c.nsequenc.s of Non-Uniformity in the Stoic.iometry of c.mponent Frac.ions within One and Two Loops Models of α-Helic.l Peptides

A 3-D elec.rostatic.density map generated using the Wavefront Topology System and Finite Element Method c.early demonstrates the non-uniformity and periodic.ty present in even a single loop of an α-helix. The four dihedral angles (N-c.-c.N, c.-c.N-c., and c.N-c.-c. fully define a helic.l shape independent of its length: the three dihedral angles, φ = -33.5°, ω = 177.3°, and Ψ = -69.4°, generate the prec.se (and identic.l) redundanc. in a one loop (or longer) α-helic.l shape (pitc. = 1.59 /residue;r = 2.25 ). Nevertheless the pattern of dihedral angles within an 11 and a 22-peptide bac.bone atom sequenc. c.nnot be distributed evenly bec.use the stoic.iometry in frac.ion of four atoms never divides evenly into 11 or 22 bac.bone atoms. Thus, three sequential sets of 11 bac.bone atoms in an α-helix will have a disc.etely different c.emic.l formula and c.rrespondingly different c.mbinations of molec.lar forc.s depending upon the assigned starting atom in an 11-step sequenc.. We propose that the unit c.ll of one loop of an α-helix oc.urs in the peptide bac.bone sequenc. c.(N-c.-c.3-N whic. c.ntains an odd number of c. plus even number of amide groups. A two-loop pattern (c.-c.N)7-c. c.ntains an even number of c. atoms plus an odd number of amide groups. Dividing the two-loop pattern into two equal lengths, one frac.ion will have an extra half amide (N-H) and the other frac.ion will have an extra half amide c.O, i.e., the stoic.iometry of eac. half will be different. Also, sinc. the length of N-c.-c.N, c.-c.N-c., and c.N-c.-c.are unequal, the summation of the number of eac. in any frac.ion of n loops of an α-helix in sequenc. will always have unequal length, depending upon the starting atom (N, c., or c..

Purpose: Anxiety disorders are frequently assoc.ated with c.ronic.stress with possible c.gnitive c.nsequenc.s. The aim of this study was to determine the verbal fluenc. performanc. in a c.hort of patients with anxiety disorders and allostatic.load, treated with alprazolam during 12 weeks. Methods: Patients with GAD (general anxiety disorders, DSM IV), with >6 in Hamilton Anxiety Rating Sc.le (HAM-A), neurotic.sm > 18 (NEO-FFI inventory), and normal Mini-Mental State Examination were inc.uded. c.inic.l and Psyc.iatric.examination, Allostatic.Load Index and c.gnitive assessment were analyzed before and after 12 weeks of treatment. The phonemic.and semantic.verbal fluenc. tests were determined in all patients. The sc.ring for eac. fluenc. task was determined by c.unting the number of c.rrec. words. The total sc.re from eac. semantic.and phonemic.verbal task was analyzed c.mparing the individual sc.re with normal data c.ntrolled by age and sex. Patients with sc.res > ?2 standard deviation (SD) from normative data were c.nsidered impaired. Results: Fifty-four patients c.mpleted the semantic.verbal fluenc. test before treatment and fifty patients c.mpleted after treatment. Ac.ording to the z-sc.res before treatment 7 patients of 54 (12.9%) had verbal fluenc. impairments. After treatment none patients showed semantic.verbal fluenc. defic.ts but 3 patients of 50 (6%) showed phonemic.impairments. Impaired group was signific.ntly assoc.ated with an older age before treatment (p = 0.033) and with a similar tendenc. but not signific.nt (p = 0.09) after treatment (Student t test). c.nc.usion: In this study older age fac.or was assoc.ated with verbal fluenc. impairment in GAD patients. Stratified treatments analyzing age and sex fac.ors, inc.uding allostatic.load measurements and c.gnitive assessments may be useful tools to determine the effec.iveness and the safety of psyc.opharmac.logic.l treatments.

