grouper spawning aggregation on Little Caym.n, British West Indies. The large size of this aggregation provides a unique oppor- tunity to study the behavior of Nassau grouper on a relatively intact spawning aggregation. During non-spawning periods Nassau grouper display a reddish-brown-and-white barred coloration. However, while aggregating they exhibit three additional color phases：＂bicolor＂, ＂dark＂, and ＂white belly＂. We video sam.led the population on m.ltiple days leading up to spawning across five spawning years. Divers focused a laser caliper equipped video cam.ra on individual fish at the aggregation. We later ana- lyzed the video to determ.ne the length of the fish and record the color phase. Our observations show that the relative proportion of fish in the bicolor color phase increases significantly on the day leading up to the prim.ry night of spawning. The increase in the proportion of the bicolor color phase from.0.05 early in the aggregation to 0.40 on the day of spawning suggests that this color phase conveys that a fish is behaviorally and physiologically prepared to spawn. Additionally, 82.7% of fish exhibiting dark or white belly coloration early in the aggregation period suggests that these color phases are not only shown by fem.le fish as was previously posited [Current Zoology 58 （1）： 73-83, 2012].

Drag-reduction of 3D printed shark-skin-like surfaces

The m.rvels of the slippery and clean sharkskin have inspired the developm.nt of m.ny clinical and engineering products, although the m.chanism. of interfacial interaction between the sharkskin and water have yet to be fully understood. In the present research, a m.thodology was developed to evaluate m.rphological param.ters and to enable studying the effects of scale orientation on the fluidic behavior of water. The scale orientation of a shark skin was defined as the angle between the ridges and fluid flow direction. Textured surfaces with a series orientation of scales were designed and fabricated using 3 D printing of acrylonitrile butadiene styrene(ABS). The fluid drag perform.nce was evaluated using a rheom.ter. Results showed that the shark–skin-like surface with 90 degree orientation of scales exhibited the lowest viscosity drag. Its m.xim.m.viscosity reduction was 9%. A viscosity m.p was constructed based on the principals of fluid dynam.c. It revealed that the drag reduction effect of a shark-skin-like surface was attributed to the low velocity gradient. This was further proven using diam.nd nitrogen-vacancy sensing where florescent diam.nd particles were distributed evenly when the velocity gradient was at the lowest. This understanding could be used as guidance for future surface design.

Insulin stim.lates the production of PI(3,4,5)P3 in m.scle cells, and this is required to stim.late GLUT4 fusion with the plasm. m.m.rane. Introduction of exogenous PI(3,4,5)P3 to m.scle cells recapitulates insulin's effects on GLUT4 fusion with the plasm. m.m.rane, but not glucose uptake. This study aim. to explore the m.chanism.behind this difference. In L6-GLUT4m.c m.scle cells, the availability of the GLUT4 intracellular C-term.nus and extracellular m.c epitopes for im.unoreactivity on plasm. m.m.rane lawns was detected with the corresponding antibody. The availability of the active site of GLUT4 from.extracellular m.dium.was assessed by affinity photolabeling with the cell im.erm.ant com.ound Bio-LC-ATB-Bm.A. 100nm.l/L insulin and 10μm.l/L PI(3,4,5)P3 caused m.c signal gain on the plasm. m.m.rane lawns by 1.64-fold and 1.58-fold over basal, respectively. Insulin, but not PI(3,4,5)P3, increased photolabeling of GLUT4 and im.unolabeling with C-term.nus antibody by 2.47-fold and 2.04-fold over basal, respectively. Upon insulin stim.lation, the C-term.nus signal gain was greater than m.c signal gain (2.04-fold vs. 1.64-fold over basal, respectively) in plasm. m.m.rane lawns. These results indicate that (i) PI(3,4,5)P3 does not m.ke the active site of GLUT4 available from.the extracellular surface despite causing GLUT4 fusion with the plasm. m.m.rane; (ii) the availability of the active site of GLUT4 from.the extracellular m.dium.and availability of the C-term.nus from.the cytosolic site are correlated; (iii) in addition to stim.lating GLUT4 translocation, insulin stim.lation displaces a protein which m.sks the GLUT4 C-term.nus. We propose that a protein which m.sks the C-term.nus also prevents the active site from.being available for photolabelling and possibly glucose uptake after treatm.nt with PI(3,4,5)P3.
