Novel polysaccharide-protein conjugates provide an immunogenic 13-valent pneumococcal conjugate vaccine for S.pneumoniae

Pneumonia remains the single leading cause of childhood death worldwide.Despite the commercial availability of multiple pneumococcal conjugate vaccines(PCVs),high dosage cost and supply shortages prevent PCV delivery to much of the developing world.The current work presents high-yield pneumococcal conjugates that are immunogenic in animals and suitable for use in human vaccine development.The 13-valent pneumococcal conjugate vaccine(PCV-13)investigated in this research incorporated serotypes 1,3,4,5,6A,6B,7F,9V,14,18C,19A,19F,and 23F.Pneumococcal polysaccharides(PnPSs)and CRM197 carrier protein were produced and purified in-house,and used to prepare PnPS-CRM conjugates using unique,cyanide-free,in vacuo glycation conjugation methods.In vitro characterization confirmed the generation of higher molecular weight PnPS-CRM conjugates low in free protein.In vivo animal studies were performed to compare PnuVax's PCV-13 to the commercially available PCV-13,Prevnar®13(Pfizer,USA).A boost dose was provided to all groups post-dose 1 at t?14 days.Post-dose 2 results at t?28 days showed that all 13 serotypes in PnuVax's PCV-13 were boostable.Per serotype IgG GMCs demonstrated that PnuVax's PCV-13 is immunogenic for all 13 serotypes,with 10 of the 13 serotypes statistically the same or higher than Prevnar®13 post-dose 2.As a result,the novel polysaccharideprotein conjugates developed in this work are highly promising for use in human PCV development.The in vacuo conjugation technique applied in this work could also be readily adapted to develop many other conjugate vaccines.

Accelerated flow softening and dynamic transformation of Ti-6Al-4V alloy in two-phase region during hot deformation via coarsening αgrain

The flow softening is an important phenomenon during hot deformation of metallic materials.In the present work,a more evident flow softening of Ti-6 Al-4 V alloy when deformed in two-phase region was observed in coarser a grain sample,which was attributed to an accelerated dynamic transformation from harder α phase into β phases.Notably,full β microstructure was observed in coarse grain samples at strain of 1.2,while retained a phase was observed in fine a grain specimens.In the views of thermodynamics and crystallographic analysis,the in-depth mechanisms of dynamic transformation were further investigated.

Exact Static Plane Symmetric Soliton-Like Solutions to the Nonlinear Interacting Electromagnetic and Scalar Field Equations in General Relativity

This research work is related to soliton solutions considered as models that can describe the complex configuration of elementary particles from the study of the interactions of their fields. It is interested in the interaction of fields between two different elementary particles by expressing their physical properties. For that, we have obtained, exact static plane symmetric soliton-like solutions to the nonlinear equations of interacting electromagnetic and scalar fields taking into account the own gravitational field of elementary particles using the calibrated invariance function P(I). It has been proved that all solutions of the Einstein, nonlinear electromagnetic and scalar field equations are regular with the localized energy density. Moreover, the total charge of particles is finite and the total energy of the interaction fields is bounded. It have been emphasized the importance to the own gravitational field of elementary particles and the role of the nonlinearity of fields in the determination of these solutions. In flat space-time, soliton-like solutions exist but the total energy of the interaction fields is equal to zero. We have also shown that in the linear case, soliton-like solutions are absent.

Resection of complex pancreatic injuries: Benchmarking postoperative complications using the Accordion classification

AIM To benchmark severity of complications using the Accordion Severity Grading System(ASGS) in patients undergoing operation for severe pancreatic injuries. METHODS A prospective institutional database of 461 patients with pancreatic injuries treated from 1990 to 2015 was reviewed. One hundred and thirty patients with AAST grade 3, 4 or 5 pancreatic injuries underwent resection(pancreatoduodenectomy, n = 20, distal pancreatectomy, n = 110), including 30 who had an initial damage controllaparotomy(DCL) and later definitive surgery. AAST injury grades, type of pancreatic resection, need for DCL and incidence and ASGS severity of complications were assessed. Uni-and multivariate logistic regression analysis was applied. RESULTS Overall 238 complications occurred in 95(73%) patients of which 73% were ASGS grades 3-6. Nineteen patients(14.6%) died. Patients more likely to have complications after pancreatic resection were older, had a revised trauma score(RTS) juries of the head and neck of the pancreas with associated vascular and duodenal injuries, required a DCL, received a larger blood transfusion, had a pancreatoduodenectomy(PD) and repeat laparotomies. Applying univariate logistic regression analysis, mechanism of injury, RTS < 7.8, shock on admission, DCL, increasing AAST grade and type of pancreatic resection were significant variables for complications. Multivariate logistic regression analysis however showed that only age and type of pancreatic resection(PD) were significant. CONCLUSION This ASGS-based study benchmarked postoperative morbidity after pancreatic resection for trauma. The detailed outcome analysis provided may serve as a reference for future institutional comparisons.

