Kerogen types exert a decisive effect on the onset and capacity of hydrocarbon generation of source rocks.Lacustrine source rocks in the Liaohe Western Depression are characterized by thick deposition,high total organic carbon(TOC)content,various kerogen types,and a wide range of thermal maturity.Consequently,their hydrocarbon generation potential and resource estimation can be misinterpreted.In this study,geochemical tests,numerical analysis,hydrocarbon generation kinetics,and basin modeling were integrated to investigate the differential effects of kerogen types on the hydrocarbon generation potential of lacustrine source rocks.Optimized hydrocarbon generation and expulsion(HGE)models of different kerogen types were established quantitatively upon abundant Rock-Eval/TOC/vitrinite reflectance(R_(o))datasets.Three sets of good-excellent source rocks deposited in the fourth(Es4),third(Es3),and first(Es1)members of Paleogene shahejie Formation,are predominantly types I-II_(1),II_(1)-II_(2),and II-III,respectively.The activation energy of types I-II_(2)kerogen is concentrated(180-230 kcal/mol),whereas that of type III kerogen is widely distributed(150-280 kcal/mol).The original hydrocarbon generation potentials of types I,II_(1),II_(2),and III kerogens are 790,510,270,and 85 mg/g TOC,respectively.The Ro values of the hydrocarbon generation threshold for type I-III source rocks gradually increase from 0.42%to 0.74%,and Ro values of the hydrocarbon expulsion threshold increase from 0.49%to 0.87%.Types I and II_(1)source rocks are characterized by earlier hydrocarbon generation,more rapid hydrocarbon expulsion,and narrower hydrocarbon generation windows than types II_(2)and III source rocks.The kerogen types also affect the HGE history and resource potential.Three types(conventional,tight,and shale oil/gas)and three levels(realistic,expected,and prospective)of hydrocarbon resources of different members in the Liaohe Western Depression are evaluated.Findings suggest that the Es3 member has considerable conventional and

BACKGROUND The overlap of imaging manifestations among distinct splenic lesions gives rise to a diagnostic dilemma.Consequently,a definitive diagnosis primarily relies on his-tological results.The ultrasound(US)-guided coaxial core needle biopsy(CNB)not only procures sufficient tissue to help clarify the diagnosis,but reduces the incidence of puncture-related complications.CASE SUMMARY A 41-year-old female,with a history of pulmonary tuberculosis,was admitted to our hospital with multiple indeterminate splenic lesions.Gray-scale ultrasono-graphy demonstrated splenomegaly with numerous well-defined hypoechoic ma-sses.Abdominal contrast-enhanced computed tomography(CT)showed an en-larged spleen with multiple irregular-shaped,peripherally enhancing,hypodense lesions.Positron emission CT revealed numerous abnormal hyperglycemia foci.These imaging findings strongly indicated the possibility of infectious disease as the primary concern,with neoplastic lesions requiring exclusion.To obtain the precise pathological diagnosis,the US-guided coaxial CNB of the spleen was ca-rried out.The patient did not express any discomfort during the procedure.CONCLUSION Percutaneous US-guided coaxial CNB is an excellent and safe option for obtaining precise splenic tissue samples,as it significantly enhances sample yield for exact pathological analysis with minimum trauma to the spleen parenchyma and sur-rounding tissue.

In this paper,the interrupted and ruptured creep tests were carried out in a novel second generation single crystal superalloy named DD11 at 1100℃/130 MPa.The alloy exhibited typical creep curve including primary,steady,and tertiary three creep stages.The microstructural evolution at different stages of the creep were analyzed by scanning electron microscopy(SEM)and transmission electron microscopy(TEM).The results show that theγ′phases transform into rafted structure at the early stage of the steady creep and keep stable during the steady creep stage.As the creep goes on,the rafted structure further coarsens and the topological inversion occurs.In addition,at the primary creep,the dislocations mainly move in theγmatrix and pile up in theγ/γ′interface since the matrix channels widen slightly.The formation of the regular interfacial dislocation networks occurs at the early stage of the steady creep.Under the low stress,the dominated deformation mechanism during steady creep stage is the climbing of the〈010〉type edge dislocation.Furthermore,the effect of the deformation mechanism on creep property was discussed in detail.

