Background and Aims:Hepatitis B surface antigen(HBsAg)loss is seldom achieved with nucleos(t)ide analog(NA)therapy in chronic hepatitis B patients but may be enhanced by switching to finite pegylated-interferon(Peg-IFN)*** assessed HBsAg loss with 48-and 96-week Peg-IFN alfa-2a in chronic hepatitis B patients with partial response to a previous ***:Hepatitis B e antigen(HBeAg)-positive patients who achieved HBeAg loss and hepatitis B virus DNA200 IU/mL with previous adefovir,lamivudine or entecavir treatment were randomized 1.1.to receive Peg-IFN alfa-2a for 48(n=1.3)or 96 weeks(n=1.0).The primary endpoint of this study was HBsAg loss at end of *** *** identifier is ***:At the end of 48 and 96 weeks'treatment,1..4%(22/1.3)and 20.7%(31.1.0)of patients,respectively,who switched from NA to Peg-IFN alfa-2a cleared *** were similar irrespective of prior NA or baseline HBeAg *** those who cleared HBsAg by the end of 48 and 96 weeks'treatment,77.8%(1./1.)and 71.4%(20/28),respectively,sustained HBsAg loss for a further 48 *** HBsAg1.00 IU/mL and week 24 HBsAg200 IU/mL were associated with the highest rates of HBsAg loss at the end of both 48-and 96-week treatment(51.4%and 58.7%,respectively).Importantly,extending treatment from 48 to 96 weeks enabled 48.3%(1./29)more patients to achieve HBsAg ***:Patients on long-term NA who are unlikely to meet therapeutic goals can achieve high rates of HBsAg loss by switching to Peg-IFN *** loss rates may be improved for some patients by extending treatment from 48 to 96 weeks,although the differences in our study cohort were not statistically *** and on-treatment HBsAg may predict HBsAg loss with Peg-IFN alfa-2a.

Background:Lipusu is the first commercialized liposomal formulation of pacli-taxel and has demonstrated promising efficacy against locally advanced lung squamous cell carcinoma(LSCC)in a small-scale ***,we conducted a multicenter,randomized,phase 3 study to compare the efficacy and safety of cis-platin plus Lipusu(LP)versus cisplatin plus gemcitabine(GP)as first-line treat-ment in locally advanced or metastatic ***:Patients enrolled were aged between 1. to 75 years,had locally advanced(clinical stage IIIB,ineligible for concurrent chemoradiation or surgery)or metastatic(Stage IV)LSCC,had no previous systemic chemother-apy and at least one measurable lesion as per the Response Evaluation Criteria in Solid Tumors(version 1.1.before administration of the trial *** primary endpoint was progression-free survival(PFS).The secondary endpoints included objective response rate(ORR),disease control rate(DCR),overall survival(OS),and safety *** explore the possible predictive value of plasma cytokines for LP treatment,plasma samples were collected from the LP group at baseline and first efficacy evaluation time and were then subjected to analysis by 45-Plex ProcartaPlex Panel 1.to detect the presence of 45 cytokines using the Luminex xMAP *** correlation between treatment outcomes and dynamic changes in the levels of cytokines were evaluated in preliminary ***:The median duration of follow-up was 1..4 months.237 patients in the LP group and 253 patients in the GP group were included in the per protocol set(PPS).In the PPS,the median PFS was 5.2 months versus 5.5 months in the LP and GP group(hazard rtio[HR]:1.03,P=0.742)*** median OS was 1..6 months versus 1..5 months in the LP and GP group(HR:0.83,P=0.21.).The ORR(41.8%versus 45.9%,P=0.41.)and DCR(90.3%versus 88.1.,P=0.443)were also similar between the LP and GP group.A significantly lower proportion of patients in the LP group experienced adverse events(AEs)le

Mechanism for ginkgolic acid(1.：1.-induced MDCK cell necrosis： Mitochondria and lysosomes damages and cell cycle arrest

Ginkgolic acids(GAs), primarily found in the leaves, nuts, and testa of ginkgo biloba, have been identified with suspected allergenic, genotoxic and cytotoxic properties. However, little information is available about GAs toxicity in kidneys and the underlying mechanism has not been thoroughly elucidated so far. Instead of GAs extract, the renal cytotoxicity of GA(1. : 1., which was isolated from the testa of Ginkgo biloba, was assessed in vitro by using MDCK cells. The action of GA(1. : 1. on cell viability was evaluated by the MTT and neutral red uptake assays. Compared with the control, the cytotoxicity of GA(1. : 1. on MDCK cells displayed a time-and dose-dependent manner, suggesting the cells mitochondria and lysosomes were damaged. It was confirmed that GA(1. : 1. resulted in the loss of cells mitochondrial trans-membrane potential(ΔΨm). In propidium iodide(PI) staining analysis, GA(1. : 1. induced cell cycle arrest at the G0/G1.and G2/M phases, influencing on the DNA synthesis and cell mitosis. Characteristics of necrotic cell death were observed in MDCK cells at the experimental conditions, as a result of DNA agarose gel electrophoresis and morphological observation of MDCK cells. In conclusion, these findings might provide useful information for a better understanding of the GA(1. : 1. induced renal toxicity.

