BAcKGROUND There is concern regarding potential long-term cardiotoxicity with systemic distribution of metals in total joint arthroplasty(TjA)*** To determine the association of commonly used implant metals with echocardiographic measures in TjA *** The study comprised 110 TjA patients who had a recent history of high chromium,cobalt or titanium *** underwent two-dimensional,three-dimensional,Doppler and speckle-strain transthoracic echocardiography and a blood draw to measure metal *** and sex-adjusted linear and logistic regression models were used to examine the association of metal concentrations(exposure)with echocardiographic measures(outcome).RESULTS Higher cobalt concentrations were associated with increased left ventricular end-diastolic volume(estimate 5.09;95%cI:0.02-10.17)as well as left atrial and right ventricular dilation,particularly in men but no changes in cardiac *** titanium concentrations were associated with a reduction in left ventricle global longitudinal strain(estimate 0.38;95%cI:0.70 to 0.06)and cardiac index(estimate 0.08;95%cI,-0.15 to-0.01).cONcLUSION Elevated cobalt and titanium concentrations may be associated with structural and functional cardiac changes in some *** studies are warranted to better understand the systemic effects of metals in TjA patients.

Prime editing is a versatile cRISPR/cas-based precise genome-editing technique for crop *** new types of prime editors(PEs)named PE6a–d were recently generated using evolved and engineered reverse transcriptase(RT)variants from three different *** this study,we tested the editing efficiencies of four PE6 variants and two additional PE6 constructs with double-RT modules in transgenic rice(Oryza sativa)***6c,with an evolved and engineered RT variant from the yeast Tf1 retrotransposon,yielded the highest prime-editing *** average fold change in the editing efficiency of PE6c compared with PEmax exceeded 3.5 across 18 agronomically important target sites from 15 *** also demonstrated the feasibility of using two RT modules to improve prime-editing *** results suggest that PE6c or its derivatives would be an excellent choice for prime editing in monocot *** addition,our findings have laid a foundation for prime-editing-based breeding of rice varieties with enhanced agronomically important traits.

Apolipoprotein D(ApoD),a member of the lipocalin superfamily of proteins,is involved in lipid transport and stress *** only a single copy of the ApoD gene is found in humans and some other vertebrates,there are typically several ApoD-like genes in *** date,there have been relatively few studies that have examined the evolution and functional differentiation of ApoD-like genes in insects,particularly hemi-metabolous *** this study,we identified 10 ApoD-like genes(NlApoD1−10)with distinct spatiotemporal expression patterns in Nilaparvata lugens(BPH),which is an important pest of ***1−10 were found to be distributed on 3 chromosomes in a tandem array of NlApoD1/2,NlApoD3−5,and NlApoD7/8,and show sequence and gene structural divergence in the coding regions,indicating that multiple gene duplication events occurred during *** analysis revealed that NlApoD1−10 can be clustered into 5 clades,with NlApoD3−5 and NlApoD7/8 potentially evolving exclusively in the Delphacidae *** screening using an RNA interference approach revealed that only NlApoD2 was essential for BPH development and survival,whereas NlApoD4/5 are highly expressed in testes,and might play roles in ***,stress response analysis revealed that NlApoD3−5/9,NlApoD3−5,and NlApoD9 were up-regulated after treatment with lipopolysaccharide,H2O2,and ultraviolet-c,respectively,indicating their potential roles in stress resistance.

Rescattering of stimulated Raman side scattering(SRSS)is observed for the first time via two-dimensional(2D)particle-in-cell(PIc)*** construct a theoretical model for the rescattering process,which can predict the region of occurrence of mth-order SRSS and estimate its *** rescattering process is identified by the 2D PIc simulations under typical conditions of a direct-drive inertial confinement fusion *** electrons produced by second-order SRSS propagate nearly perpendicular to the density gradient and gain nearly the same energy as in first-order SRSS,but there is no cascade acceleration to produce superhot *** studies for a wide range of ignition conditions show that SRSS and associated rescatterings are robust and important processes in inertial confinement fusion.

BAcKGROUND Severe pneumonia is a common severe respiratory infection worldwide,and its treatment is challenging,especially for patients in the intensive care unit(IcU).AIM To explore the effect of communication and collaboration between nursing teams on the treatment outcomes of patients with severe pneumonia in *** We retrospectively analyzed 60 patients with severe pneumonia who were treated at the IcU of the hospital between january 1,2021 and December 31,*** compared and analyzed the respiratory mechanical indexes[airway resistance(Raw),mean airway pressure(mPaw),peak pressure(PIP)],blood gas analysis indexes(arterial oxygen saturation,arterial oxygen partial pressure,and oxygenation index),and serum inflammatory factor levels[c-reactive protein(cRP),procalcitonin(PcT),cortisol(cOR),and high mobility group protein B1(HMGB1)]of all patients before and after *** Before treatment,there was no significant difference in respiratory mechanics index and blood gas analysis index between 2 groups(P>0.05).However,after treatment,the respiratory mechanical indexes of patients in both groups were significantly improved,and the improvement of Raw,mPaw,plateau pressure,PIP and other indexes in the combined group after communication and collaboration with the nursing team was significantly better than that in the single care group(PcRP and PcT levels of patients were significantly decreased,and the difference was statistically significant compared with that of nursing group alone(PcOR and HMGB1 before and after treatment were also significantly decreased between the two *** The communication and collaboration of the nursing team have a significant positive impact on respiratory mechanics indicators,blood gas analysis indicators and serum inflammatory factor levels in the treatment of severe pneumonia patients in IcU.