It has been demonstrated that microplastics (MPs) can accumulate heavy metals from the environment and transfer them into organisms via the food chain. However, adsorption and desorption capacities for biodegradable MPs relative to those for conventional MPs remain poorly understood. In this study, cadmium (Cd(II)) adsorption and desorption characteristics of polylactic acid (PLA), a typical biodegradable MP, were investigated. Two conventional MPs, i.e., polypropylene (PP) and polyamide (PA) were used for comparison. The maximum Cd(II) adsorption capacities of the MPs studied in the adsorption experiments decreased in the order PA (0.96 ± 0.07 mg/g) > PLA (0.64 ± 0.04 mg/g) > PP (0.22 ± 0.03 mg/g). The Pseudo-second-order kinetic model and Freundlich isothermal model described the Cd(II) adsorption behaviors of PLA MPs well. X-ray photoelectron spectroscopy and two-dimensional Fourier transform infrared correlation spectroscopy analysis indicated that oxygen functional groups were the major and preferential binding sites of PLA MPs, which contributed to their high Cd(II) adsorption capacities. Simulated gastric and intestinal fluids both significantly enhanced the desorption capacities of the examined MPs. Notably, degradation of the PLA MPs during in vitro human digestion made the Cd(II) on the PLA MPs more bioaccessible (19% in the gastric phase and 62% in the intestinal phase) than Cd(II) on the PP and PA MPs. These results indicate the remarkable capacities of biodegradable MPs to accumulate Cd(II) and transfer it to the digestive system and show that biodegradable MPs might pose more severe threats to human health than conventional nonbiodegradable MPs.

Comprehensive reutilization of herbal waste:Coproduction of magnolol,honokiol,and β-amyrin from Magnolia officinalis residue

Herbal extraction residues(HERs)cause serious environmental pollution and resource waste.In this study,a novel green route was designed for the comprehensive reutilization of all components in HERs,taking Magnolia officinalis residues(MOR)as an example.The reluctant structure of MOR was first destroyed by alkali pretreatment to release the functional ingredients(magnolol and honokiol)originally remaining in MOR and to make MOR more accessible for hydrolysis.A metal–organic frame material MIL-101(Cr)with a maximum absorption capacity of 255.64 mg g^(-1)was synthesized to absorb the released honokiol and magnolol from the pretreated MOR solutions,and 40 g L^(-1)reducing sugars were obtained with 81.8%enzymatic hydrolysis rate at 10%MOR solid loading.Finally,382 mg L-1β-amyrin was produced from MOR hydrolysates by an engineered yeast strain.In total,1 kg honokiol,8 kg magnolol,and 7.64 kg β-amyrin could produce from 1 ton MOR by this cleaner process with a total economic output of 170,700 RMB.

The endoplasmic reticulum(ER),which is composed of a continuous network of tubules and sheets,forms the most widely distributed membrane system in eukaryotic cells.As a result,it engages a variety of organelles by establishing membrane contact sites(MCSs).These contacts regulate organelle positioning and remodeling,including fusion and fission,facilitate precise lipid exchange,and couple vital signaling events.Here,we systematically review recent advances and converging themes on ER-involved organellar contact.The molecular basis,cellular influence,and potential physiological functions for ER/nuclear envelope contacts with mitochondria,Golgi,endosomes,lysosomes,lipid droplets,autophagosomes,and plasma membrane are summarized.