Serotonin regulates brain-derived neurotrophic factor expression in select brain regions during acute psychological stress

Previous studies suggest that serotonin （5-HT） might interact with brain-derived neurotrophic factor （BDNF） during the stress response. However, the relationship between 5-HT and BDNF expression under purely psychological stress is unclear. In this study, one hour before psychological stress exposure, the 5-HT1. receptor agonist 8-OH-DPAT or antagonist MDL73005, or the 5-HT2A receptor agonist DOI or antagonist ketanserin were administered to rats exposed to psychological stress. Immunohistochemistry and in situ hybridization revealed that after psychological stress, with the exception of the ventral tegmental area, BDNF protein and mRNA expression levels were higher in the 5-HT1. and the 5-HT2A receptor agonist groups compared with the solvent control no-stress or psychological stress group in the CA1.and CA3 of the hippocampus, prefrontal cortex, central amygdaloid nucleus, dorsomedial hypothalamic nucleus, dentate gyrus, shell of the nucleus accumbens and the midbrain periaqueductal gray. There was no significant difference between the two agonist groups. In contrast, after stress exposure, BDNF protein and mRNA expression levels were lower in the 5-HT1. and 5-HT2A receptor antagonist groups than in the solvent control non-stress group, with the exception of the ventral tegmental area. Our findings suggest that 5-HT regulates BDNF expression in a rat model of acute psychological stress.

The TEA domain(TEAD)family proteins(TEAD1.4)are essential transcription factors that control cell differentiation and organ size in the Hippo *** the sequences and structures of TEAD family proteins are highly conserved,each TEAD isoform has unique physiological and pathological ***,the development and discovery of subtype selective inhibitors for TEAD protein will provide important chemical probes for the TEAD-related function studies in development and ***,we identified a novel TEAD 1.3 covalent inhibitor(DC-TEADin1.72)with biochemical IC50 values of 0.61.0.02 and 0.58±0.1.μmol/L against TEAD1.and TEAD3,*** chemical optimization based on DC-TEAD in 1.72 yielded a selective TEAD3 inhibitor DCTEAD3 in03 with the IC_(50) value of 0.1.±0.03μmol/L,which shows 1.0-fold selectivity over other TEAD isoforms in activity-based protein profiling(ABPP)*** cells,DC-TEAD3 in03 showed selective inhibitory effect on TEAD3 in GAL4-TEAD(1.4)reporter assays with the IC50 value of1.1.μmol/*** administered to zebrafish juveniles,experiments showed that DC-TEAD3 in03 reduced the growth rate of zebrafish caudal fins,indicating the importance of TEAD3 activity in controlling proportional growth of vertebrate appendages.

Controllable synthesis of novel nanoporous manganese oxide catalysts for the direct synthesis of imines from alcohols and amines

A novel template-free oxalate route was applied to synthesize different mesoporous manganese oxides(amorphous manganese oxide(AMO),Mn5 O8,Mn3 O4,Mn O2)in the narrow temperature range from 350°C to 400°C by controlling the calcination conditions,which were employed as the efficient catalysts for the oxidative coupling of alcohols with amines to *** chemical and structural properties of the manganese oxides were characterized by the methods of thermogravimetry analysis and heat flow(TG-DSC),X-ray diffraction(XRD),nitrogen sorption,scanning electron microscope(SEM),transmission electron microscopy(TEM),X-ray photoelectron spectroscopy(XPS),H2 temperature-programmed reduction(H2-TPR),and inductively coupled plasma optical emission spectrometry(ICP-OES)*** structures of different manganese oxides were confirmed by *** M-350(AMO)presented the maximum surface area,amorphous nature,the lowest reduction temperature,the higher(Mn3++Mn4+)/Mn2+ratio,and the higher adsorbed oxygen species compared to other *** the catalysts,M-350 showed the best catalytic performance using air as an oxidant,and the conversion of benzyl alcohol(BA)and the selectivity of N-benzylideneaniline(NBA)reached as high as 1.0%and 97.1.respectively at the lower reaction temperature(80°C)for 1.h.M-350 had also the highest TOF value(0.01.0 mmol·mg-1.h-1.compared to the other manganese oxide *** catalyst was reusable and gave 95.8%conversion after 5 reuse tests,the XRD pattern of the reactivated M-350 did not show any obvious *** oxygen mobility and(Mn3++Mn4+)/Mn2+ratio were found to play the important roles in the catalytic activity of aerobic reactions.

Although carbapenem use is prohibited in animals in china,carbapenem-resistant Escherichia coli(CREC),especially New Delhi metallo-β-lactamase(NDM)-producing strains,are widely prevalent in foodproducing *** present,the impact of livestock-associated CREC strains on {1.man populations at the national level is ***,we conduct a retrospective cross-sectional study to investigate the prevalence of CREC from clinical settings across 22 Chinese provinces or municipalities and analyze anthropogenic factors associated with their *** also ascertain the blaNDMand blaKPCabundance among pig and chicken farms and present a detailed genomic framework for CREC of animal and {1.man ***,631.29799(2.1.)clinical Escherichia coli(***)isolates were identified as *** analysis revealed that being male,an age below 1.an age between 1. and 1.,provinces with greater chicken production,and provinces with higher pig production were associated with higher odds of CREC *** general,73.8%(n=45/61.of pig farms and 62.2%(n=28/45)of chicken farms had a blaNDMabundance of 1.1.^(-5)to 1.1.^(-3)and 1.1.^(-3)to 1.1.^(-2),*** all the Chinese NDM-positive ***(n=463)available at the National Center for Biotechnology Information(NCBI),the genomic analysis revealed that blaNDM-5and Inc X3 were the predominant carbapenemase gene-plasmid combination,while a highly homogeneous relationship between NDM-positive isolates from {1.mans and animals was demonstrated at the plasmid and core genome *** the findings suggest frequent CREC transmission between {1.mans and animals,indicating that further discussions on the use of antibiotics in animals and {1.mans are needed,both in china and across the globe.
