A Light-Independent Allele of Phytochrome B Faithfully Recapitulates Photomorphogenic Transcriptional Networks

Dominant gain-of-function alleles of Arabidopsis phytochrome B were recently shown to confer lightindependent, constitutive photomorphogenic （cop） phenotypes to transgenic plants （Su and lagarias, 2007）. In the present study, comparative transcription profiling experiments were performed to assess whether the pattern of gene expression regulated by these alleles accurately reflects the process of photomorphogenesis in wild-type Arabidopsis. Whole- genome transcription profiles of dark-grown phyAphyB seedlings expressing the Y^276H mutant of phyB （YHB） revealed that YHB reprograms about 13% of the Arabidopsis transcriptome in a light-independent manner. The YHB-regulated transcriptome proved qualitatively similar to but quantitatively greater than those of wild-type seedlings grown under 15 or 50 μmol m^-2 m^-1 continuous red light （Rc）. Among the 2977 genes statistically significant two-fold （SSTF） regulated by YHB in the absence of light include those encoding components of the photosynthetic apparatus, tetrapyrrole/pigment biosynthetic pathways, and early light-responsive signaling factors. Approximately 80% of genes SSTF regulated by Rc were also YHB-regulated. Expression of a notable subset of 346 YHB-regulated genes proved to be strongly attenuated by Rc, indicating compensating regulation by phyC-E and/or other Rc-dependent processes. Since the maj.rity of these 346 genes are regulated by the circadian clock, these results suggest that phyA- and phyB-independent light signaling pathway（s） strongly influence clock output. Together with the unique plastid morphology of dark-grown YHB seedlings, these analyses indicate that the YHB mutant induces constitutive photomorphogenesis via faithful reconstruction of phyB signaling pathways in a light-independent fashion.

Obj.ctive:To provide an overview of the various treatment options available in the clinic to achieve periorbital rej.venation.Data Sources:Literature review and expert opinion.Conclusions:Periorbital rej.venation in the clinic can be accomplished through a variety of treatment modalities,including topical therapies,skin resurfacing,and fillers and inj.ctables.Furthermore,some surgical approaches,such as upper blepharoplasties,ptosis,and brow lifts,can be performed in the clinic under local anesthesia with only mild oral sedation.However,the successful execution of such procedures depends on proper patient selection and maximizing patient comfort.

Use of topical vancomycin powder in total j.int arthroplasty:Why the current literature is inconsistent?

Periprosthetic j.int infection(Pj.)is a rare but terrible complication in hip and knee arthroplasty,and the use of topical vancomycin powder(VP)has been investigated as a tool to potentially reduce its incidence.However,there remains no consensus on its efficacy.Therefore,the aim of this review is to provide an overview on the application of topical vancomycin in orthopaedic surgery focusing on the recent evidence and results in total j.int arthroplasty.Several systematic reviews and meta-analyses on topical VP in hip and knee arthroplasty have been recently published reporting sometimes conflicting results.Apart from all being limited by the quality of the included studies(mostly level III and IV),confounding variables are often included potentially leading to biased conclusions.If taken into consideration the exclusive use of VP in isolation,the available data,although very limited,suggest that it does not reduce the infection rate in routine primary hip and knee arthroplasty.Therefore,we still cannot advise for a routinary application.A properly powered randomized-controlled trial would be necessary to clarify the role of VP in hip and knee arthroplasty.Based on the analysis of the current evidence,the use of topical VP appears to be safe when used locally in terms of systemic adverse reactions,hence,if proven to be effective,it could bring great benefits due to its low cost and accessibility.

The Middle-Late j.rassic transition period is a critical period for the evolution of terrestrial vertebrates,but the global fossil record from this time is relatively poor.The Shishugou Fauna of this period has recently produced significant fossil remains of dinosaurs and other vertebrate groups,some representing the earliest known members of several dinosaurian groups and other vertebrate groups and some representing the best-known specimens of their group.These discoveries are significant for our understanding of the origin and evolution of several vertebrate lineages.Radiometric dating indicates that the fauna is aged approximately 159-164 Ma.Comparisons with other similarly-aged terrestrial faunas such as Shaximiao and Yanliao show both taxonomic similarities and differences between these faunas and indicate that the j.nggar deposits might have preserved the most complete vertebrate fossil record for a Middle-Late j.rassic Laurasian terrestrial fauna.

Investigation of serum calcium and vitamin D levels in superior semicircular canal dehiscence syndrome: A case control study

Obj.ctive: It remains unknown whether calcium metabolism has any effect on the clinical presentation of superior semicircular canal dehiscence(SSCD). Our aim was to analyse the adj.sted calcium and vitamin D levels in SSCD patients compared to a control group.Methods: This was a prospective case-control study performed in a tertiary referral center, university teaching hospital in the UK. It included all new patients with SSCD seen in a dedicated skull base clinic over a 5-year period(2015-2019) compared to a gender and age matched control group. The main outcome of the study was adj.sted calcium and Vitamin D levels between the two groups.Results: A total of 31 SSCD patients were recruited with a matched number of control patients. The mean Vitamin D level on the SSCD group was 44.8 nmoL/l(SD: 20.8) compared to 47.5 nmoL/l(SD: 27.4) on the control group(p = 0.702). Mean Adj.sted calcium level was 2.34 mmoL/l(SD: 0.7) for SSCD compared to2.41 mmoL/l(SD: 0.11) for controls(p = 0.01), being within normal limits for both the SSCD and the control group.Conclusion: Our study did not identify a link between Vitamin D levels and presence of SSCD. Normal adj.sted calcium values were found in both groups. Despite that a statistically significant lower calcium level was found in the SSCD group which could indicate that suboptimal levels of calcium may affect the micro-environment of the otic capsule at the SSC region.

