Investigation of HcAR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving the herd. In vitro administration of beta-hydroxybutyric acid(BHBA;the primary blood ketone) has inhibitory effects on the function of bovine leukocytes. BHBA is a ligand of HcAR2 and the activation of these receptors promotes an anti-inflammatory response which may be related with immunosuppression observed in transition dairy cattle. The objective of this study was to identify and test antagonists for HcAR2 in bovine immune cells cultured with *** We observed expression of HcAR2 at the protein level within lymphocytes, monocytes, and granulocytes. The proportion of cells expressing HcAR2 tended to be greater in mid-lactation compared to early lactation cows;the increase was a result of increased proportion of T and B cells expressing HcAR2. Stimulation of HcAR2 with niacin or BHBA promoted ca^(2+) mobilization in neutrophils and mononuclear cells. Mononuclear cells treated with BHBA had diminished intracellular ca^(2+) responses when HcAR2 was knocked down by si RNA silencing, indicating ca^(2+) mobilization was mediated by HcAR2 signaling. Two candidate antagonists for HcAR2, synthesized from niacin(NA-1 and NA-5), were tested;monocytes and neutrophils pre-treated with NA-1 and NA-5 had reduced ca^(2+) mobilization after incubation with BHBA. Furthermore, NA-5 but not NA-1 prevented BHBA-associated reductions in cyclic *** We demonstrated that HcAR2 is present on bovine leukocytes and has greater expression later in lactation. We confirmed that BHBA and niacin derived HcAR2 antagonists alter bovine leukocyte activity. Our results demonstrate that both BHBA and niacin affect bovine leukocyte ca^(2+) mobilization in a HcAR2-dependent manner.

Home Energy Management Systems(HEMS)are increasingly relevant for demand-side management at the residential level by collecting data(energy,weather,electricity prices)and controlling home appliances or storage *** control can be performed with classical models that find optimal solutions,with high real-time computational cost,or data-driven approaches,like Reinforcement Learning,that find good and flexible solutions,but depend on the availability of load and generation data and demand high computational resources for *** this work,a novel HEMS is proposed for the optimization of an electric battery operation in a real,online and data-driven environment that integrates state-of-the-art load forecasting combining cNN and LSTM neural networks to increase the robustness of *** Reinforcement Learning agents are trained with different algorithms(Double DQN,Dueling DQN,Rainbow and Proximal Policy Optimization)in order to minimize the cost of electricity purchase and to maximize photovoltaic self-consumption for a PV-Battery residential *** show that the best Reinforcement Learning agent achieves a 35%reduction in total cost when compared with an optimization-based agent.

Na-ion cathode materials with a fast charge and discharge behavior are needed to develop future high energy sodium-ion batteries(SIBs).However,inevitably complicated phase transitions and sluggish kinet ics during insertion and removal of Na+in P_(2)-type layered transition metal oxides generate structura instability and severe capacity *** get rid of such a dilemma,we report a structural optimization strategy to promote P2-type layered transition metal oxides with more(010)active planes as an efficien cathode for *** a result,as-prepared hexagonal-prism P2-type layered Na_(0.71)Ni_(0.16)Li_(0.09)co_(0.16)Mn0.6O_(2)cathode with more(010)active planes delivers a reversible capacity of 120.1 mAh/g at 0.1 c,impressive rate capability of 52.7 m Ah/g at 10 c,and long-term cycling stability(capacity retention of 95.6%ove200 cycles).The outstanding electrochemical performance benefited from the unique hexagonal-prism with more(010)active facets,which can effectively shorten the diffusion distances of Na+,increase the Na-ion migration dynamics and nanostructural stability during cycling verified by morphology character ization,Rietveld refinement,GITT,density functional theory calculations and operando XRD.

BAcKGROUND There is concern regarding potential long-term cardiotoxicity with systemic distribution of metals in total joint arthroplasty(TjA)*** To determine the association of commonly used implant metals with echocardiographic measures in TjA *** The study comprised 110 TjA patients who had a recent history of high chromium,cobalt or titanium *** underwent two-dimensional,three-dimensional,Doppler and speckle-strain transthoracic echocardiography and a blood draw to measure metal *** and sex-adjusted linear and logistic regression models were used to examine the association of metal concentrations(exposure)with echocardiographic measures(outcome).RESULTS Higher cobalt concentrations were associated with increased left ventricular end-diastolic volume(estimate 5.09;95%cI:0.02-10.17)as well as left atrial and right ventricular dilation,particularly in men but no changes in cardiac *** titanium concentrations were associated with a reduction in left ventricle global longitudinal strain(estimate 0.38;95%cI:0.70 to 0.06)and cardiac index(estimate 0.08;95%cI,-0.15 to-0.01).cONcLUSION Elevated cobalt and titanium concentrations may be associated with structural and functional cardiac changes in some *** studies are warranted to better understand the systemic effects of metals in TjA patients.

