Effects of undigested protein-rich ingredients on polarised small intestinal organoid monolayers

Here, we describe the use of monolayers of intestinal epithelial cells derived from intestinal organoids and transcriptomics to investigate the direct effects of dietary protein sources on epithelial function. Mechanically dissociated 3 D organoids of mouse duodenum were used to generate a polarized epithelium containing all cell types found in the tissue of origin. The organoid-derived cell monolayers were exposed to 4%(w/v) of ‘undigested(non-hydrolysed)-soluble' fraction of protein sources used as feed ingredients [soybean meal(SBM) and casein], or alternative protein sources(spray dried plasma protein, and yellow meal worm), or controls for 6 h prior to RNA isolation and transcriptomics. All protein sources altered expression of unique biological processes in the epithelial cells. Exposure of intestinal organoids to SBM downregulated expression of retinol and retinoid metabolic processes as well as cholesterol and lipid biosynthetic pathways, consistent with the reported hypotriglyceridaemic effect of soy protein in vivo. These findings support the use of intestinal organoids as models to evaluate complex interactions between dietary ingredients and the intestinal epithelium and highlights some unique host effects of alternative protein sources in animal feed and potentially human food.

Aberrant activation of the epidermal growth factor receptor(EGFR)is a driving force for cancer growth in a subgroup of non-small cell lung cancer *** patients can be identified by the presence of activating EGFR *** three generations of EGFR-tyrosine kinase inhibitors(TKIs)have been approved by the Food and Drug Administration and European Medicine *** paper reviews the structure of EGFR and the downstream signaling pathways of EGFR and describes the mechanisms of intrinsic and acquired resistance against *** mechanisms include secondary or tertiary mutations in EGFR,the activation of bypassing signaling pathways or a histological transformation to small cell lung ***,drug efflux transporters will affect the cellular accumulation of EGFR-TKIs and penetration of the first generation of EGFR-TKI into the *** sequestration of some EGFR-TKIs may also prevent the drugs to reach their *** conclusion,resistance to EGFR-TKIs is multifactorial,including primary and acquired mutations in the EGFR gene,activation of bypassing pathways and limited uptake of drugs in the cells or target *** pharmacological studies are needed in order to develop new specific compounds targeted to overcome new resistance mechanisms in order to enable a personalized treatment approach.

The association between radiological spreading pattern and clinical outcomes in necrotizing external otitis

Objectives: Necrotizing external otitis(NEO) is a rare infectious disease of the skull base. The purpose of this study was to determine whether clinical outcomes of NEO can be correlated to different infectious spread ***: Retrospective chart review from 2010 to 2019 with NEO patients, who were divided into two cohorts: single spreading patterns(group A) or complex spreading patterns(group B) as diagnosed by *** symptoms, diagnostic and treatment delay, course of disease, complications, and duration of antibiotic exposure were retrospectively collected from patient ***: 41 NEO patients were included, of which 27 patients belonged to group A(66%). The diseaserelated mortality rate was 12.2% among the entire cohort, no differences were found between group A and B. Higher rates of ***(42.9% vs 14.8% P = 0.047) and N. IX palsies were found in group B compared to group A(28.6% vs 3.7%, P = 0.039). The median duration of antibiotic use was significantly different for a complex spreading pattern, clinical recovery and hospitalizations. Complications were associated with higher diagnostic delay and with a complex spread pattern. The median duration of follow-up was 12.0(IQR 6.0-19.5) ***: NEO is a severe disease, with significant mortality and morbidity(cranial nerve palsies). The radiological spread pattern may assist in predicting clinical outcome. Furthermore, complex spread patterns are associated with higher rates of clinical nerve palsies(N. VII and ***), complications, surgery rates and longer duration of antibiotic use. Diagnostic delay was associated with mortality, complications and facial palsies.

Four different fruit thinning severities were *** thinning treatments were carried out in October before the occurrence of physiological fruit *** parameters were measured(both qualitative and quantitative)and the results compared among treatments as well as to that of the *** results showed that where fruit on‘Sensation’mango pani-cles were thinned to two fruit per panicle,a significant difference was obtained for most of the quantitative *** fruits of the treatment where one fruit per panicle was retained and50%panicles removed,produced the best results for most of the qualitative parameters.

In this study,Ni catalysts supported on Pr-doped Ce O_(2) are studied for the CO_(2) methanation reaction and the effect of Pr doping on the physicochemical properties and the catalytic performance is thoroughly *** is shown,that Pr^(3+)ions can substitute Ce^(4+)ones in the support lattice,thereby introducing a high population of oxygen vacancies,which act as active sites for CO_(2) *** doping can also act to reduce the crystallite size of metallic Ni,thus promoting the active metal *** performance evaluation evidences the promoting effect of low Pr loadings(5 at%and 10 at%)towards a higher catalytic activity and lower CO_(2) activation *** the other hand,higher Pr contents negate the positive effects on the catalytic activity by decreasing the oxygen vacancy population,thereby creating a volcano-type trend towards an optimum amount of aliovalent substitution.

Pyrrololactam type of alkaloids was found in many marine sponges which belong to the family Axinellidae. In the course of our systematic research for bioactive substances in marine organisms, we studied the sponge Phacellia fusca Schmidt which also belongs to the family Axinelldae. From the chloroform-soluble fraction, ten A-nor-sterols were isolated and identified, while from the water-soluble portion, three known pyrrololactam alkaloids and a new pyrrololactam alkaloid, which was named fuscin, were obtained. The present paper contains the isolation and structure determination of fuscin (1).
