In vivo and ex vivo confocal endomicroscopy of pancreatic cystic lesions: A prospective study

To investigate the reproducibility of the in vivo endoscopic ultrasound (EUS) - guided needle based confocal endomicroscopy (nCLE) image patterns in an ex vivo setting and compare these to surgical histopathology for characterizing pancreatic cystic lesions (PCLs).mETHODSIn a prospective study evaluating EUS-nCLE for evaluation of PCLs, 10 subj.cts underwent an in vivo nCLE (AQ-Flex nCLE miniprobe; Cellvizio, maunaKea, Paris, France) during EUS and ex vivo probe based CLE (pCLE) of the PCL (Gastroflex ultrahigh definition probe, Cellvizio) after surgical resection. Biopsies were obtained from ex vivo CLE-imaged areas for comparative histopathology. All subj.cts received intravenous fluorescein prior to EUS and pancreatic surgery for in vivo and ex vivo CLE imaging *** total of 10 subj.cts (mean age 53 ± 12 years; 5 female) with a mean PCL size of 34.8 ± 14.3 mm were enrolled. Surgical histopathology confirmed 2 intraductal papillary mucinous neoplasms (IPmNs), 3 mucinous cystic neoplasms (mCNs), 2 cystic neuroendocrine tumors (cystic-NETs), 1 serous cystadenoma (SCA), and 2 squamous lined PCLs. Characteristic in vivo nCLE image patterns included papillary proj.ctions for IPmNs, horizon-type epithelial bands for mCNs, nests and trabeculae of cells for cystic-NETs, and a “fern pattern” of vascularity for SCA. Identical image patterns were observed during ex vivo pCLE imaging of the surgically resected PCLs. Both in vivo and ex vivo CLE imaging findings correlated with surgical *** vivo nCLE patterns are reproducible in ex vivo pCLE for all maj.r neoplastic PCLs. These findings add further support the application of EUS-nCLE as an imaging biomarker in the diagnosis of PCLs.

＜正＞Non-melanoma skin cancer （NmSC） is the most common human cancer worldwide. In Australia, NmSC is three times as common as all other cancer types combined. NmSC includes solar keratosis （SK）, a benign tumour, which can progress into the malignant squamous cell carcinoma （SCC）. The greatest risk factor for the development of SCC is exposure to ultraviolet radiation （UVR）;an early event in skin cancer development is UVR-induced mutations in p53. Little is known as to whether additional mutations to other molecules are necessary to cause SK to progress to SCC. Brm （Brahma） might be implicated in the progression of SK to SCC. Our group found via microarray analyses that Brm expression was significantly reduced in SCC relative to SK, suggesting a role for the molecule as a tumour suppressor. Brm is the core ATPase-containing component of the SWI/SNF chromatin-remodelling complex. Loss of Brm has been identified in -10% of primary lung adenocarcinomas, whilst knockout of Brm in mice leads to abnormalities in cell growth. Our aim is to determine the role of Brm in skin cancer progression via the manipulation of Brm expression in immortalised human keratinocytes （HaCaT cells）. This will be achieved using two methods: RNA-interference will be used to knock-down Brm mRNA expression in HaCaT cells, whilst a dominant-negative mutant of Brm will be used to competitively inhibit the function of wild-type Brm. Effects of Brm manipulation will be assayed using engineered human skin and Boyden chamber assays. We hypothesise that reduced functionality of Brm will lead to abnormal differentiation and stratification of the engineered epidermis, as well as enhancing invasion of the basement membrane.

＜正＞Background/Aims: The porphyrin drug Photofrin is used in photodynamic therapy applications to clinically treat oral and upper aerodigestive tract tumors;however, this drug is limited by its inadequate tumor selectivity and poor optical properties-common problems for members of the porphyrin family. We are currently studying a class of porphyrin derivatives, the porphyrazines （pzs）, which possess superior optical properties and greater synthetic flexibility than the porphyrins and show promise as both PDT agents and traditional anti - tumor agents. Previous work in our laboratory has shown that a subfamily of acid - functionalized pzs exhibit selective anti - tumor activity in A549 human lung adenocarcinoma cells. We here expand this study to test these same pzs in a number of head and neck squamous cell carcinoma （SCC） cell lines. methods: mTT proliferation/viability assays were used to test the toxicity of three H2[pz（ An;B4-n） ] pzs, where n = 4, 3, or 2 （ in a trans conformation）, A is [S（CH2）3COOH], and B is a fused β,β’ - diisopropyloxybenzo ring. Five SCC cell lines were exposed to the pzs at concentrations of 12.5 - 50 μm, for periods of 24, 48, and 72 hours. Results: As the number of terminal acid groups on the pzs was increased, the amount of toxicity observed in the SCC cell lines upon 72 hour exposure also increased-a trend which was also observed previously in the A549 adenocarcinoma cell line. Additionally, at any given concentration, each compound was found to kill a greater percentage of tumor cells than normal cells. Normal cells were found to be unaffected by the H2A2B2 pz （four acid groups） at concentrations of 25 μm and below, whereas all five SCC cell lines exhibited toxicity in the presence of each of the three pzs at all concentrations studied （12.5 - 50 μm）. Conclusion: The acid - appended pzs were found to exhibit the same structure - function relationship in both adeno-carcinomas and squamous cell carcinomas. The selective tumor toxicity

Biochar(BC)has gained attention for removal of toxic elements(TEs)from aqueous media;however,pristine biochar often exhibits low adsorption ***,various modification strategies in BC have been developed to improve its removal capability against *** zero-valent iron(nZVI)and iron oxides(FeOx)have been used as sorbents for TE ***,these materials are prone to agglomeration and also expensive,which make their usage limited for large-scale *** nZVI technical demerits could be resolved by the development of BC-based composite sorbents through the loading of nZVI or FeOx onto BC *** zero-valent iron modified BC(nZVIBC),FeOx-modified BC(FeOxBC)have attracted attention for their capability in removing pollutants from the aqueous ***,a potential use of nZVIBC and FeOxBC for TE removal from aqueous environments has not been well-realized or *** such,this article reviews:(i)the preparation and characterization of nZVIBC and FeOxBC;(ii)the capacity of nZVIBC and FeOxBC for TE retention in line with their physicochemical properties,and(iii)TE removal mechanisms by nZVIBC and *** nZVI and FeOx in BC increases its sporptive capability of TEs due to surface modifications in morphology,functional groups,and elemental *** combined effects of BC and nZVI,FeOx or Fe salts on the sorption of TEs are complex because they are very specific to *** review identified significant opportunities for research and technol-ogy advancement of nZVIBC and FeOxBC as novel and effective sorbents for the remediation of TEs contaminated water.
