Excessive fructose diet is closely associated with colorectal cancer(CRC)***,fructose’s specific function and precise mechanism in colorectal cancer liver metastasis(CRLM)is rarely ***,this study reported that the fructose absorbed by primary colorectal cancer could accelerate CRLM,and the expression of KHK-A,not KHK-C,in liver metastasis was higher than in paired primary ***,KHK-A facilitated fructose-dependent CRLM in vitro and in vivo by phosphorylating PKM2 at ***2 phosphorylated by KHK-A inhibited its tetramer formation and pyruvic acid kinase activity but promoted the nuclear accumulation of *** and aerobic glycolysis activated by nuclear PKM2 enhance CRC cells’migration ability and anoikis resistance during CRLM ***-46 treatment,targeting the phosphorylation of PKM2,inhibited the pro-metastatic effect of ***,c-myc activated by nuclear PKM2 promotes alternative splicing of KHK-A,forming a positive feedback loop.

Transcriptome-Wide Identification and Validation of Reference Genes in Black Rockfish(Sebastes schlegelii)

The quantitative real-time reverse transcription PCR(qRT-PCR)is a widely used technique to analyze gene expression *** a suitable reference gene is a crucial step to obtain an accurate result in ***,most previous studies on fishes adopted reference genes that were commonly used in mammals without *** this study,we utilized 89 transcrip-tome datasets covering early developmental stages and different adult tissues,and carried out transcriptome-wide identification and validation of reference genes in Sebastes ***,121 candidate reference genes were identified based on four *** candidates(METAP2,BTF3L4,EIF5A1,TCTP,UBC,PAIRB,RAB10,and DLD)and four commonly used reference genes in mam-mals(TUBA,ACTB,GAPDH,RPL17)were selected for validation via qRT-PCR and four statistical analysis methods(delta-Ct,Best-Keeper,geNorm,and NormFinder).The results indicated that when the black rockfish are cultured in a general condition,the eight candidate reference genes are more stable than traditional reference genes in mammals,and RAB10,EIF5A1,PAIRB and BTF3L4 are the best reference genes in *** is the first study to conduct transcriptome-wide identification and validation of reference genes for quantitative RT-PCR in the black rockfish,and lay an important foundation for gene expression analysis in teleost.

Cavity-enhanced single quantum dots(QDs)are the main approach towards ultra-high-performance solid-state quantum light sources for scalable photonic quantum ***,harnessing the Purcell effect requires precise spectral and spatial alignment of the QDs’emission with the cavity mode,which is challenging for most *** we have successfully integrated miniaturized Fabry-Perot microcavities with a piezoelectric actuator,and demonstrated a bright single-photon source derived from a deterministically coupled QD within this *** the cavity-membrane structures,we have achieved large spectral tunability via strain *** resonance,a high Purcell factor of~9 is *** source delivers single photons with simultaneous high extraction efficiency of 0.58,high purity of 0.956(2)and high indistinguishability of 0.922(4).Together with its compact footprint,our scheme facilitates the scalable integration of indistinguishable quantum light sources on-chip,therefore removing a major barrier to the development of solid-state quantum information platforms based on QDs.

为了探究microRNAs(miRNAs)在斑马鱼生殖细胞发育过程中所起的作用,本研究在斑马鱼胚胎期过表达miR-430,结果发现原始生殖细胞(Primordial germ cells,PGCs)迁移出现紊乱,并且通过qRT-PCR发现nanos3、piwil2及tdrd7等生殖质基因的表达水平显著下调。此外,在斑马鱼性腺发育过程中,通过组织学观察发现过表达miR-430会导致雌性斑马鱼卵母细胞发育异常、卵巢发育迟缓,但是对雄性生殖细胞及性腺的发育没有造成明显的影响,并且进一步统计分析表明,miR-430不影响斑马鱼的性别分化。综上所述,本研究初步明确了过表达miR-430对斑马鱼PGCs迁移、性腺发育以及性别分化的影响,并为进一步探究miR-430在斑马鱼生殖细胞发育过程中的功能及分子机制奠定了理论基础。