The evolution and functional divergence of 10 Apolipoprotein D-like genes in Nilaparvata lugens

Apolipoprotein D(ApoD),a member of the lipocalin superfamily of proteins,is involved in lipid transport and stress *** only a single copy of the ApoD gene is found in humans and some other vertebrates,there are typically several ApoD-like genes in *** date,there have been relatively few studies that have examined the evolution and functional differentiation of ApoD-like genes in insects,particularly hemi-metabolous *** this study,we identified 10 ApoD-like genes(NlApoD1−10)with distinct spatiotemporal expression patterns in Nilaparvata lugens(BPH),which is an important pest of ***1−10 were found to be distributed on 3 chromosomes in a tandem array of NlApoD1/2,NlApoD3−5,and NlApoD7/8,and show sequence and gene structural divergence in the coding regions,indicating that multiple gene duplication events occurred during *** analysis revealed that NlApoD1−10 can be clustered into 5 clades,with NlApoD3−5 and NlApoD7/8 potentially evolving exclusively in the Delphacidae *** screening using an RNA interference approach revealed that only NlApoD2 was essential for BPH development and survival,whereas NlApoD4/5 are highly expressed in testes,and might play roles in ***,stress response analysis revealed that NlApoD3−5/9,NlApoD3−5,and NlApoD9 were up-regulated after treatment with lipopolysaccharide,H2O2,and ultraviolet-c,respectively,indicating their potential roles in stress resistance.

Prime editing is a versatile cRISPR/cas-based precise genome-editing technique for crop *** new types of prime editors(PEs)named PE6a–d were recently generated using evolved and engineered reverse transcriptase(RT)variants from three different *** this study,we tested the editing efficiencies of four PE6 variants and two additional PE6 constructs with double-RT modules in transgenic rice(Oryza sativa)***6c,with an evolved and engineered RT variant from the yeast Tf1 retrotransposon,yielded the highest prime-editing *** average fold change in the editing efficiency of PE6c compared with PEmax exceeded 3.5 across 18 agronomically important target sites from 15 *** also demonstrated the feasibility of using two RT modules to improve prime-editing *** results suggest that PE6c or its derivatives would be an excellent choice for prime editing in monocot *** addition,our findings have laid a foundation for prime-editing-based breeding of rice varieties with enhanced agronomically important traits.

Rescattering of stimulated Raman side scattering(SRSS)is observed for the first time via two-dimensional(2D)particle-in-cell(PIc)*** construct a theoretical model for the rescattering process,which can predict the region of occurrence of mth-order SRSS and estimate its *** rescattering process is identified by the 2D PIc simulations under typical conditions of a direct-drive inertial confinement fusion *** electrons produced by second-order SRSS propagate nearly perpendicular to the density gradient and gain nearly the same energy as in first-order SRSS,but there is no cascade acceleration to produce superhot *** studies for a wide range of ignition conditions show that SRSS and associated rescatterings are robust and important processes in inertial confinement fusion.

Effects of nursing team communication and collaboration on treatment outcomes in intensive care unit patients with severe pneumonia

BAcKGROUND Severe pneumonia is a common severe respiratory infection worldwide,and its treatment is challenging,especially for patients in the intensive care unit(IcU).AIM To explore the effect of communication and collaboration between nursing teams on the treatment outcomes of patients with severe pneumonia in *** We retrospectively analyzed 60 patients with severe pneumonia who were treated at the IcU of the hospital between january 1,2021 and December 31,*** compared and analyzed the respiratory mechanical indexes[airway resistance(Raw),mean airway pressure(mPaw),peak pressure(PIP)],blood gas analysis indexes(arterial oxygen saturation,arterial oxygen partial pressure,and oxygenation index),and serum inflammatory factor levels[c-reactive protein(cRP),procalcitonin(PcT),cortisol(cOR),and high mobility group protein B1(HMGB1)]of all patients before and after *** Before treatment,there was no significant difference in respiratory mechanics index and blood gas analysis index between 2 groups(P>0.05).However,after treatment,the respiratory mechanical indexes of patients in both groups were significantly improved,and the improvement of Raw,mPaw,plateau pressure,PIP and other indexes in the combined group after communication and collaboration with the nursing team was significantly better than that in the single care group(PcRP and PcT levels of patients were significantly decreased,and the difference was statistically significant compared with that of nursing group alone(PcOR and HMGB1 before and after treatment were also significantly decreased between the two *** The communication and collaboration of the nursing team have a significant positive impact on respiratory mechanics indicators,blood gas analysis indicators and serum inflammatory factor levels in the treatment of severe pneumonia patients in IcU.

A Sharding Scheme Based on Graph Partitioning Algorithm for Public Blockchain

Blockchain technology,with its attributes of decentralization,immutability,and traceability,has emerged as a powerful catalyst for enhancing traditional industries in terms of optimizing business ***,transaction performance and scalability has become the main challenges hindering the widespread adoption of *** to its inability to meet the demands of high-frequency trading,blockchain cannot be adopted in many *** improve the transaction capacity,researchers have proposed some on-chain scaling technologies,including lightning networks,directed acyclic graph technology,state channels,and shardingmechanisms,inwhich sharding emerges as a potential scaling ***,excessive cross-shard transactions and uneven shard workloads prevent the sharding mechanism from achieving the expected *** paper proposes a graphbased sharding scheme for public blockchain to efficiently balance the transaction *** crossshard transactions and evening-out workloads among shards,the scheme reduces transaction confirmation latency and enhances the transaction capacity of the ***,the scheme can achieve a high-frequency transaction as well as a better blockchain *** results show that the scheme effectively reduces the cross-shard transaction ratio to a range of 35%-56%and significantly decreases the transaction confirmation latency to 6 s in a blockchain with no more than 25 shards.
