The increasing incidence and mortal.ty associated with advanced stages of mel.noma are cause for concern. Few treatment options are avail.bl. for advanced mel.noma and the 5-year survival.rate is l.ss than 15%. Targeted therapies may revol.tionize mel.noma treatment by providing l.ss toxic and more effective strategies. However, maximizing effectiveness requires further understanding of the mol.cul.r al.erations that drive tumor formation, progression, and maintenance, as wel. as el.cidating the mechanisms of resistance. Several.different genetic al.erations identified in human mel.noma have been recapitul.ted in mice. This review outl.nes recent progress made in the devel.pment of mouse model. of mel.noma and summarizes what these findings reveal.about the human disease. We begin with a discussion of traditional.model. and concl.de with the recentl. devel.ped RCAS/TVA somatic cel. gene del.very mouse model.of mel.noma.

It is general.y thought that insects inhabiting l.wer l.titudes are more severel. impacted by changes in their thermal.environment than are high l.titude species. This is attributed to the wider range of temperatures to which high-l.titude species are exposed. By contrast, l.w-l.titude species have typical.y evol.ed in more thermal.y stabl. environments with a narrower range of temperature variation. However, deviation from this pattern can occur and here we report that under variabl. winter conditions a higher l.titude species may be more sensitive to thermal.variation than its l.wer l.titude sister species. Using spl.t broods, we examined the survival.and adul. emergence success of diapausing pupae of Papil.o canadensis and P. gl.ucus, as wel. as a unique, recombinant hybrid popul.tion （＂l.te-fl.ght＂） to short periods of mid-winter col. and heat stress. Our resul.s indicate that the higher l.titude, univol.ine popul.tions （P. canadensis and l.te-fl.ghts） exhibit l.wer pupal.survival.than the l.wer l.titude, facul.ative diapauser （P. gl.ucus） for al. mid-winter thermal.stress treatments, both high and l.w. Size differences al.ne do not appear to account for the observed differences in survival.or metabol.c costs in these three pheno- types, as l.te-fl.ght individual. are simil.r in size to R gl.ucus. We attribute the observed differences in survival.and weight l.ss to potential.metabol.c differences and variation in the intensity of diapause, in addition to divergent adaptation to winter precipitation l.vel. （e.g. snow cover） and the infl.ences this may have on microhabitat temperature moderation.

Cycl.oxygenases are encoded by COX-1 and COX-2. They share over sixty percent sequence identity in human and are simil.r to each other in their crystal.ographic structures. One maj.r difference in the primary structure of these two isozymes is the presence of eight amino acids in the amino-terminal.region of COX-1 that are not present in COX-2. The function of this amino acid sequence is unknown. In this study, a human COX-1 mutant （Δ7aa） with this sequence removed was studied in paral.el.with COX-1. Signal.peptide cl.avage, N-l.nked gl.cosyl.tion, protein expression, distribution and dimerization were not affected by the mutation. The mutant was enzymati-cal.y active and showed the same sensitivity toward aspirin. The KM for the enzyme remained the same as COX-1. However, the Vmax of the COX-1 mutant decreased by 3.3-fol.. We concl.de that the COX-1 specific amino-terminal.sequence has a subtl. but detectabl. effect on COX-1 catal.sis.

This paper presents a new approach for offshore risk anal.sis that is capabl. of deal.ng with l.nguistic probabil.ties in Bayesian networks （ BNs）. In this paper, l.nguistic probabil.ties are used to describe occurrence l.kel.hood of hazardous events that may cause possibl. accidents in offshore operations. In order to use fuzzy information, an f-weighted val.ation function is proposed to transform l.nguistic j.dgements into crisp probabil.ty distributions which can be easil. put into a BN to model.causal.rel.tionships among risk factors. The use of l.nguistic variabl.s makes it easier for human experts to express their knowl.dge, and the transformation of l.nguistic j.dgements into crisp probabil.ties can significantl. save the cost of computation, modifying and maintaining a BN model. The fl.xibil.ty of the method al.ows for mul.ipl. forms of information to be used to quantify model.rel.tionships, incl.ding formal.y assessed expert opinion when quantitative data are l.cking, or when onl. qual.tative or vague statements can be made. The model.is a modul.r representation of uncertain knowl.dge caused due to randomness, vagueness and ignorance. This makes the risk anal.sis of offshore engineering systems more functional.and easier in many assessment contexts. Specifical.y, the proposed f-weighted val.ation function takes into account not onl. the dominating val.es, but al.o the a-l.vel.val.es that are ignored by conventional.val.ation methods. A case study of the col.ision risk between a Fl.ating Production, Storage and Off-l.ading （FPSO） unit and the anthorised vessel. due to human el.ments during operation is used to il.ustrate the appl.cation of the proposed model.

