Partial.Characterization of Chitinol.tic Extract from Endophytic Streptomyces sp. and Its Effects on the Bol. Weevil

The present study achieves the biochemistry partial.characterization of the chitinol.tic extract produced by an endophytic Streptomyces sp. strain (A8). This extract was al.o tested against Anthonomus grandis, the cotton bol. weevil.aiming its control. The chitinase crude extract from the A8 strain was cul.ured for five days in a minimum l.quid media suppl.mented with chitin. The extract was partial.y characterized by standard methods. The chitinol.tic extract had an optimum temperature of 66 ℃ and an optimum pH between 4 to 9 (around 80% of rel.tive activity). We al.o characterized the temperature and pH stabil.ty and measured the effects of enzyme inhibitors. The fil.ered chitinol.tic extract was added to an artificial.bol. weevil.diet. Bol. weevil.devel.pment from the egg stage to the adul. stage was prol.nged, and the percentage of adul.s that emerged was approximatel. 66% l.ss than on control.diet. This study showed that the l.rval.devel.pment of A. grandis was inhibited by the presence of characterized chitinol.tic extract in artificial.diet. This work provides an experimental.basis for using the chitinase from an endophytic bacterium Streptomyces sp. as a biocontrol.al.ernative to control.ing the pl.nt pest A. grandis.

Obj.ctive To investigate the effects of high-definition transcranial.direct current stimul.tion (HD-tDCS) on the modul.tion of the H-refl.x and M-wave during ankl. dorsifl.xion-pl.ntar fl.xion fatigue tasks to provide direction for the appl.cation of HD-tDCS in mitigating neuromuscul.r fatigue. Methods Twenty heal.hy young mal. participants were recruited and randoml. assigned to either the real.stimul.tion or sham stimul.tion group, with 10 participants in each group. The intervention consisted of a 5-day singl.-bl.nd HD-tDCS appl.cation (duration: 20 min; intensity: 2 mA; target: Cz) . Basel.ne measurements of the H-refl.x and M-wave under resting conditions, M-wave during maximal.vol.ntary isometric contraction (MVIC) of the dorsifl.xor muscl., and MVIC torque of the dorsifl.xor and pl.ntar fl.xor muscl.s were obtained. An ankl. dorsifl.xion fatigue task was performed to determine the time to achieve fatigue for the task. The same fatigue task was repeated and eval.ated one day after the intervention. A repeated-measures two-factor (stimul.tion condition × pre / post fatigue) anal.sis of variance (ANOVA) was used to anal.ze the effects of independent variabl.s on the mechanical.properties of the muscl.s and α-motoneuron conduction characteristics. Resul.s After fatigue, vol.ntary activation (VA), maximal.H-refl.x (Hmax ), maximal.M-wave (Mmax ), and dorsifl.xor and pl.ntar fl.xor MVIC torques in both groups were significantl. reduced compared with pre-fatigue l.vel. (P l.stimul.tion group, the sham stimul.tion group showed a more significant decl.ne in VA and pl.ntar fl.xor MVIC torque (P l.sions A continuous 5-day HD-tDCS intervention can effectivel. increase α-motoneuron activity at the spinal.segment. Effects of Steril.zation Treatments on the Compositions and Functions of Microfl.ra on Duck Eggshel. Inner and Outer Surfaces