Previously we have used Singular Spec.rum Analysis (SSA) to dec.nstruc. the global-mean near-surfac. temperature observations of the Hadley c.ntre—c.imate Researc. Unit that extend from 1850 through 2012. While SSA is a very powerful tool, it is rather like a statistic.l “blac. box” that gives little intuition about its results. Ac.ordingly, here we use the simplest statistic.l tool to provide suc. intuition, the Simple Moving Average (SMA). Firstly we use a 21-year SMA. This reveals a nonlinear trend and an osc.llation of about 60 years' length. Sec.ndly we use a 61-year SMA on the raw observations. This yields a nonlinear trend. We subtrac. this trend from the raw observations and apply a 21-year SMA. This yields a Quasi-periodic.Osc.llation (QPO) with a period and amplitude of about 62.4 years and 0.11°c. This is the QPO we disc.vered in our 1994 Nature paper, whic. has c.me to be c.lled the Atlantic.Multidec.dal Osc.llation. We then subtrac. QPO-1 from the detrended observations and apply an 11-year SMA. This yields QPO-2 with a period and amplitude of about 21.0 years and 0.04°c. We subtrac. QPO-2 from the detrended observations minus QPO-1 and apply a 3-year SMA. This yields QPO-3 with a period and amplitude of about 9.1 years and 0.03°c. QPOs 1, 2 and 3 are suffic.ently regular in period and amplitude that we fit them by sine waves, thereby yielding the above periods and amplitudes. We then subtrac. QPO-3 from the detrended observations minus QPOs 1 and 2. The result is too irregular in period and amplitude to be fit by a sine wave. Ac.ordingly we represent this unpredic.able part of the temperature observations by a Gaussian probability distribution (GPD) with a mean of zero and standard deviation of 0.08°c. The sum of QPOs 1, 2 and 3 plus the GPD c.n be used to proj.c. the natural variability of the global-mean near-surfac. temperature to add to, and be c.mpared with, the c.ntinuing temperature trend c.used predominantly by humanity’s c.ntinuing c.mbustion

The c.talytic.ac.ivity of two c.mmon bac.erial enzymes, lac.ate dehydrogenase （LDH） and c.toc.rome c.oxidase （c.X） from Esc.eric.ia c.li was examined following bac.erial exposure to mic.owave （MW） radiation under well-defined experimental c.nditions. The experiments were c.nduc.ed in a spec.alized mic.owave proc.ssing apparatus, with an exposure frequenc. of 18 GHz, and a temperature profile that was restric.ed to below 40℃ to avoid thermal degradation of the bac.eria. The absorbed power was c.lc.lated to be 1,500 kW/m3 and the elec.ric.field was determined to be 300 Wm. Both values were theoretic.lly c.nfirmed using c.mputer Simulation Tec.nology （c.T） Mic.owave Studio 3D Elec.romagnetic.Stimulation Software. Results showed that the ac.ivity of both enzymes was inc.eased following MW radiation c.mpared to negative c.ntrols and thermally treated samples subj.c.ed to similar temperature profiles. It is suggested that the inc.ease in c.X and LDH enzyme ac.ivity c.uld not be explained by c.nventional heating alone, but was rather a result of mic.o-thermal effec.s that inc.rporated ＇undetec.able＇ thermal mec.anisms.

We have developed a new radiography setup with a short-pulse laser-driven x-ray sourc.. Using a radiography axis perpendic.lar to both long- and short-pulse lasers allowed optimizing the inc.dent angle of the short-pulse laser on the x-ray sourc. target. The setup has been tested with various x-ray sourc. target materials and different laser *** to noise ratios are presented as well as ac.ieved spatial resolutions. The high quality of our tec.nique is illustrated on a plasma flow radiograph obtained during a laboratory astrophysic. experiment on POLARs.
