The impact of weather on summer and winter exercise behaviors

background: Outdoor exercise is an enjoyable way for individuals to improve fitness, but it is dependent on weather conditions. This study examines the association between weather conditions and outdoor exercise after adjustment for age, sex, race, and socioeconomic status.Methods: We used data representative of American adults from the University of Michigan/Thomson Reuters june 2013 surveys of consumers(core and supplement) to investigate self-reported exercise behavior in summer and winter. Multivariate multinomial logistic regression models estimated the odds of delayed or indoor exercise compared with outdoor exercise.Results: Of the 502 respondents, 16.3% did not regularly exercise outdoors(i.e., at least once a week), and many would delay exercise both in summer(51.8%) and winter(43.9%). Individuals listing rain as the predominant adverse weather condition had 3.33 times higher odds of exercising indoors(95% confidence interval(CI): 1.34-8.28) and 3.49 times higher odds of delaying exercise(95%CI: 1.69-7.21) compared with those mentioning heat as the predominant adverse condition. Individuals for whom ice or snow was an adverse winter weather condition were more likely to delay exercise(odds ratio(OR) = 3.34; 95%CI: 1.19-9.36), compared with those concerned with cold.Conclusion: This study found that race, age, and education exacerbate the negative effects of adverse weather conditions on the decision to exercise outdoors. Accordingly, any recommendation for an individual to exercise outdoors should be combined with an evaluation of the individual's outdoor environment along with strategies for the individual to continue exercising, indoors or outdoors, when adverse weather is present.

The Sulfate Transporter Family in Wheat： Tissue-Specific Gene Expression in Relation to Nutrition

Sulfate uptake and distribution in plants are managed by the differential expression of a family of transporters, developmentally, spatially, and in response to sulfur nutrition. Elucidation of the signaling pathways involved requires a knowledge of the component parts and their interactions. Here, the expression patterns of the full complement of sulfate transporters in wheat, as influenced by development and sulfur nutrition, are described. The 10 wheat sulfate transporters characterized here are compared to the gene families for both rice and Brachypodium, for whom full genome information is available. Expression is reported in young seedlings with a focus on roles in uptake from nutrient solution and differential expression in relation to sulfate deprivation. In addition, patterns of expression in all organs at the grain filling stage are reported and indicate differential responses to nutritional signals of the individual transporters in specific tissues and an overall coordination of uptake, storage, and remobilization to deliver sulfur to the developing grain.

Heavy Metals in a Sulfidic Minespoil:Fractions and Column Leaching

Fractions of various heavy metals in a sulfidic minespoil were investigated. Column leaching experimentwas also conducted to simulate 'acid mine drainage' (AMD) from the minespoil. The results show thatleaching of heavy metals from the minespoil was extremely significant during the initial water flushing.The amounts of heavy metals leached out dramatically reduced after leaching twice. It is worthwhile tonote that in this study, Zn, Mn, Fe, As and Ni in the first leachate exceeded the total amount of eachcorresponding water-extractable (1:5, soil:water) metal contained in the minespoil sample. This appears tosuggest that 1:5 water extraction did not allow accurate estimation of water-leachable concentrations of theabove heavy metals. This work has implications for the management of sulfidic minespoils. Acid drainageof great environmental concerns is likely to occur only during heavy rainfall events after substantial solubleand readily exchangeable acid and metals are accumulated in the minespoils. The slow-reacting fractionsother than water-soluble and readily exchangeable fractions may pose little environmental hazards. This isparticularly true for Pb, As and Ni.

Cytoprotective Effect of Silymarin against Diabetes-Induced Cardiomyocyte Apoptosis in Diabetic Rats

Objective The beneficial effects of silymarin have been extensively studied in the context of inflammation and cancer treatment, yet much less is known about its therapeutic effect on diabetes. The present study was aimed to investigate the cytoprotective activity of silymarin against diabetes-induced cardiomyocyte apoptosis. Methods Rats were randomly divided into： control group, untreated diabetes group and diabetes group treated with silymarin （120 mg/ks·d） for 10 d. Rats were sacrificed, and the cardiac muscle specimens and blood samples were collected. The immunoreactivity of caspase-3 and Bcl-2 in the cardiomyocytes was measured. Total proteins, glucose, insulin, creatinine, AST, ALT, cholesterol, and triglycerides levels were estimated. Results Unlike the treated diabetes group, cardiomyocyte apoptosis increased in the untreated rats, as evidenced by enhanced caspase-3 and declined Bcl-2 activities. The levels of glucose, creatinine, AST, ALT, cholesterol, and triglycerides declined in the treated rats. The declined levels of insulin were enhanced again after treatment of diabetic rats with silymarin, reflecting a restoration of the pancreatic β-cells activity. Conclusion The findings of this study are of great importance, which confirmed for the first time that treatment of diabetic subjects with silymarin may protect cardiomyocytes against apoptosis and promote survival-restoration of the pancreatic β-cells.