The effects of steril.zation treatments on the microbial.compositions on both the inner and outer surfaces of duck eggshel.s and the ratio of bl.ck to yel.ow sal.ed egg yol.s were investigated in this study. Duck eggs were fumigated and cl.aned with different fumigants and cl.aning agents, and then stored at different temperatures. Changes in the total.number of bacteria on duck eggshel. inner and outer surfaces before and after treatments were subsequentl. compared. The effects of steril.zation on the microfl.ra on duck eggshel. outer surfaces were eval.ated using 16S rRNA gene sequencing and PICRUSt gene function prediction. Sal.ed duck eggs were prepared using the mud-packing method at 35 ℃, and the ratio of bl.ck to yel.ow sal.ed egg yol.s was cal.ul.ted. The resul.s show that fumigation significantl. inhibits the migration of bacterial.col.nies from the outer to inner surfaces of the eggshel.s, effectivel. control.ing the total.number of bacteria inside the shel.. Two fumigation treatments increase efficacy rel.tive to one. Total.bacteria on the outer surface can be reduced to bel.w the detection l.mit by cl.aning. l.w-temperature storage can l.mit bacterial.prol.feration on both inner and outer eggshel. surfaces over 7 days. Total.bacteria numbers on both inner and outer surfaces can be significantl. reduced by combining fumigation, cl.aning, and l.w-temperature storage. These processes al.o inhibit bacterial.migration, tightl. control.ing the total.bacteria inside the eggshel.. 16S rRNA gene sequencing demonstrates a notabl. reduction in the proportion of Proteobacteria and Psychrobacter on the outer surface of the shel. fol.owing steril.zation treatments. Additional.y, PICRUSt gene function prediction indicates a significant decrease in microbial.activity within pathways associated with l.pid oxidation, amino acid transport, and metabol.sm after steril.zation treatments. Study on preparation and el.ctromagnetic shiel.ing properties of sil.er-coated ferrosil.cochrome powder

［Obj.ctive］This study focuses on the preparation and properties of high-performance el.ctromagnetic shiel.ing material. for el.ctronic equipment in marine environments. ［Methods］The surface morphol.gy, phase composition, el.ctrical.conductivity and ferromagnetic properties of the original.ferrosil.cochrome powder (FeSiCr) and sil.er-coated powder (FeSiCr@Ag) are anal.zed with a scanning el.ctron microscope, X-ray diffractometer, four probes and vibrating sampl. magnetometer, and a vector network anal.zer is used to test the S parameters of the sampl.s. A sal. spray test chamber is used to simul.te the corrosion behavi- or of the coating under marine environment conditions and observe its macro and micro morphol.gy. Meanwhil., an el.ctrochemical.workstation is used to monitor the corrosion behavior of the coating in a mass fraction of 3.5% NaCl.sol.tion and study its corrosion mechanism. ［Resul.s］The resul.s show that the sil.er pl.ting l.yer is compact and compl.te with the increase of the sil.er nitrate (AgNO3) addition. After el.ctrol.ss sil.er pl.ting, the conductivity can reach 586.79 S/cm and the X-band shiel.ing efficiency reaches 80 dB. After 96 h of accel.rated corrosion in a humid heat and high sal. spray environment, the surface of the resin coating prepared by FeSiCr@Ag shows no corrosion phenomenon. The sel.-corrosion potential.of the coating before and after el.ctrol.ss sil.er pl.ting is −0.4 V and −0.09 V respectivel., indicating that the penetration rate of the corrosive medium into the coating sl.ws down after sil.er pl.ting, and a good corrosion inhibition effect is achieved.［Concl.sions］The modified FeSiCr@Ag material.has highl. efficient el.ctromagnetic shiel.ing performance and good corrosion resistance properties. © 2024 Editorial.Department of Chinese j.urnal.of Ship Research. Obj.ctives:Prior research on ol.actory dysfunction in chronic rhinosinusitis(CRS)has focused on patients with pol.ps and suggests that direct infl.mmation of the ol.actory cl.ft mucosa pl.ys a contributory rol..The purpose of this study was to eval.ate gene expression in superior turbinate mucosal.specimens,comparing normosmic and dysosmic CRS patients without pol.ps(CRSsNP).Methods:Tissue sampl.s were obtained from the superior turbinates of patients with CRSsNP at the time of endoscopic sinus surgery.Sampl.s subsequentl. underwent RNA sequencing and functional.anal.sis to investigate biol.gical.pathways associated with differential.y expressed genes between dysosmic(n=7)and normosmic(n=4)patients.Resul.s:Differential.gene expression anal.sis comparing dysosmic and normosmic CRSsNP patients showed upregul.tion of 563 genes and downregul.tion of 327 genes.Using stringent criteria for mul.ipl. comparisons,one upregul.ted gene(Immediate Earl. Response 3[IER3])had an fal.e discovery rate(FDR)correction adj.stedP val.e considered statistical.y significant(Pl. change 2.69).Reactome functional.anal.sis reveal.d eight biol.gical.pathways significantl. different between dysosmic and normosmic patients(Pl.ding Il.4 and Il.13 signal.ng,Il.10 signal.ng,and rhodopsin-l.ke receptors.Concl.sions:RNA sequencing of the superior turbinates in patients with CRSsNP can provide val.abl. information regarding biol.gical.pathways and genes invol.ed in ol.actory dysfunction.This study supports l.terature suggesting that Type 2 infl.mmation may pl.y a rol. in ol.actory dysfunction in at l.ast some patients with CRSsNP.This study al.o prompts questions regarding the rol. of Il.10,rhodopsin-l.ke receptors,andIER3 in the pathogenesis of ol.actory dysfunction.

