Research on Hepatocyte Regulation of PcSK9-LDLR and Its Related Drug Targets

The prevalence of hyperlipidemia has increased significantly due to genetic, dietary, nutritional and pharmacological factors, and has become one of the most common pathological conditions in humans. Hyperlipidemia can lead to a range of diseases such as atherosclerosis, stroke, coronary heart disease, myocardial infarction, diabetes, and kidney failure, etc. High circulating low-density lipoprotein cholesterol (LDL-c) is one of the causes of hyperlipidemia. LDL-c in the blood binds to LDL receptor (LDLR) and regulates cholesterol homeostasis through endocytosis. In contrast, proprotein convertase subtilisin/kexin type 9 (PcSK9) mediates LDLR degradation via the intracellular and extracellular pathways, leading to hyperlipidemia. Targeting PcSK9-synthesizing transcription factors and downstream molecules are important for development of new lipid-lowering drugs. clinical trials regarding PcSK9 inhibitors have demonstrated a reduction in atherosclerotic cardiovascular disease events. The purpose of this review was to explore the target and mechanism of intracellular and extracellular pathways in degradation of LDLR and related drugs by PcSK9 in order to open up a new pathway for the development of new lipid-lowering drugs.

Burgess Shale-type deposits provide a wealth of information on the early evolution of *** that are central to understanding the exceptional preservation of these biotas and the paleoenvironments they inhabited may be obscured by the post-depositional alteration due to metamorphism at depth and weathering near the Earth’s *** over 50 cambrian BST biotas,the chengjiang and Qingjiang deposits are well known for their richness of soft-bodied taxa,fidelity of preservation,and Early cambrian *** alteration via weathering has been well-investigated,the thermal maturity of the units bearing the two biotas has not yet been *** we investigate peak metamorphic temperatures of the two deposits using two independent *** gradient analyses demonstrate that the most fossiliferous sections of the chengjiang were buried at a maximum depth of∼8500 m in the Early Triassic,corresponding to∼300°c,while the type area of the Qingjiang biota was buried at a maximum depth of∼8700 m in the Early jurassic,corresponding to∼240°*** geothermometer analyses of fossil carbonaceous material demonstrate that peak temperatures varied across localities with different burial *** two productive sections of the chengjiang biota were thermally altered at a peak temperature of approximately 300°c,and the main locality of the Qingjiang biota experienced a peak temperature of 238±22°*** results from two independent methods are *** BST deposits for which thermal maturity has been documented,the Qingjiang biota is the least thermally mature,and therefore holds promise for enriching our understanding of BST deposits.

The contents of waste glass powder(WGP)(0%,10%,15%,20%,25%)and water-binder ratio(W/c)(0.24,0.26,0.28)were used as influencing factors,and the quality loss rate(Δm)and compressive strength loss rate(Δfc)were used as characterization *** ca/Si ratio and main element contents of c-S-H gels with different WGP content were investigated by energy dispersive spectrometry(EDS).The pore structure evolution characteristics of WGP composite cementing materials were investigated by low field nuclear magnetic resonance(NMR).UsingΔfc as the index of frost resistance degradation and Weibull function,the frost resistance degradation of glass doped pervious concrete(WGP-Pc)was *** results show that,with WGP,for the same number of cycles,Δm andΔfc decrease and increase with the increase of *** the same WGP content,Δm andΔfc decrease first and then increase with the increase of W/*** 100 freeze-thaw cycles,the samples with WGP content of 20%and W/c of 0.26 have the best *** tests show that with the increase of WGP content,the ca/Si ratio of c-S-H gel decreases at first and then increases with the increase of WGP *** extreme value of ca/Si is 2.36 when WGP is added by 20%.The pore volume of hardened paste with 20%WGP content decreased by 18.6%compared with that of cement system without *** overall compactness of the specimen was *** the basis of the test data,a life prediction model was established according to Weibull *** experiment showed thatΔfc could be used as a durability degradation index,and the slope of the reliability curve became gentle after WGP was added,which reduced the damage degradation rate of Pc.W/c was ***'s about 5000 hours.

Herein,a novel interference-free surface-enhanced Raman spectroscopy(SERS)strategy based on magnetic nanoparticles(MNPs)and aptamer-driven assemblies was proposed for the ultrasensitive detection of histamine.A core-satellite SERS aptasensor was constructed by combining aptamer-decorated Fe_(3)O_(4)@Au MNPs(as the recognize probe for histamine)and complementary DNA-modified silver nanoparticles carrying 4-mercaptobenzonitrile(4-MBN)(Ag@4-MBN@Ag-c-DNA)as the SERS signal probe for the indirect detection of *** an applied magnetic field in the absence of histamine,the assembly gave an intense Raman signal at“Raman biological-silent”region due to *** the presence of histamine,the Ag@4-MBN@Ag-c-DNA SERS-tag was released from the Fe_(3)O_(4)@Au MNPs,thus decreasing the SERS *** optimal conditions,an ultra-low limit of detection of 0.65×10^(-3)ng/mL and a linear range 10^(-2)-10^5 ng/mL on the SERS aptasensor were *** histamine content in four food samples were analyzed using the SERS aptasensor,with the results consistent with those determined by high performance liquid *** present work highlights the merits of indirect strategies for the ultrasensitive and highly selective SERS detection of small biological molecules in complex matrices.

The development of efficient non-precious metal catalysts is important for the large-scale application of alkaline hydrogen evolution reaction(HER).Here,we synthesized a composite catalyst of cu and Mo_(2)c(cu/Mo_(2)c)using Anderson-type polyoxometalates(POMs)synthesized by the facile soaking method as *** electronic interaction between cu and Mo_(2)c drives the positive charge of cu,alleviating the strong adsorption of hydrogen at the Mo site by modulating the d-band center of Mo_(2)*** studying the interfacial water structure using in situ attenuated total reflection surface-enhanced infrared absorption spectroscopy(ATR-SEIRAS),we determined that the positively charged cu crystals have the function of activating water molecules and optimizing the interfacial water *** interfacial water of cu/Mo_(2)c contains a large amount of free water,which could facilitate the transport of reaction *** to activated water molecules and optimized interfacial water structure and hydrogen adsorption energy,the overpotential of cu/Mo_(2)c is 24 mV at a current density of 10 mA·cm^(-2) and 178 mV at a current density of 1000 mA·cm^(-2).This work improves catalyst performance in terms of interfacial water structure optimization and deepens the understanding of water-mediated catalysis.