Tooth loss patterns in older adults with special needs:a Minnesota cohort

This study was conducted to detail tooth loss patterns in older adults with special needs. A total of 491 elderly subj.cts with special needs were retrospectively selected and followed during 10/1999-12/2006. Medical, dental, cognitive, and functional assessments were abstracted from dental records and used to predict risk of tooth loss. Tooth loss events were recorded for subj.cts during follow-up. Chi-squared tests were used to study the association between tooth loss and the selected risk factors. Logistic, poisson, and negative binomial regressions were developed to study tooth loss patterns. Overall, 27% of the subj.cts lost at least one tooth during follow-up. Fourteen subj.cts had tooth loss events per 100 person-years. Tooth loss pattern did not differ significantly among different special-needs subgroups （i.e. community-dwelling vs. long-term care, physically disabled vs. functionally independent）. Special-needs subj.cts with three or more active dental conditions at arrival had more than twice the risk of losing teeth than those without any existing conditions. After adj.sting other factors, the number of carious teeth or retained roots at arrival was a significant predictor of tooth loss for older adults with special needs （P=0.001）. These findings indicate that appropriately managing active caries and associated conditions is important to prevent tooth loss for older adults with special needs.

Full Scale SBR Municipal Wastewater Treatment Facility Utilization of Simultaneous Nitrification/Denitrification Coupled with Traditional Nitrogen Removal to Meet Water Criterion

Simultaneous nitrification denitrification (SND) is a well-established phenomenon in biological nutrient removal activated sludge systems. Study at a municipal wastewater treatment facility sought to determine nitrogen removal effectiveness within a full-scale sequential batch reactor (SBR) system utilizing SND in conj.nction with traditional nitrogen removal. In addition to characterizing extent of SND, the research examined the ability of SND to meet state-based effluent water quality standards. At the selected facility, the average SND efficiency during a two-month sampling period was 52.8%, paralleling results from similar SBR municipal wastewater systems. The observed SBR system had removal efficiencies > 99% for the influent to effluent -N concentrations. The SND process also resulted in average NO3-NO2-N concentration that was 82% lower than the theoretical concentration under comparable circumstances. Overall, nitrogen removal for this SBR system was >99% which typified results in other SND systems, but at a higher Total Nitrogen removal rate.

Physical Characteristics Explain Ball-Carrying Capability in Sub-Elite Rugby Union Players

Purpose The aims of the present study were two-fold:(i)to investigate the relationship between physical characteristics and the game statistics associated with ball-carrying capability amongst sub-elite rugby union players,and(ii)to predict the level of change in these physical characteristics required to improve the associated game statistic via regression analysis.Methods Thirty-eight senior professional players(forwards,n=22;backs,n=16)were assessed for body mass(BM),back squat(BS)single-repetition maximum(1RM)normalised to BM(1RM/BM),10 m sprint velocity(S10),10 m sprint momentum(SM10),and the game statistics from 22 games within the 2019/20 RFU Championship season.The relationship between these measures and the predicted level of change in a physical measure required to improve the total number of the associated game statistic by one were assessed by Pearson’s correlation coefficient and simple regression analyses.Results In forwards,an~11.5%reduction in BM,an~11.8%improvement in BS 1RM/BM,or an~11.5%increase in S10 was required to improve the game statistics associated with ball-carrying capability.In backs,a~19.3%increase in BM or a~15.6%improvement in SM10 was required.Conclusions These findings demonstrate that improvements in lower-body relative strength,acceleration performance,and position-specific alterations in body mass are required to maximise the ball-carrying capability and therefore match outcome of sub-elite rugby union players.

The identification of j.ts originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large Hadron Collider,as Standard Model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced background.This paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS experiment.Two taggers with high performances in rej.cting j.ts from gluon over j.ts from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the j.t,and the other combines several j.t substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the j.t pseudorapidity.Differences in tagging efficiency between data and simulation are provided for j.ts with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.

Assessing the academic achievement of United States orthopaedic departments

BACKGROUND Assessing academic productivity allows academic departments to identify the strengths of their scholarly contribution and provides an opportunity to evaluate areas for improvement.AIM To provide obj.ctive benchmarks for departments seeking to enhance academic productivity and identify those with significant improvement in recent past.METHODS Our study retrospectively analyzed a cohort of orthopaedic faculty at United States-based academic orthopaedic programs.5502 full-time orthopaedic faculty representing 178 programs were included in analysis.Variables included for analysis were National Institutes of Health funding(2014-2018),leadership positions in orthopaedic societies(2018),editorial board positions of top orthopaedic j.urnals(2018),total number of publications and Hirsch-index.A weighted algorithm was used to calculate a cumulative score for each academic program.This study was performed at a large,United States medical school.RESULTS All 178 programs included in analysis were evaluated using the comprehensive weighted algorithm.The five institutions with the highest cumulative score,in decreasing order,were:Washington University in St.Louis,the Hospital for Special Surgery,Sidney Kimmel Medical College(SKMC)at Thomas j.fferson University,the University of California,San Francisco(UCSF)and Massachusetts General Hospital(MGH)/Brigham and Women’s/Harvard.The five institutions with the highest score per capita,in decreasing order,were:Mayo Clinic(Rochester),Washington University in St.Louis,Rush University,Virginia Commonwealth University(VCU)and MGH/Brigham and Women’s/Harvard.The five academic programs that had the largest improvement in cumulative score from 2013 to 2018,in decreasing order,were:VCU,SKMC at Thomas j.fferson University,UCSF,MGH/Brigham and Women’s/Harvard,and Brown University.CONCLUSION This algorithm can provide orthopaedic departments a means to assess academic productivity,monitor progress,and identify areas for improvement as they seek to expand their academic contributions t