Strategies to improve T cell therapy efficacy in solid tumors such as hepatocellular carcinoma(HCC)are urgently needed.The common cytokine receptorγchain(γc)family cytokines such as IL-2,IL-7,IL-15 and IL-21 play fundamental roles in T cell development,differentiation and effector phases.This study aims to determine the combination effects of IL-21 in T cell therapy against HCC and investigate optimized strategies to utilize the effect of IL-21 signal in T cell therapy.The antitumor function of AFP-specific T cell receptor-engineered T cells(TCR-T)was augmented by exogenous IL-21 in vitro and in vivo.IL-21 enhanced proliferation capacity,promoted memory differentiation,downregulated PD-1 expression and alleviated apoptosis in TCR-T after activation.A novel engineered IL-21 receptor was established,and TCR-T armed with the novel engineered IL-21 receptors(IL-21R-TCR-T)showed upregulated phosphorylated STAT3 expression without exogenous IL-21 ligand.IL-21R-TCR-T showed better proliferation upon activation and superior antitumor function in vitro and in vivo.IL-21R-TCR-T exhibited a less differentiated,exhausted and apoptotic phenotype than conventional TCR-T upon repetitive tumor antigen stimulation.The novel IL-21 receptor in our study programs powerful TCR-T and can avoid side effects induced by IL-21 systemic utilization.The novel IL-21 receptor creates new opportunities for next-generation TCR-T against HCC.

Background:Accumulating evidence suggests that metabolic disorders,including type 2 diabetes mellitus(T2DM),can be treated with traditional Chinese medicine formulas,such as the Gegen Qinlian decoction(GQD).This study elucidates the mechanisms by which gut microbes mediate the anti-diabetic effects of GQD.Methods:We conducted a double-blind randomized clinical trial involving 120 untreated participants with T2DM.During the 12-week intervention,anthropometric measurements and diabetic traits were recorded every 4 weeks.Fecal microbiota and serum metabolites were measured before and after the intervention using 16S rDNA sequencing,liquid chromatography-mass spectrometry,and Bio-Plex panels.Results:Anti-diabetic effects were observed in the GQD group in the human trial.Specifically,glycated hemoglobin,fasting plasma glucose,and two-hour postprandial blood glucose levels were significantly lower in the GQD group than in the placebo group.Additionally,Faecalibacterium was significantly enriched in the GQD group,and the short-chain fatty acid levels were higher and the serum inflammation-associated marker levels were lower in the GQD group compared to the placebo group.Moreover,Faecalibacterium abundance negatively correlated with the levels of serum hemoglobin,fasting plasma glucose,and pro-inflammatory cytokines.Finally,the diabetes-alleviating effect of Faecalibacterium was confirmed by oral administration of Faecalibacterium prausnitzi(DSMZ 17677)in T2DMmousemodel.Conclusions:GQD improved type 2 diabetes primarily by modulating the abundance of Faecalibacterium in the gut microbiota,alleviating metabolic disorders and the inflammatory state.

Alzheimer's disease （AD） has become an important public health issue worldwide. P-Coumaric acid （CA） and lactoferrin （Lf） possessed antioxidant， anti-inflammatory， and anti-AD activities. Herein， we hypothesized that a combination treatment of CA and Lf would synergistically improve AD symptoms and studied its mechanisms. Twelve-week-old APP/PS1 mice were treated with CA， Lf， or both CA and Lf for 8 weeks. Results showed that individual and combined treatments could ameliorate cognitive deficits to varying degrees， with stronger effects for combined intervention than for CA or Lf alone. Specifically， combined treatment was most effective in improving nesting ability and reducing Aβ1-42deposition in mice. Moreover， combined treatment was more effective in suppressing APP， BACE1， and inflammation; it inhibited the CCAAT-enhancer-binding protein （C/EBPβ）/asparagine endopeptidase （AEP） signaling pathway in the hippocampus and colon and upregulated the expression of tight junction protein ZO-1 in the colon; it also decreased the F/B ratio at the phylum level and increased the relative abundance of Prevotellaceae， Lachnospira， and Eubacterium at the genus level. Overall， the combination of CA and Lf may ameliorate cognitive deficits in APP/PS1 mice by improving inflammation and inhibiting the C/EBPβ/AEP signaling pathway via modulating gut microbiome.