Well aligned quasi-straight single-walled carbon nanotubes (SWCNTs) and straight SWCNTs bundle have been prepared in large scale by anode-arc vaporization of gr aphite with metallic catalysts. Various parameters such as the catalyst preparat ion, the kinds and pressure of the buffer gases, the quantity of anode-arc curre nt intensity, and the method of purification have been examined. The influence o f these parameters on the deposited carbon yield is reported, together with obse rvations of the produced material. Improvement in synthetic techniques has resul ted in the optimal conditions for the production of large quantities of high qua lity SWCNTs in our semi-continuous synthesis method. The formation of carbon nan otubes (CNTs) was studied briefly in this paper. Owing to the magnetic pinching effect of arc current, the CNTs arrange in parallel lines along the arc current direction.

A peptide containing the receptor binding site of insulin-like growth factor binding protein-2 enhances bone mass in ovariectomized rats

Male Igfbp2-/-mice have a significant reduction in bone mass and administration of a peptide that contains the insulin-like growth factor binding protein-2(IGFBP-2) receptor-binding domain stimulates bone formation in these animals. Female Igfbp2-/-mice do not have this phenotype but following ovariectomy(OVX) lose more bone than OVX wild-type mice. This suggests that in the absence of estrogen, IGFBP-2 is required to maintain bone mass. Therefore these studies were undertaken to determine if this peptide could stimulate bone acquisition in OVX rats. OVX rats were divided into seven treatment groups: sham animals, OVX animals, OVX animals receiving a control scrambled peptide, or one of three doses of the active peptide termed PEG-HBD-1(0.7, 2,and 6 mg·kg^(-1)) and an OVX group receiving parathyroid hormone(PTH)(50 μg·kg-1 per day). The peptides were administered for8 weeks. DXA revealed a significant reduction in femoral and tibial areal bone mineral density(aBMD) after OVX, whereas treatment with the high-dose peptide increased aBMD by 6.2% ± 2.4%(P < 0.01) compared to control peptide; similar to the increase noted with PTH(5.6% ± 3.0%, P < 0.01). Similar increases were noted with two lower doses of the peptide(3.8% ± 1.5%, P < 0.05 for low dose; 3.1% ± 1.6%, P = 0.07 for middle dose). Micro CT showed that the OVX control peptide animals had reductions of 41% and64% in femoral trabecular BV/TV and trabecular number, respectively. All three doses of the peptide increased bone volume/total volume(BV/TV) significantly, while the low and middle doses increased trabecular number. Cortical BV/TV and thickness at the midshaft increased significantly with each dose of peptide(18.9% ± 9.8%, P < 0.01 and 14.2% ± 7.9%, P < 0.01 for low dose; 23.7% ±10.7%, P < 0.001 and 15.8% ± 6.1%, P < 0.001 for middle dose; 19.0% ± 6.9%, P < 0.01 and 16.2% ± 9.7%, P < 0.001 for high dose)and with PTH(25.8% ± 9.2%, P < 0.001 and 19.4% ± 8.8%, P < 0.001). Microchimerism in recurrent miscarriage

Maternal-fetal cell exchange during pregnancy results in acquisition of microchimerism, which can durably persist in both recipients. Naturally acquired microchimerism may impact maternal-fetal interaction in pregnancy. We conducted studies to ask whether microchimerism that a woman acquired from her own mother is detectable before or during pregnancy in women with recurrent miscarriage. Fetal microchimerism was also assayed. Women with primary idiopathic recurrent miscarriage （n=23） and controls （n=31） were studied. Genotyping was conducted for probands, their mothers and the fetus, a non-shared polymorphism identified and quantitative polymerase chain reaction performed to measure microchimerismin peripheral blood mononuclear cells. Preconception comparisons were made between recurrent miscarriage subjects and controls, using logistic regression and Wilcoxon rank sum. Longitudinal microchimerism in subsequent pregnancies of recurrent miscarriage subjects was described. There was a trend toward lower preconception detection of microchimerism in recurrent miscarriage versus controls, 6% vs. 19% （1/16 vs. 6/31, P=0.2）. During pregnancy, 3111 （27%） of recurrent miscarriage subjects who went on to have a birth had detection of microchimerism from their own mother, whereas neither of two subjects who went on to miscarry had detection （0/2）. This initial data suggest that microchimerism from a woman＇s own mother, while detectable in women with recurrent miscarriage, may differ from controls and according to subsequent pregnancy outcome. Further studies are needed to determine the cell types,quantities and any potential functional role of microchimerism in recurrent miscarriage.

