Biomonitoring the <i>Vitex gardneriana</i>Shauer (Lamiaceae) Toxic Effects to Shed Light on Bioactive Compounds against a Major Coconut Pest Mite

The coconut mite, Aceria guerreronis Keifer (Acari: Eriophyidae), is a major pest of coconut plantations (Cocos nucifera L.) worldwide. Here, we conducted a bioguided phytochemical approach using toxicity and repellency bioassays of nonpolar extract and its fractions of Vitex gardneriana Schauer (Lamiaceae) leaves to this pest. Nonpolar crude extract was fractionated by column chromatography using solvents with increased polarity and binary mixtures, resulting in five semipurified groups. The biomonitoring bioassay provided active fractions and led to the isolation and characterization of the bioactive compound squalene, a biosynthetic precursor of 20-hydroxyecdysone, which plays an important role in plant defense against arthropods. The LC50 of the crude extract of V. gardneriana for A. guerreronis was estimated to be 0.185 mg·mL-1 and LC80 = 4.123 mg·mL-1. Also, the extract was highly repellent to this pest for up to 24 h. The fractions of V. gardneriana, and also squalene, caused mortality to A. guerreronis. The potential of V. gardneriana fractions/squalene as biopesticides for controlling A. guerreronis in coconut plantations is discussed herein.

Semi-Quantitative Histological Analysis of the Effect of Intense Pulsed Light (IPL) and Carbon Dioxide (CO<sub>2</sub>) Intradermic Injection on Fibroblast and Collagen Proliferation in the Skin of Wistar Rats

Background: In recent years, so-called “non-ablative rejuvenation” has been carried out with the use of lasers or intense pulsed light (IPL) to stimulate collagen production by dermal fibroblasts. Intradermal infusion of CO2 stimulates fibroblasts and the synthesis of collagen and elastin, contributing to the retraction of the skin and tissue rejuvenation. Objectives: To evaluate the effects of IPL and the intradermal infusion of CO2 on fibroblast proliferation and collagen in the skin of female rats. Methods: Sixteen adult female Wistar rats were divided into two groups of eight animals. Group 1 underwent IPL and group 2 underwent intradermal CO2 infusion. There was a total of 8 weeks of treatment. We conducted a punch in each animal before any procedure (T0), another punch in the middle of treatment at 4 weeks post-procedure (T1) and a punch at the end of treatment at 8 weeks post-procedure (T2). The cells involved in inflammation, fibrosis and vascularization of the injured tissue by histopathology were analyzed. Results: There was statistically significant fibroblast proliferation and collagen proliferation noted when analyzing all 16 animals together and also when considering the two study groups separately. In both groups, the greatest proliferation of fibroblasts coincided with periods of increased collagen production. Conclusion: Both IPL and intradermal CO2 infusion stimulated fibroblast and collagen proliferation in the skin of the rats studied.

The objective of this work was to evaluate the effect of different water deficiency and rehydration levels on the concentrations of osmoregulators in two plant species（Hymenaea courbaril and H. Stigonocarpa） in the Amazon. We adopted a 2×5×5 factorial system,referring to 2 species（H. courbaril and H. stigonocarpa）and 5 stages of hydration and rehydration. The five hydration and rehydration stages were established in:（1）Control treatment E0;（2） Plants with 13 days of stress after incubation—E13;（3） Plants with 26 days of stress E26;（4）The plants that were established after 26 days after incubation and rehydrated for two days（RD2）;（5） rehydrated for two days（RD4）. The plants that were established after 26 days after incubation and rehydrated for four days. The experiment totaled fifty young plants with five *** measurements were performed at the beginning of the experiment（E0） at 13（E13） and 26（E26） days after the water stress, in which the plants were rehydrated,repeating the analyses after two（RD2） and four（RD4）days. Both species increased the sucrose concentration by18%, with a decrease of 52% in starch content. The RD4 time presented the highest mean starch concentration(0.19 mmol g-1 of the residue for H. courbaril and0.27 mmol g;of residue for H. stigonocarpa). Increased proline concentrations were recorded for controls until RD2 for both species. For glycine betaine, the highest increases in treatments E26 and RD2 were observed for the H. courbaril species. Our rehydration period was not sufficient for total recovery of pre-stress concentrations of all studied solutes.