Soliton-Like Spherical Symmetric Solutions to the Electromagnetic and Scalar Nonlinear Induction Field Equations in the General Relativity Theory

In this paper, we have used the static spherical symmetric metric. The parameter of the nonlinearity fields is included in the arbitrary function characterizing the interaction between the electromagnetic and scalar fields. Taking into account the own gravitational field of elementary particles, we have obtained exact static spherical symmetric solutions to the electromagnetic and scalar field equations of nonlinear induction. Considering all forms of the solution of Liouville equation, we proved that the metric functions are regular with localized energy density. Moreover, the total energy of the nonlinear induction fields is bounded and the total charge of the elementary particles has a finite value (soliton-like). In the flat space-time, soliton-like solutions exist.

Organizational commitment and turnover intention among rural nurses in the Philippines: Implications for nursing management

Objectives:The unrelenting migration trend of Filipino nurses to other countries has threatened the quality of patient care services in the country.This study explored the extent of nurses'organizational commitment and turnover intention in the Philippines.Furthermore,predictors of nurses'organizational commitment and turnover intention were identified.Methods:A cross-sectional research design was adopted for this study.Two hundred nurses from nine rural hospitals in the Central Philippines were asked to participate in the study and 166 nurses responded(an 83%response rate).Two standardized instruments were used:the Organizational Commitment Questionnaire and the Six-item Turnover Intention Inventory Scale.Results:Findings revealed that Philippine nurses were moderately committed(3.13±0.24)to and were undecided(2.42±0.67)whether or not to leave their organization.Nurses'age(P=0.006),gender,(t=-2.25,P=0.026),education(t=2.38,P<0.001),rank(t=4.38,P<0.001),and work experience(t=2.18,P=0.031)correlated significantly with organizational commitment,while nurses'age(P=0.028)and education(t=1.99,P=0.048)correlated significantly with turnover intention.An inverse relationship was identified between the organizational commitment and turnover intention(r=-0.22,P=0.005).Conclusion:The findings of this study highlight the need for formulation and implementation of interventions to promote life-long commitment in nurses and to reduce turnover rates.

Control of light emission of quantum emitters coupled to silicon nanoantenna using cylindrical vector beams

Light emission of europium(Eu^(3+))ions placed in the vicinity of optically resonant nanoantennas is usually controlled by tailoring the local density of photon states(LDOS).We show that the polarization and shape of the excitation beam can also be used to manipulate light emission,as azimuthally or radially polarized cylindrical vector beam offers to spatially shape the electric and magnetic fields,in addition to the effect of silicon nanorings(Si-NRs)used as nanoantennas.The photoluminescence(PL)mappings of the Eu^(3+)transitions and the Si phonon mappings are strongly dependent of both the excitation beam and the Si-NR dimensions.The experimental results of Raman scattering and photoluminescence are confirmed by numerical simulations of the near-field intensity in the Si nanoantenna and in the Eu^(3+)-doped film,respectively.The branching ratios obtained from the experimental PL maps also reveal a redistribution of the electric and magnetic emission channels.Our results show that it could be possible to spatially control both electric and magnetic dipolar emission of Eu^(3+)ions by switching the laser beam polarization,hence the near field at the excitation wavelength,and the electric and magnetic LDOS at the emission wavelength.This paves the way for optimized geometries taking advantage of both excitation and emission processes.

Trends in Biodiversity Research — A Bibliometric Assessment

Research on biodiversity has grown considerably during the last decades. The present study applies bibliometric methods to evaluate efforts in this field of study. We retrieved roughly 69,000 bibliographic records from the Web of Science database that matched the word biodiversity (and derivatives) in keywords, title or abstract. Article contributions and number of involved authors and journals increased exceptionally fast since the 1980s, when the term biodiversity was coined. But since the year 2008, a decelerated growth rate leads to an average rate of knowledge generation. Using the frequency of terms extracted from publication titles, we inferred that the community-level focus has increased in biodiversity studies, while molecular biodiversity is still not strongly represented. Climate-related topics are rapidly gaining importance in biodiversity research. The geographical imbalance between allocation of research efforts and distribution of biological diversity is apparent.

Flexible Defense Succeeds Creative Attacks!—A Simulation Approach Based on Position Data in Professional Football

Introduction: The key to success is finding the perfect mixture of tactical patterns and sudden breaks of them, which depends on the behavior of the opponent team and is not easy to estimate by just watching matches. According to the specific tactical team behavior of “attack vs. defense” professional football matches are investigated based on a simulation approach, professional football matches are investigated according to the specific tactical team behavior of “attack vs. defense.” Methods: The formation patterns of all the sample games are categorized by SOCCER© for defense and attack. Monte Carlo-Simulation can evaluate the mathematical, optimal strategy. The interaction simulation between attack and defense shows optimal flexibility rates for both tactical groups. Approach: A simulation approach based on 40 position data sets of the 2014/15 German Bundesliga has been conducted to analyze and optimize such strategic team behavior in professional soccer. Results: The results revealed that both attack and defense have optimal planning rates to be more successful. The more complex the success indicator, the more successful attacking player groups get. The results also show that defensive player groups always succeed in attacking groups below a specific planning rate value. Conclusion: Groups are always succeeding. The simulation-based position data analysis shows successful strategic behavior patterns for attack and defense. Attacking player groups need very high flexibility to be successful (stay in ball possession). In contrast, defensive player groups only need to be below a defined flexibility rate to be guaranteed more success.
