*** strongest locus which associated with type 2 diabetes(T2D)by the common variant rs7903146 is the transcription factor 7-like 2 gene(TCF7L2).We aimed to quantify the interaction of diet/lifestyle interventions and the genetic effect of TCF7L2 rs7903146 on glycemic traits,body weight,or waist circumference in overweight or obese adults in several randomized controlled trials(RCTs).*** October 2016 to May 2018,a large collaborative analysis was performed by pooling individualparticipant data from 7 *** RCTs reported changes in glycemic control and adiposity of the variant rs7903146 after dietary/lifestyle-related interventions in overweight or obese *** treatment interaction models which used the genetic effect encoded by the allele dose and common covariates were applicable to individual participant data in all *** the joint analysis,a total of 7 eligible RCTs were included(n=4,114).Importantly,we observed a significant effect modification of diet/lifestyle-related interventions on the TCF7L2 variant rs7903146 and changes in fasting *** with the control group,diet/lifestyle interventions were related to lower fasting glucose by-3.06(95%CI,-5.77 to-0.36)mg/dL(test for heterogeneity and overall effect:I^(2)=45:1%,p<0:05;z=2:20,p=0:028)per one copy of the TCF7L2 T risk ***,regardless of genetic risk,diet/lifestyle interventions were associated with lower waist ***,there was no significant change for diet/lifestyle interventions in other glycemic control and adiposity traits per one copy of TCF7L2 risk *** findings suggest that carrying the TCF7L2 T risk allele may have a modestly greater benefit for specific diet/lifestyle interventions to improve the control of fasting glucose in overweight or obese adults.

Complementary analysis of microsatellite tumor profile and mismatch repair defects in colorectal carcinomas

Microsatellite instability (MSI) is a prognostic factor and a marker of defi cient mismatch repair (MMR) in colorectal adenocarcinomas (CRC). However, a proper application of this marker requires understanding the following: (1) The MSI concept: The PCR approach must amplify the correct locus and accurately identify the microsatellite pattern in the patient’s normal tissue. MSI is demonstrat- ed when the length of DNA sequences in a tumor differs from that of nontumor tissue. Any anomalous expansion or reduction of tandem repeats results in extra-bands normally located in the expected size range (100 bp, above or below the expected product), differ from the germline pattern by some multiple of the repeating unit, and must show appropriate stutter. (2) MSI mechanisms: MMR gene inactivation (by either mutation or protein down-regulation as frequently present in deep CRC com- partments) leads to mutation accumulation in a cell with every cellular division, resulting in malignant transforma- tion. These mechanisms can express tumor progression and result in a decreased prevalence of aneuploid cells and loss of the physiologic cell kinetic correlations in the deep CRC compartments. MSI molecular mechanisms are not necessarily independent from chromosomal in- stability and may coexist in a given CRC. (3) Because of intratumoural heterogeneity, at least two samples from each CRC should be screened, preferably from the su- perfi cial (tumor cells above the muscularis propria) and deep (tumor cells infi ltrating the muscularis propria) CRC compartments to cover the topographic tumor hetero- geneity. (4) Pathologists play a critical role in identify- ing microsatellite-unstable CRC, such as occur in young patients with synchronous or metachronous tumors or with tumors showing classic histologic features. Anemia in inflammatory bowel disease: A neglected issue with relevant effects

Anemia, a common complication associated with inflammatory bowel disease (IBD), is frequently overlooked in the management of IBD patients. Unfortunately, it represents one of the major causes of both decreased quality of life and increased hospital admissions among this population. Anemia in IBD is pathogenically complex, with several factors contributing to its development. While iron deficiency is the most common cause, vitamin B12 and folic acid deficiencies, along with the effects of pro-inflammatory cytokines, hemolysis, drug therapies, and myelosuppression, have also been identified as the underlying etiology in a number of patients. Each of these etiological factors thus needs to be identified and corrected in order to effectively manage anemia in IBD. Because the diagnosis of anemia in IBD often presents a challenge, combinations of several hematimetric and biochemical parameters should be used. Recent studies underscore the importance of determining the ferritin index and hepcidin levels in order to distinguish between iron deficiency anemia, anemia due to chronic disease, or mixed anemia in IBD patients. With regard to treatment, the newly introduced intravenous iron formulations have several advantages over orally-administered iron compounds in treating iron deficiency in IBD. In special situations, erythropoietin supplementation and biological therapies should be considered. In conclusion, the management of anemia is a complex aspect of treating IBD patients, one that significantly influences the prognosis of the disease. As a consequence, its correction should be considered a specific, first-line therapeutic goal in the management of these patients.

Bio-Imitation of Mexican Migration Routes to the USA with Slime Mould on 3D Terrains

Plasmodium ofPhysarum polycephalum （P. polycephalum） is a large single cell visible by an unaided eye. It shows sophisticated behavioural traits in foraging for nutrients and developing an optimal transport network of protoplasmic tubes spanning sources of nutrients. When placed in an environment with distributed sources of nutrients the cell computes＇ an optimal graph spanning the nutrients by growing a network of protoplasmic tubes. P. polycephalum imitates development ofman-made transport networks of a country when configuration of nutrients represents major urban areas, We employed this feature of the slime mould to imitate mexican migration to USA. The Mexican migration to USA is the World＇s larger migration system. We bio-physically imitated the migration using slime mould P. poIycephalum. In laboratory experiments with 3D Nylon terrains of USA we imitated development of migratory routes from Mexico-USA border to ten urban areas with high concentration of Mexican migrants. From results of laboratory experiments we extracted topologies of migratory routes, and highlighted a role of elevations in shaping the human movement networks.

