Aberrant sperm protamination is linked to sperm dysmorphology and nuclear chromatin ***,its effects on sperm cytoplasmic maturation remain largely *** relationships of protamines,sperm morphology,DNA damage,and cytoplasmic remodeling were illustrated in this study to provide fresh perspectives on the mechanisms of male infertility.A total of 205 infertile males were allocated into 5 groups according to the percentage of spermatozoa exhibiting abnormal morphology within their *** concentration,motility,abnormal sperm morphology,cytoplasmic droplets(CDs),and excess residual cytoplasm(ERC)were analyzed according to the World Health Organization manual(2010).Sperm nuclear vacuoles(NVs)were determined by propidium iodide(PI)*** protamine expressions(P1 and P2)were detected by western *** damage was measured by acridine orange test(AOT)to calculate the proportion of sperm with single-strand DNA breaks(SSBs).Our data showed that sperm concentration and motility in infertile males significantly decreased with the severity of abnormal sperm morphology(both Pen the SSB rate and the P1/P2 ratio were also significant(Pentially serving as a valuable indicator of sperm quality and male fertility.

Lidocaine hydrochloride(LIDH) as an anesthetic is widely used in local anesthesia. Dissolving microneedles(MNs) have great application value in the field of skin anesthesia. However, the limited drug-loading of dissolving MNs is an existing challenge that affects clinical use. In this study, we have screened isomaltulose(ISO) as the proper matrix material for the MNs by using molecular dynamics(MD) simulation. Our findings indicate that ISO has good compatibility with LIDH, and the LIDH-loaded ISO MNs(LI-MNs) have high drug-loading capacity. The drug-loading capacity of LI-MNs could reach 80%, and it could effectively puncture the skin. In addition, the preparation method of customized LI-MNs was established based on three-dimensional(3D) printing technology. It was shown that the administration time of LI-MNs could be controlled within 3 min. Also, the LI-MNs were able to provide the local anesthetic efficacy within2 min and sustained for more than 2 h. Significantly, LI-MNs had more efficient drug efficacy compared to the topical creams and the majority of existing LIDH-loaded dissolving MNs. They even provided a longer duration of action than the injections. Overall, the LI-MNs with high drug-loading have a promising application prospect.

In recent years, the authors have extended the traditional interval method into the time dimension to develop a new mathematical tool called the “interval process model” for quantifying time-varying or dynamic uncertainties. This model employs upper and lower bounds instead of precise probability distributions to quantify uncertainty in a parameter at any given time point. It is anticipated to complement the conventional stochastic process model in the coming years owing to its relatively low dependence on experimental samples and ease of understanding for engineers. Building on our previous work, this paper proposes a spectrum analysis method to describe the frequency domain characteristics of an interval process, further strengthening the theoretical foundation of the interval process model and enhancing its applicability for complex engineering problems. In this approach, we first define the zero midpoint function interval process and its auto/cross-power spectral density(PSD) functions. We also deduce the relationship between the auto-PSD function and the auto-covariance function of the stationary zero midpoint function interval process. Next, the auto/cross-PSD function matrices of a general interval process are defined, followed by the introduction of the concepts of PSD function matrix and cross-PSD function matrix for interval process vectors. The spectrum analysis method is then applied to random vibration problems, leading to the creation of a spectrum-analysis-based interval vibration analysis method that determines the PSD function for the system displacement response under stationary interval process excitations. Finally, the effectiveness of the formulated spectrum-analysis-based interval vibration analysis approach is verified through two numerical examples.
