Preprotein Import into Chloroplasts via the Toc and Tic Complexes Is Regulated by Redox Signals in Pisum sativum

The import of nuclear-encoded preproteins is necessary to maintain chloroplast function. The recognition and transfer of most precursor proteins across the chloroplast envelopes are facilitated by two membrane-inserted protein complexes, the translocons of the chloroplast outer and inner envelope （Toc and Tic complexes, respectively）. Several signals have been invoked to regulate the import of preproteins. In our study, we were interested in redox-based import regulation mediated by two signals： regulation based on thiols and on the metabolic NADP＋/NADPH ratio. We sought to identify the proteins participating in the regulation of these transport pathways and to characterize the preprotein subgroups whose import is redox-dependent. Our results provide evidence that the formation and reduction of disulfide bridges in the Toc receptors and Toc translocation channel have a strong influence on import yield of all tested preproteins that depend on the Toc complex for translocation. Furthermore, the metabolic NADP＋/NADPH ratio influences not only the composition of the Tic complex, but also the import efficiency of most, but not all, preproteins tested. Thus, several Tic subcomplexes appear to participate in the translocation of different preprotein subgroups, and the redox-active compo- nents of these complexes likely play a role in regulating transport.

Application of wave equation theory to improve dynamic cone penetration test for shallow soil characterisation

Among the geotechnical in situ tests,the dynamic penetration test(DPT)is commonly used around the ***,DPT remains a rough technique and provides only one failure parameter:blow count or cone *** paper presents an improvement of the dynamic cone penetration test(DCPT)for soil characterisation based on the wave equation *** on an instrumented lightweight dynamic penetrometer driving with variable energy,the main process of the test involves the separation and reconstruction of the waves propagating in the rods after each blow and provides a dynamic cone load-penetration(DCLT)*** analytical methodology is used to analyse this curve and to estimate additional strength and deformation parameters of the soil:dynamic and pseudo-static cone resistances,deformation modulus and wave *** carried out in the laboratory on different specimens(wood,concrete,sand and clay)in an experimental sand pit and in the field demonstrated that the resulting DCLT curve is reproducible,sensitive and reliable to the test conditions(rod length,driving energy,etc.)as well as to the soil properties(nature,density,etc.).Obtained results also showed that the method based on shock polar analysis makes it possible to evaluate mechanical impedance and wave velocity of soils,as demonstrated by the comparisons with cone penetration test(CPT)and shear wave velocity measurements made in the *** technique improves the method and interpretation of DPT and provides reliable data for shallow foundation design.

Ultra-sensitive bioanalysis of the therapeutic peptide exenatide for accurate pharmacokinetic analyses at effective plasma concentrations utilizing UPLC-MS/MS

Exenatide is the first approved glucagon-like peptide 1 receptor agonist subcutaneously or intramuscularly inj.cted for the treatment of type 2 diabetes *** therapeutic plasma concentrations are in the low pg/mL range,therefore requiring ultra-sensitive *** enable the accurate evaluation of pharmacokinetic studies,we established a UPLC-MS/MS assay with a lower limit of quantification(LLOQ)of 5 pg/mL(1.2 pM)using 200μL of plasma,validated acco rding to FDA's and EMA's pertinent *** was isolated from plasma with solid phase extraction utilizing anionexchange *** was performed with positive electrospray ionization tandem mass spectrometry in the selected reaction monitoring *** calibrated concentration range of 5-10,000 pg/mL was linear showing co rrelation coefficients>*** and intraday accuracy ranged from 97.5%to 105.4%with corre sponding precision of<10.9%.Accuracy at the LLOQ ranged from 93.0%to 102.5%with corresponding precision of<15.9%.Because of the validity of a 10-fold dilution QC(accuracy 111.2%),the assay is suitable for exenatide quantification up to 100,000 pg/*** ultra-sensitive assay's applicability was demonstrated by the quantification of exenatide plasma concentrations and pharmacokinetics after intravenous and nasal administration to beagle dogs.

Liver surgery in cirrhosis and portal hypertension

The prevalence of hepatic cirrhosis in Europe and the United States, currently 250 patients per 100000 inhabitants, is steadily increasing. Thus, we observe a significant increase in patients with cirrhosis and portal hypertension needing liver resections for primary ormetastatic lesions. However, extended liver resections in patients with underlying hepatic cirrhosis and portal hypertension still represent a medical challenge in regard to perioperative morbidity, surgical management and postoperative outcome. The Barcelona Clinic Liver Cancer classification recommends to restrict curative liver resections for hepatocellular carcinoma in cirrhotic patients to early tumor stages in patients with Child A cirrhosis not showing portal hypertension. However, during the last two decades, relevant improvements in preoperative diagnostic, perioperative hepatologic and intensive care management as well as in surgical techniques during hepatic resections have rendered even extended liver resections in higher-degree cirrhotic patients with portal hypertension possible. However, there are few standard indications for hepatic resections in cirrhotic patients and risk stratifications have to be performed in an interdisciplinary setting for each individual patient. We here review the indications, the preoperative risk-stratifications, the morbidity and the mortality of extended resections for primary and metastatic lesions in cirrhotic livers. Furthermore, we provide a review of literature on perioperative management in cirrhotic patients needing extrahepatic abdominal surgery and an overview of surgical options in the treatment of hepatic cirrhosis.