Host-parasite interactions under extreme climatic conditions

The effect that climatic changes can exert on parasitic interactions represents a multifactor problem whose results are difficult to predict. The actual impact of changes will depend on their magnitude and the physiological tolerance of affected organisms. When the change is considered extreme （i.e. unusual weather events that are at the extremes of the historical distribution for a given area）, the probability of an alteration in an organisms＇ homeostasis increases dramatically. However, factors determining the altered dynamics of host-parasite interactions due to an extreme change are the same as those acting in response to changes of lower magnitude. Only a deep knowledge of these factors will help to produce more accurate predictive models for the effects of extreme changes on parasitic interactions. Extreme environmental conditions may affect pathogens directly when they include free-living stages in their life-cycles and indirectly through reduced resource availability for hosts and thus reduced ability to produce efficient anti-parasite defenses, or by effects on host density affecting transmission dynamics of diseases or the frequency of intraspecific contact. What are the consequences for host-parasite interactions？ Here we summarize the present knowledge on three principal factors in determining host-parasite associations; biodiversity, population density and immunocompetence In addition, we analyzed examples of the effects of environmental alteration of anthropogenic origin on parasitic systems because the effects are analogous to that exerted by an extreme climatic change [Current Zoology 57 （3）： 390405, 2011].

Ligand effects on the structure and vibrational properties of the thiolated Au18 cluster

Most of the studies devoted to thiolated gold clusters suppose that their core and Au-S framework do not suffer from distortion independently of the protecting ligands（-SR） and it is assumed as correct to simplify the ligand as SCH3. In this work is delivered a systematic study of the structure and vibrational properties（IR and Raman）of the Au18（SR）14cluster. The pursued goal is to understand the dependency of the displayed vibrational properties of the thiolated Au18cluster with the ligands type. A set of six ligands was considered during calculations of the vibrational properties based on density functional theory（DFT） and in its dispersioncorrected approach（DFT-D）.

This study investigates the influence of electropolymerization conditions on the deposition of polypyrrole(PPy)onto cotton-derived carbon fiber(CF)modified with reduced graphene oxide(rGO)for supercapacitors applications using an experimental/theorical *** surface modification of CF by rGO and/or by PPy electrodeposited at 10,25 and 50 mV s^(-1) was thoroughly examined physicochemical and *** electrodes comprising CF-rGo-PPy,synthesized via electropolymerization at 25 mV s^(-1),demonstrated a remarkable increase in capacitance,showcasing~742 F g^(-1) compared to 153 F g^(-1) for ***,N_(2)-surface area,XPS,and TD-DFT approach revealed that the higher capacitance observed in CF-rGo-PPy electrodes underscores the influence of morphology and charged nitrogen species on the electrochemical performance of these modified ***,this electrode material achieves a specific capacitance retention of~96%of their initial capacitance after 10000 cycles at 0.5 A g^(-1) measured in a two-electrodes cell *** work also discusses the influence of the scan rate used for pyrrole electropolymerization on the pseudocapacitance contribution of PPy and its possible effect on the porosity of the *** results highlight the importance of appropriate electropolymerization conditions that allow obtaining the synergistic effect between CF,rGO and PPy.

Isolated energy systems in archipelagos are characterized for having a great dependence on fossil sources due to isolation and territorial *** island of La Palma is situated on the northwest of the Canary Islands,and its electric system is very *** policies planned by local authorities are aimed to increase the share of renewable energies and the reduction of fossil ***,intermittence and the concentration of unmanageable renewable energies in few locations may hinder the operation of the *** order to solve these problems,energy storage plays an essential *** aim of this paper is to analyse the effects of the introduction of two possible alternatives as a way of energy storage:pumped hydro storage and electric *** this,we use a simulation model adapted to the features of La Palma,considering different *** show that,in the most favourable scenario,the installation of an additional 25 MW from renewables(more than double the current power),supported by 20 MW of pumped hydro storage and a fleet of 3361 electric vehicles,would allow the current share of renewables to increase from11%(in 2015)to 49%.Furthermore,this would mean a 26%reduction in CO2 emissions,10%in costs of generated k Wh and 19%in energy dependence.

Winkler Support Model and Nonlinear Boundary Conditions Applied to 3D Elastic Contact Problem Using the Boundary Element Method

This work presents a numerical methodology for modeling the Winkler supports and nonlinear conditions by proposing new boundary conditions. For the boundary conditions of Winkler support model, the surface tractions and the displacements normal to the surface of the solid are unknown, but their relationship is known by means of the ballast coefficient, whereas for nonlinear boundary conditions, the displacements normal to the boundary of the solid are zero in the positive direction but are allowed in the negative direction. In those zones, detachments of nodes might appear, leading to a nonlinearity, because the number of nodes that remain fixed or of the detached ones (under tensile tractions) is unknown. The proposed methodology is applied to the 3D elastic receding contact problem using the boundary element method. The surface t r actions and the displacements of the common int erface bet ween the two solids in contac t under the influence of different supports are calculated as well as the boundary zone of the solid where the new boundary conditions are applied. The problem is solved by a double-iterative met hod, so in the final solut ion, t here are no t r act ions or pene trations between the two solids or at the boundary of the solid where the nonlinear boundary conditions are Simula ted. The effectiveness of the proposed method is verified by examples.
