A functional tacrolimus-releasing nerve wrap for enhancing nerve regeneration following surgical nerve repair

Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

Dobutamine Infusion and Absence of Pulmonary Hypertension Are Associated with Decreased Mortality in a Cohort of 249 Patients with Cardiogenic Shock

Objective: Prognostic analysis of 249 patients admitted for cardiogenic shock (CS) of various origins. Background: Little is known about prognosis of CS from non-ischemic cardiomyopathy. Methods: Retrospective monocentric study of patients referred to an ICU during 2 years. Results: Despite aggressive management including intra-aortic balloon pump (31%), extra-renal replacement therapy (36%), extra-corporeal life support (8%), and catecholamine infusion (97%), in-hospital mortality was 46%. Toxic CS or CS related to deficiency carried a better outcome (mortality 5%). Post-myocardial infarction or post-cardiac arrest CS was associated with higher mortality. In the multivariate analyses, only SAPS II (OR 1.037;1.013 - 1.056;p = 0.0001), pulmonary hypertension (OR 4.8;1.3 - 17;p = 0.02), extra-renal replacement therapy (OR 2.9;1.3 - 6;p = 0.006), and dobutamine infusion (OR 0.44;0.2 - 0.96;p = 0.04) were significantly associated with in-hospital mortality. Conclusion: Dobutamine infusion was associated with a better outcome. Higher SAPS II, pulmonary hypertension, and extra-renal replacement therapy were associated with increased in-hospital mortality.

Spin-correlation function of the fully frustrated Ising model and ±j Ising spin glass on a square lattice

In this work we study the correlation function of the ground state of a two-dimensional fully frustrated Ising model as well as spin glass. The Pfaffian method is used to calculate free energy and entropy as well as the correlation function. We estimate the exponent of spin correlation function for the fully frustrated model and spin glass. In this paper an overview of the latest results on the spin correlation function is presented.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

The nature of the companion of PSR j1719-1438: a white dwarf or an exotic object?

We raise the possibility that the very dense, compact companion of PSR j1719-1438, which has a jupiter-like mass, is an exotic quark object rather than a light helium or carbon white dwarf. The exotic hypothesis naturally explains some of the observed features, and provides quite strong predictions for this system, to be confirmed or refuted in feasible future studies.

It is crucial to determine the spin and parity （jR） of Zc（3900）＋ for understanding its structure. We perform a helicity amplitude analysis on Zc（3900）＋ in the process e＋e- →π＋π-j/φ＊ with the hypotheses jP = 0-, 1＋, 1-, 2＋ and 2-. To estimate the significance of jP = 1＋ over other hypotheses, we perform a Monte Carlo simulation study, and we also discuss the prospect of measuring the spin parity in the future experiment with a large data sample.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.

We discuss the association between the candidate magnetar CXOU j171405.7-381031 and the supernova remnant CTB *** recent detection of the period derivative of the object allowed an estimation of a young characteristic age of only ～ *** value is too small to be compatible even with the minimum radius of the remnant being ≥ 10 pc,the value corresponding to the lower limit of the estimated distance of 10.2 ± 3.5 kpc,unless the true distance happens to be even smaller than the lower *** argue that a consistent scenario for the remnant＇s origin,in which the latter is powered by the energy injected by a young magnetar,is indeed more accurate to explain the young age,and demonstrates its non-standard（***-driven） nature.

Safe mining practices under wide spans in underground non-caving mines——Case studies

Mining under wide span is of concern as it increases the probability of back caving causing personnel injury and equipment damage in underground mines in open stoping and underhand drift-and-fill *** restricting personnel access to well supported lateral development is commonly practiced,it is not always possible to implement this requirement due to various factors such as ore loss control,drilling equipment limitations,availability of remote operating capacity and consideration of *** with rules implemented to limit personnel entry into openings with wide spans,the hazards of equipment damage and back caving still *** the years,different practices have been reported and adopted to minimize risks associated with exposure to large spans in various underground *** from these practices are beneficial to current and future mines with challenges of safe extraction of thick deposits in a non-caving *** paper briefly summarizes practices in mining wide orebodies using the open stoping method without personnel access and underhand mining using the drift-andfill method with personnel exposure in the industry and presents cases from Kinross mines where the hanging pillar design was tested,and stope backs were naturally and artificially supported for extraction under wide spans using the open stoping method.