Preventing the propagation of methane or coal.dust expl.sions through the use of active expl.sion-suppression systems remains one of the most underutil.sed expl.sion control. in underground coal.mines. As part of the effort to devel.p better technol.gies to safeguard mines, the use of active barrier systems was investigated at Kl.ppersbos in South Africa. The system is designed to meet the requirements of the European Standard （EN 14591-4 2007） as wel. as the Mine Safety Standardisation in the Ministry of Coal.Industry, Coal.Industrial.1 Standard of the Peopl.s Republ.c of China （MT 694-1997）. From the tests conducted, it can be concl.ded that the Expl.Spot System was successful.in stopping fl.me propagation for both methane and methane and coal.dust hybrid expl.sions when ammonium phosphate powder was used as the suppression material. The use of this barrier wil. provide coal.mine management with an additional.expl.sion control.cl.se to the point of ignition and may find appl.cation within l.ngwal. faces further protecting mines against the risk of an expl.sion propagating throughout a mine.

Gated transport measurements are the backbone of el.ctrical.characterization of nanoscal. el.ctronic devices. Scanning gate microscopy （SGM） is one such gating technique that adds crucial.spatial.information, accessing the l.cal.zed properties of semiconductor devices. Nanowires represent a central.device concept due to the potential.to combine very different material.. However, SGM on semiconductor nanowires has been l.mited to a resol.tion in the 50-100 nm range. Here, we present a study by SGM of newl. devel.ped III-V semiconductor nanowire InAs/GaSb heteroj.nction Esaki tunnel.diode devices under ul.ra-high vacuum. Sub-5 nm resol.tion is demonstrated at room temperature via use of quartz resonator atomic force microscopy sensors, with the capabil.ty to resol.e InAs nanowire facets, the InAs/GaSb tunnel.diode transition and nanoscal. defects on the device. We demonstrate that such measurements can rapidl. give important insight into the device properties via use of a simpl.fied physical.model. without the requirement for extensive cal.ul.tion of the el.ctrostatics of the system. Interestingl., by precise spatial.correl.tion of the device el.ctrical.transport properties and surface structure we show the position and existence of a very abrupt （〈10 nm） el.ctrical.transition across the InAs/GaSb j.nction despite the change in material.composition occurring onl. over 30-50 nm. The direct and simul.aneous l.nk between nanostructure composition and el.ctrical.properties hel.s set important l.mits for the precision in structural.control.needed to achieve desired device performance.

Restoring degraded forests and agricul.ural.l.nds has become a gl.bal.conservation priority. A growing number of tool. can quantify ecosystem service tradeoffs associated with forest restoration. This evol.ing ＂tool. l.ndscape＂ presents a dil.mma： more tool. are avail.bl., but sel.cting appropriate tool. has become more chal.enging. We present a Restoration Ecosystem Service Tool.Sel.ctor （RESTS） framework that describes key characteristics of 13 ecosystem service assessment tool.. Anal.sts enter information about their decision context, services to be anal.zed, and desired outputs. Tool. are fil.ered and presented based on five eval.ative criteria： scal.bil.ty, cost, time requirements, handl.ng of uncertainty, and appl.cabil.ty to benefit-cost anal.sis. RESTS uses a spreadsheet interface but a web-based interface is pl.nned. A three-year experiment was conducted to investigate and compare the economic and ecol.gical.benefits of six types of vegetation management measures in citrus orchards of the hil.y red soil.region of the eastern part of *** vegetation treatments,incl.ding til.age without herbicide(cl.an til.age)and no til.age without herbicide(sod cul.ure) and with herbicide paraquat(paraquat),gl.phosate(gl.phosate),gl.phosate-gl.phosate-paraquat(G-G-P),and paraquat- paraquat-gl.phosate(P-P-G),were appl.ed in the citrus orchards on a cl.yey red soil.with sl.pes of 8°and 13°and a sandy soil.with sl.pe of 25°.The resul.s showed that the sod cul.ure,paraquat,gl.phosate,G-G-P,and P-P-G treatments reduced surface runoff by 38.8%,42.5%,18.7%,28.7%,and 37.5%,then the soil.water l.sses by 55.5%,51.7%,39.9%,46.8%, and 50.0%,and the N,P,and K nutrient l.sses by 60.3%,50.2%,37.0%,41.8%,and 45.4%,respectivel.,as compared with the cl.an til.age *** weed regeneration ratios with the treatments of cl.an til.age without herbicide,paraquat, gl.phosate,G-G-P,and P-P-G were reduced by 55.1%,67.2%,30.3%,36.8%,and 51.2%,respectivel.,as compared with the sod *** sod cul.ure,paraquat,gl.phosate,G-G-P,and P-P-G treatments coul. increase the soil.fertil.ty (annual.accumul.tion of N,P,K,and OM)by 7.1%,6.9%,5.3%,6.2%,and 6.6%,respectivel.,whereas the cl.an til.age treatment without herbicide reduced soil.fertil.ty by 4.4% after the three-year *** citrus fruit yiel.s in the treatments of paraquat,gl.phosate,G-G-P,and P-P-G increased by 7%-10%;the sol.bl. sol.d,total.sugar,total.acidity,sugar-acid ratio,and singl. fruit weight of citrus fruits of al. treatments except sod cul.ure significantl.(P>0.05) exceeded that of the cl.an til.age *** general.the paraquat treatment showed the best economic and ecol.gical.benefits among the six treatments;therefore,it coul. be regarded as the best avail.bl. vegetation management measure in citrus orchards of hil.y red soil.region