Platelets are reprogrammed by cancer via a process called education,which favors cancer development.The transcriptional profile of tumor-educated platelets(TEPs)is skewed and therefore practicable for cancer detection.This intercontinental,hospital-based,diagnostic study included 761 treatment-naive inpatients with histologically confirmed adnexal masses and 167 healthy controls from nine medical centers(China,n=3;Netherlands,n=5;Poland,n=1)between September 2016 and May 2019.The main outcomes were the performance of TEPs and their combination with CA125 in two Chinese(VC1 and VC2)and the European(VC3)validation cohorts collectively and independently.Exploratory outcome was the value of TEPs in public pan-cancer platelet transcriptome datasets.The AUCs for TEPs in the combined validation cohort,VC1,VC2,and VC3 were 0.918(95%CI 0.889-0.948),0.923(0.855-0.990),0.918(0.872-0.963),and 0.887(0.813-0.960),respectively.Combination of TEPs and CA125 demonstrated an AUC of 0.922(0.889-0.955)in the combined validation cohort;0.955(0.912-0.997)in VC1;0.939(0.901-0.977)in VC2;0.917(0.824-1.000)in VC3.For subgroup analysis,TEPs exhibited an AUC of o.858,0.859,and 0.920 to detect early-stage,borderline,non-epithelial diseases and 0.899 to discriminate ovarian cancer from endometriosis.TEPs had robustness,compatibility,and universality for preop.erative diagnosis of ovarian cancer since it withstood validations in populations of different ethnicities,heterogeneous histoiogical subtypes,and early-stage ovarian cancer.However,these observations warrant prospective validations in a larger population beforeclinicalutilities.

Modeling the oxidative consumption of curcumin from controlled released poly(betaamino ester)microparticles in the presence of a free radical generating system

Despite the promise of its therapeutic benefits,curcumin as a free molecule has failed to demonstrate significant clinical success.Arguably,its inherently poor stability and rapid clearance is a significant reason for these negative outcomes.The incorporation of curcumin into the backbone of a crosslinked hydrogel that utilizes poly(beta-amino ester)(PBAE)chemistry can provide a tunable protective network with the ability to release at a controlled rate while improving its therapeutic potential.Kinetics of curcumin conjugated PBAE microparticles controlled release delivery system in the presence of oxidative environments was studied for the first time,where consumption rates of active curcumin and release products were obtained.The constituent amount of curcumin present in solution was improved by incorporating the active into the network in comparison to curcumin as a free drug.Modeling curcumin conjugated PBAE microparticles will provide a design platform to improve translation and overall success in delivering a therapeutic agent that matches levels of oxidative stress.

Liver Fibrosis Helps to Distinguish Autoimmune Hepatitis from DILI with Autoimmune Features: A Review of Twenty Cases

background and Aims:Drug-induced liver injury with autoimmune features(AI-DILI)mimics the clinical presentation,and laboratory and pathologic features of idiopathic autoimmune hepatitis(AIH).We aimed to identify histopathologic hallmarks to differentiate these entities.Methods:All liver biopsies archived for the past 10 years were reviewed retrospectively to identify cases of recently detected liver injury associated with predominantly lymphoplasmacytic interphase hepatitis,positive markers for liver autoimmunity,and negative tests for viral hepatitis.Twenty cases were divided into AIH(n=12)or AI-DILI(n=8)groups.Blind qualitative evaluation of necroinflammatory changes and liver fibrosis were performed according to the Scheuer scoring system.Cellular densities were determined using Imagej(V1.51t,National Institutes of Health,Bethesda,MD,USA).Fibrosis was assessed on Masson trichrome-stained slides,and collagen deposition was estimated following a protocol of color deconvolution.Results:Necroinflammatory changes as well as densities(portal and lobular)of neutrophils and eosinophils,intracellular cholestasis,and regenerative changes did not differ between the two groups(P≥0.05).Neutrophil densities but not eosinophils showed a positive correlation with the severity of hepatocellular damage(r=0.6and 0.58,vs.alanine aminotransferase,P<0.05).Ceroidladen macrophages but not histiocytic aggregates appeared to be more common in AI-DILI(P<0.05).AIH patients presented more often with evidence of chronic damage,including higher scores of fibrosis and collagen deposition,in comparison to AI-DILI(P<0.05).Conclusions:Although there is no histologic feature pathognomonic for AI-DILI or AIH,advanced stages of liver fibrosis can be used to support the diagnosis of AIH in some cases.Definitive diagnosis of AI-DILI requires follow-up and demonstration of complete remission after drug withdrawal with no need for immunosuppression.