现有婚姻市场理论无法有效解释部分农村流动青年“结婚难”问题。本次调查研究发现:选择现代婚恋模式的农村流动青年遇到的“结婚难”问题与婚姻市场理论下的“结婚难”问题不同,主要表现为恋爱难、由恋爱到结婚难。本文从婚恋失范视角对部分农村流动青年的“结婚难”问题进行解释,即因为婚恋观和恋爱行为缺乏社会规范指导和约束而导致“结婚难”问题。

Medical devices are instruments and other tools that act on the human body to aid clinical diagnosis and disease treatment,playing an indispensable role in modern ***,the increasing demand for personalized medical devices poses a significant challenge to traditional manufacturing *** emerging manufacturing technology of three-dimensional(3D)printing as an alternative has shown exciting applications in the medical field and is an ideal method for manufacturing such personalized medical devices with complex ***,the application of this new technology has also brought new risks to medical devices,making 3D-printed devices face severe challenges due to insufficient regulation and the lack of standards to provide guidance to the *** review aims to summarize the current regulatory landscape and existing research on the standardization of 3D-printed medical devices in China,and provide ideas to address these *** focus on the aspects concerned by the regulatory authorities in 3D-printed medical devices,highlighting the quality system of such devices,and discuss the guidelines that manufacturers should follow,as well as the current limitations and the feasible path of regulation and standardization work based on this *** key points of the whole process quality control,performance evaluation methods and the concept of whole life cycle management of 3D-printed medical devices are ***,the significance of regulation and standardization is pointed ***,aspects worthy of attention and future perspectives in this field are discussed.

Efficient polymeric room-temperature phosphorescence(PRTP)with excellent processability and flexibility is highly desirable but still faces formidable ***,a general strategy is developed for efficient PRTP through photo-polymerization of phosphor monomers and N-isopropylacrylamide(NIPAM)spontaneously without a *** ultralong lifetime of 3.54 s with afterglow duration time of 25 s and decent phosphorescent quantum efficiency of 13%are *** efficient PRTP has been demonstrated to be derived from the synergistic effect of the covalent and hydrogen bonds networks formed through photo-polymerization of *** electron paramagnetic resonance(EPR)spectra confirmed that methyl radicals are generated under the irradiation of ultraviolet light and promote the formation of covalent cross-linking *** strategy has also been proved to be generalizable to several other phosphor ***,the polymer films display ultrahigh temperature resistance with long afterglows even at 140℃ and unexampled ultralong lifetime of 2.45 s in aqueous *** work provides a simple and feasible avenue to obtain efficient PRTP.

白消安处理对红鳍东方鲀性腺的影响

为了获取生殖细胞移植的不育受体,本研究使用白消安(1,4-丁二醇二甲磺酸酯)对二龄半红鳍东方鲀(Takifugu rubripes)进行处理,使用40 mg/kg白消安注射处理两次后,进行了对照组、注射组的性腺取样和转录组测序,分析了白消安的处理效果和白消安对性腺基因表达的影响。研究表明:40 mg/kg白消安其对精巢的处理效果显著,生精小管中的生殖细胞大量减少;对卵巢的处理效果不佳,实验组与对照组的卵巢在组织形态上无显著差异。性腺转录组的分析结果显示,与注射二甲基亚砜(Dimethyl sulfoxide,DMSO)溶剂的对照组相比,经过白消安处理后,精巢组中获得差异表达基因576个,卵巢组中获得差异表达基因74个,白消安处理对精巢基因的转录水平影响更大。差异表达基因的GO(Gene Ontology)富集分析显示,氧化还原酶活性、单加氧酶活性、双加氧酶活性等氧化代谢相关条目被显著富集;KEGG(Kyoto Encyclopedia of Genes and Genomes)富集分析显示,氧化磷酸化、核苷酸切除修复、范可尼贫血、同源重组、DNA复制和错配修复等通路被显著富集。GSEA(Gene set enrichment analysis)结果显示,错配修复、范可尼贫血、同源重组等通路被上调,DNA复制和核苷酸切除修复通路下调。研究结果表明,白消安处理导致了雄性受体的内源生殖细胞数量减少,并使精巢处于DNA损伤和氧化应激的状态。不同性腺组织对白消安的不同反应可能与其细胞类型对氧化应激和DNA损伤的抵抗有关。