Cuticular hydrocarbon profiles differ between ant body parts:implications for communication and our understanding of CHC diffusion

Insect cuticular hydrocarbons(CHCs)serve as communication signals and protect against *** form complex blends of up to 150 different *** to differences in molecular packing,CHC classes differ in melting *** is especially important in social insects like ants,which use CHCs to communicate within the colony and to recognize *** recognition models often assume a homogenous colony odor,where CHCs are collected,mixed,and redistributed in the postpharyngeal gland(PPG).Via diffusion,recognition cues should evenly spread over the body ***,CHC composition should be similar across body parts and in the *** test this,we compared CHC composition among whole-body extracts,PPG,legs,thorax,and gaster,across 17 ant species from 3 *** CHC composition differed between body parts,with consistent patterns across species and CHC ***-melting CHC classes were most abundant in the *** contrast,whole body,gaster,thorax,and legs had increasing proportions of CHC classes with higher melting *** CHC variation was highest for rather solid,late-melting CHC classes,suggesting that CHCs differ in their diffusion rates across the body *** results show that body parts strongly differ in CHC composition,either being rich in rather solid,late-melting,or rather liquid,early-melting *** implies that recognition cues are not homogenously present across the insect ***,the unequal diffusion of different CHCs represents a biophysical mechanism that enables caste differences despite continuous CHC exchange among colony members.

Effect of nanoparticle concentration on zeta-potential measurement results and reproducibility

The effect of nanoparticle concentration on zeta-potential measurement results at dilute concentrations was evaluated. The values of the zeta-potential for four different types of nanoparticles, Ludox （silica）, multi-walled carbon nanotubes （bamboo-shaped and hollow nanotubes） and gold, at various concentrations, were obtained using a laser Doppler electrophoresis instrument. The size of the nanoparticles on dilution was measured using dynamic light scattering （DLS）. The results show that there is a concentration range within which the zeta-potential, and particle size, are not affected by nanoparticle concentration. The lower concentration limit for the system to produce consistent results was dependent on the nature of the sample under study and ranged between 10-2 and 10-4 wt%. Below this concentration, there was an apparent shift in zeta-potential values to less negative values, which was accompanied by an increase in the particle size. The shift in zeta-potential was attributed to an increase in contribution of the signal from extraneous particulate matter. The increase in particle size was attributed to the nature of the homodyne optical configuration of the instrument. The aim of this study was to elucidate the range in nanoparticle concentration that allows for accurate and reliable measurement of the zeta-potential and DLS data.

The ever increasing availability of supercomputing resources led computer-based materials science into a new era of highthroughput ***,Pizzi et *** the AiiDA framework that provides a way to automate calculations while allowing to store the full provenance of complex workflows in a *** present the development of the AiiDA-KKR plugin that allows to perform a large number of ab initio impurity embedding calculations based on the relativistic full-potential Korringa-Kohn-Rostoker Green function *** capabilities of the AiiDA-KKR plugin are demonstrated with the calculation of several thousand impurities embedded into the prototypical topological insulator *** results are collected in the j.DiT database which we use to investigate chemical trends as well as Fermi level and layer dependence of physical properties of *** includes the study of spin moments,the impurity’s tendency to form in-gap states or its effect on the charge doping of the *** properties depend on the detailed electronic structure of the impurity embedded into the host crystal which highlights the need for ab initio calculations in order to get accurate predictions.

Background and purpose Retinal pathologies are an independent risk factor for ischaemic stroke,but research on the predictive value of retinal abnormalities for recurrent vascular events in patients with prior stroke is *** investigated the association of retinal pathologies with subsequent vascular *** In a substudy of the Intensified secondary prevention intending a reduction of recurrent events in TIA and minor stroke patients(INSPiRE-TMS)trial,we enrolled patients with recent transient ischaemic attack(TIA)or minor stroke with at least one modifiable risk *** outcome was the composite of subsequent vascular *** photographs were taken at baseline and categorised into three different fundus groups by a telemedically linked *** 722 patients participated in the current study and 109 maj.r vascular events *** multivariable adj.stments,we did not find a significant association between fundus categories and risk for subsequent vascular events(HRs for moderate vascular retinopathy and vascular retinopathy with vessel rarefaction in comparison to no vascular retinopathy 1.03(95%CI 0.64 to 1.67),p=0.905 and 1.17(95%CI 0.62 to 2.20),p=0.626).In a selective post hoc analysis in patients with diabetes mellitus and hypertension,patients with vascular retinopathy with vessel rarefaction had a higher risk for recurrent stroke(HR 24.14(95%CI 2.74 to 212.50),p=0.004).Conclusions Retinal changes did not predict maj.r subsequent vascular events in patients with recent TIA or minor *** studies are needed to examine the utility of fundus photography in assessing the risk of stroke recurrence in patients with diabetes mellitus and hypertension.

Early Identification of Stable Transformation Events by Combined Use of Antibiotic Selection and Vital Detection of Green Fluorescent Protein (GFP) in Carrot (Daucus carota L.) Callus

Genetic transformation is a useful technique to complement conventional breeding in crop improvement. Although carrot has been a model organism for in vitro embryogenesis study, genetic transformation of carrot is still lengthy and labor intensive. An efficient transformation and detection system is desirable. Direct infection of Agrobacterium to carrot calli has provided an easy way for carrot genetic transformation. To improve the efficiency of antibiotic selection in this method, we report the combined use of an improved green-fluorescent protein, referred to as smGFP, to establish a versatile selection method for carrot callus transformation system. By combining antibiotic selection with the bright fluorescence observed in the callus tissue, we were able to easily identify stable transformants in early stage of the transformation process. In addition to the GFP expression of the callus cells, the transgenic nature of callus cells was confirmed with Southern and Western analysis. We found we can link the simplicity of carrot-callus-cell transformation, early detection of stable transformants with antibiotic selection, visualization of GFP fluorescence, and molecular analysis （Southern and Western） of callus tissue （non-photosynthetic tissue） to provide a more efficient way in identifying stable transformants at early stage of carrot transformation.