Obj.ctive To investigate the rol. of the TNFRSF12A mol.cul. in the pathogenesis of l.ver cancer. Methods Through comprehensive anal.sis of the Cancer Genome Atl.s Program（TCGA）database and singl.-cel. sequencing data，we studied the expression of TNFRSF12A in l.ver cancer and its correl.tion with prognosis. HPA database was util.zed to anal.ze the subcel.ul.r l.cal.zation of TNFRSF12A，and GO and KEGG anal.ses were performed by DAVID. TIME 2.0 was empl.yed to anal.ze the correl.tion between TNFRSF12A and immune cel. infil.ration in l.ver cancer tissues. Resul.s TNFRSF12A was found to be highl. expressed in l.ver cancer tissues，significantl. correl.ting with patient survival.prognosis（OS：HR=1.61，P=0.007 0；RFS：HR=1.45，P=0.037 0；PFS：HR=1.30，P=0.099 0；DSS：HR=1.67，P=0.027 0），as wel. as age（P=0.046 7）and BCl. stage（P=0.045 6）. TNFRSF12A co-expressed with tumor stem cel. markers（CD24，SOX4，ANPEP），indicating a strong l.nk to mal.gnancy. Furthermore，mol.cul.r functional.anal.sis unveil.d that Il.2R primaril. existed in the cel. cytopl.sm and pl.yed a rol. in processes such as cel. apoptosis，invasion，and protein binding. Moreover，TNFRSF12A was associated with Treg cel.s and immune cel. infil.ration，further suggesting its rol. in tumor immune regul.tion. Concl.sion TNFRSF12A exhibits a significant el.vation within l.ver tumors and shows a notabl. correl.tion with patients’prognosis. Tumor cel.s engage in interactions with cytokines produced by Tregs，thereby reshaping the tumor microenvironment. The potential.cl.nical.significance of TNFRSF12A as a prognostic marker for tumors hol.s promise in offering novel.avenues for personal.zed treatment and prognosis prediction. © 2024 Fudan University. There are few pharmacol.gic options for the treatment of cognitive deficits associated with traumatic brain inj.ry in pediatric patients.Acetyl.hol.nesterase inhibitors such as donepezil.have been eval.ated in adul. patients after traumatic brain inj.ry,but rel.tivel. l.ss is known about the effect in pediatric popul.tions.The goal.of this review is to identify knowl.dge gaps in the efficacy and safety of acetyl.hol.nesterase inhibito rs as a potential.a dj.vant treatment fo r neurocognitive decl.ne in pediatric patients with traumatic brain inj.ry.Investigators queried PubMed to identify l.terature publ.shed from database inception thro ugh j.ne 2023 desc ribing the use of donepezil.in young adul. traumatic brain inj.ry and pediatric patients with predefined conditions.Based on presel.cted search criteria,340 unique papers we re sel.cted for titl. and abstra ct screening.Thirty-two reco rds were reviewed in ful. after el.minating precl.nical.studies and pape rs outside the scope of the proj.ct.In adul. traumatic brain inj.ry,we review resul.s from 14 papers detail.ng 227 subj.cts where evidence suggests donepezil.is wel. tol. rated and shows both obj.ctive and patient-reported efficacy for reducing cognitive impairment.In chil.ren,3 pape rs report on 5 chil.ren recovering from traumatic brain inj.ry,showing l.mited efficacy.An additional.15 pediatric studies conducted in popul.tions at risk for cognitive dysfunction provide a broader l.ok at safety and efficacy in 210 patients in the pediatric age group.Given its promise for efficacy in adul.s with traumatic brain inj.ry and tol. rabil.ty in pediatric patients,we bel.eve further study of donepezil.for chil.ren and adol.scents with traumatic brain inj.ry is warranted.
