Understanding brain structure and function,and the complex relationships bet ween them,is one of the grand challenges of contemporary *** to their fexiblity,optical techniques could be the key to explore this complex *** this manuscript,we briefly review recent adv ancements in optical methods applied to three main issues:anatomy,plasticity and *** describe novel implement ations of light-sheet microscopy to resolve neuronal anat-omy in whole fixed brains with cellular *** to liv ing samples,we show how real-time dynamics of brain rewiring can be visualized through two-photon microscopy with the spatial resolution of single synaptic *** plasticity of the injured brain can also be dissected through cut ting edge optical methods that specifically ablate single neuronal ***,we report how nonlinear microscopy in combination with novel voltage sensitive dyes allow optical registrations of action potential across a population of neurons opening promising prospective in understanding brain *** knowledge acquired from these complementary optical methods may provide a deeper comprehension of the brain and of its unique features.

Impact of chronic exposure to bevacizumab on EpCAM-based detection of circulating tumor cells

Background： Circulating tumor cells （CTCs） are often undetected through the immunomagnetic epithelial cell adhesion molecule （EpCAM）-based CellSearch~ System in breast and colorectal cancer （CRC） patients treated with bevacizumab （BEV）, where low CTC numbers have been reported even in patients with evidence of progression of disease. To date, the reasons for this discrepancy have not been clarified. This study was carried out to investigate the molecular and phenotypic changes in CRC cells after chronic exposure to BEV in vitro. Methods： The human CRC cell line WiDr was exposed to a clinically relevant dose of BEV for 3 months in vitro. The expression of epithelial and mesenchymal markers and EpCAM isoforms was determined by western blotting and immunofluorescence. To evaluate the impact of EpCAM variant isoforms expression on CTC enumeration by CellSearch, untreated and treated colon cancer cells were spiked into 7.5 mL of blood from a healthy donor and enumerated by CellSearch. Results： Chronic exposure of CRC cell line to BEV induced decreased expression of EpCAM 40 kDa isoform and increased expression EpCAM 42 kDa isoform, together with a decreased expression of cytokeratins （CK）, while no evidence of epithelial to mesenchymal transition （EMT） in treated cells was observed. The recovery rate of cells through CellSearch was gradually reduced in course of treatment with BEV, being 84% , 70% and 40% at l, 2 and 3 months, respectively. Conclusions： We hypothesize that BEV may prevent CellSearch from capturing CTCs through altering EpCAM isoforms.

Extracellular polymeric substances as paper coating biomaterials derived from anaerobic granular sludge

Recovering extracellular polymeric substances(EPS)from waste granular sludge offers a cost-effective and sustainable approach for transforming wastewater resources into industrially valuable ***,the application potential of these EPS in real-world scenarios,particularly in paper manufacturing,remains *** we show the feasibility of EPS-based biomaterials,derived from anaerobic granular sludges,as novel coating agents in paper *** systematically characterised the rheological properties of various EPS-based *** applied as surface sizing agents,these EPS-based biomaterials formed a distinct,ultra-thin layer on paper,as evidenced by scanning electron microscopy.A comprehensive evaluation of water and oil penetration,along with barrier properties,revealed that EPS-enhanced coatings markedly diminished water absorption while significantly bolstering oil and grease *** performance was observed in EPS variants with elevated protein and hydrophobic contents,correlating with their superior rheological *** enhanced water-barrier and grease resistance of EPS-coated paper can be attributed to its non-porous,fine surface structure and the functional groups in EPS,particularly the high protein content and hydrophobic humic-like *** research marks the first demonstration of utilizing EPS from anaerobic granular sludge as paper-coating biomaterials,bridging a critical knowledge gap in the sustainable use of biopolymers in industrial applications.