Blockchain technology,with its attributes of decentralization,immutability,and traceability,has emerged as a powerful catalyst for enhancing traditional industries in terms of optimizing business ***,transaction performance and scalability has become the main challenges hindering the widespread adoption of *** to its inability to meet the demands of high-frequency trading,blockchain cannot be adopted in many *** improve the transaction capacity,researchers have proposed some on-chain scaling technologies,including lightning networks,directed acyclic graph technology,state channels,and shardingmechanisms,inwhich sharding emerges as a potential scaling ***,excessive cross-shard transactions and uneven shard workloads prevent the sharding mechanism from achieving the expected *** paper proposes a graphbased sharding scheme for public blockchain to efficiently balance the transaction *** crossshard transactions and evening-out workloads among shards,the scheme reduces transaction confirmation latency and enhances the transaction capacity of the ***,the scheme can achieve a high-frequency transaction as well as a better blockchain *** results show that the scheme effectively reduces the cross-shard transaction ratio to a range of 35%-56%and significantly decreases the transaction confirmation latency to 6 s in a blockchain with no more than 25 shards.

Obj.c.ive To explore the neuroprotective effects of the Shaoyao Gancao decoction(SGD)against excitatory damage in Pc12 cells and the role of the Src-NR2-nNOS pathway mediation by SGD in regulatingγ-aminobutyric acid(GABA)-glutamate(Glu)*** N-Methyl-d-aspartic acid(NMDA)was used to establish a Pc12 cell excitability injury *** investigate the neuroprotective effect of SGD,a cell counting kit-8(ccK-8)assay was used to determine Pc12 cell viability,Annexin V/Propidium Iodide(Annexin V/PI)double staining was used to determine Pc12 cell apoptosis,and ca^(2+)concentration was observed using laser confocal *** receptor agonists and antagonists were used to analyze the neuroprotective interactions betweenγ-aminobutyric acid(GABA)and NMDA ***,molecular biology techniques were used to determine mRNA and protein expression in the Src-NR2-nNOS *** analyzed the correlations between the regulatory sites of GABA and NMDA interactions,excitatory neurotoxicity,and brain damage at the molecular *** NMDA excitotoxic injury manifested as a significant decrease in cell activity,increased apoptosis and caspase-3 protein expression,and a significant increase in intracellular ca^(2+)*** of SGD,a GABAA receptor agonist(muscimol),or a GABAB receptor agonist(baclofen)decreased intracellular ca^(2+)concentrations,attenuated apoptosis,and reversed NMDA-induced upregulation of caspase-3,Src,NMDAR2A,NMDAR2B,and ***,a GABA_(A)receptor antagonist(bicuculline)and a GABA_(B)receptor antagonist(saclofen)failed to significantly increase excitatory *** Taken together,these results not only provide an experimental basis for SGD administration in the clinical treatment of central nervous system injury diseases,but also suggest that the Src-NR2A-nNOS pathway may be a valuable target in excitotoxicity treatment.

The evolution of light-skin pigmentation among Eurasians is considered as an adaptation to the high-latitude *** Asians are ideal populations for studying skin color evolution because of the complex environment of East ***,we report a strong selection signal for the pigmentation gene phenylalanine hydroxylase(PAH)in light-skinned Han chinese *** intron mutation rs10778203 in PAH is enriched in East Asians and is significantly associated with skin color of the back of the hand in Han chinese males(Pciferase and transcription factor binding assays show that the ancestral allele of rs10778203 could bind to SMAD2 and has a significant enhancer activity for ***,the derived T allele(the major allele in East Asians)of rs10778203 decreases the binding activity of transcription factors and enhancer ***,the derived T allele of rs10778203 shows a weaker ultraviolet radiation response in A375 cells and zebrafish ***,rs10778203 decreases melanin production in transgenic zebrafish embryos after ultraviolet B(UVB)***,PAH is a potential pigmentation gene that regulates skin tanning *** selection has enriched the adaptive allele,resulting in weakened tanning ability in East Asians,suggesting a unique genetic mechanism for evolutionary skin lightening in East Asians.