Levels of Trihalomethanes in Stored Water from High and Fundamental Schools:Comparison between Two Temporal Data Sets

Epidemiological studies have been investigating the relationship between chlorination byproducts exposure and cancer. Studies showed the incidence of colon, rectum and bladder cancer in laboratory animals when halogenated byproducts were administered to them, such as trihalomethanes. Based on this fact, in this work, two data sets of water quality parameters were analyzed with focus on total trihalomethanes (THMt). These two data sets are from two different time periods (one in 2014 and other in 2017). All the samples were collected in the same months, in both data sets. The samples were taken from its same corresponding sampling points in both periods of time. Trihalomethanes (THMs) are undesired byproducts of chlorination and its formation occurs when the chlorine used in water treatment reacts with natural organic matter, which is present in natural waters, during the disinfection process. The aim of this research was to investigate the THMs levels in storage water from the chlorination performed by the Water Treatment Station (WTS) of the Maringá-Parana-Brazil;also, to compare the results obtained with the maximum allowable values (MAVs) established by the Consolidation Resolution n.05/2017, current law of water quality in Brazil. Water samples were collected in eight high and fundamental schools of Maringá-Paraná-Brazil and analyzed through the gas chromatography method by the use of mass spectrometry detector with purge-and-trap concentrator (GC-MS) for THM. Furthermore, parameters such as pH and residual chlorine were analyzed following the methodology proposed by the Standard Methods for the Examination of Water and Wastewater. The study results to THMt show that the maximum value of 21.5 μg/L obtained is within the MAV of 100 μg/L. Chloroform was the compound with higher concentrations in all samples in THMt analysis. In Silico Pharmacokinetics Studies for Quinazolines Proposed as EGFR Inhibitors

In silico pharmacokinetics studies can aid the search for molecules with potential ability to be drug candidates. In this paper, a number of quinazoline candidates for epidermal growth factor receptor inhibitors—EGFR, important targets for the treatment of cancer, are computationally analyzed. The literature described that 69 quinazoline molecules were synthesized and the respective half maximum inhibitory concentrations (IC50) were obtained. A bilinear parabolic model was built to investigate the druglikeness by correlating the corresponding lipophilicities, which can be represented by the ideal Log P , with the optimal biological activity in terms of pIC50 values. Structural characteristics leading to improved pharmacokinetics parameters were then analyzed. Compound 56 exhibited the lowest IC50 and, therefore, it had the highest ability to inhibit the EGFR. In the present work, the most potent inhibitor 56 is not calculated to be the most promising drug candidate, since it’s out of the parabolic model obtained due to a Log P above 5, which is not within the expected optimum range. Finally, this work is an example of computational prediction that an experimentally, highly active EGFR inhibitor can be unsuccessful as drug candidate because of pitfalls in pharmacokinetics parameters.

Clinical Trial demonstrates Efficacy of Transcranial Direct Current Stimulation (tDCS) in Improving Pain Management from Post-Laminectomy Syndrome

Chronic pain, a multidimensional experience affecting individuals’ sensory, cognitive, and emotional aspects, significantly impacts their quality of life. Post-laminectomy syndrome, a condition characterized by persistent back pain following spinal surgery, often leads to disability and increased healthcare utilization. Methods: This randomized, controlled, blind clinical trial aimed to investigate the efficacy of Transcranial Direct Current Stimulation (tDCS) in managing pain from post-laminectomy syndrome in patients. Twenty-four participants were assigned to three groups: sham stimulation, active stimulation over primary motor cortex (M1), or stimulation over dorsolateral prefrontal cortex (DLPFC). Stimulation was administered for five consecutive days, 20 minutes per session, using a current of 1.5 mA through 25 cm2 electrodes. Pain intensity was assessed using Visual Analog Scale (VAS) before, during, and after intervention. Results: An ANOVA model demonstrates significant reduction in pain intensity compared to baseline in VAS, (F(7, 285) = 12.292;p 0.001;Power = 1.000;η2p = 0.534), in tDCS applied to M1, after five days of intervention. After stimulation, a significant improvement was observed in WHOQoL-Bref Quality of life item 1 (p = 0.04), considering statistical significant difference p 0.05. Correlation between the variables: quality of life, depression, anxiety and pain also demonstrates reduction in depression and anxiety according to Beck’s depression and Anxiety Inventories (BDI and BAI), p 0.05. This effect was not observed in DLPFC stimulation group. Patients who believed they received active stimulation, in sham group, demonstrated potential for effective blinding. Conclusion: The tDCS applied to primary motor cortex effectively improved pain management and psychiatry symptoms in post-laminectomy syndrome patients. Variables Associated with Cirrhosis Diagnosis in Patients with Chronic Hepatitis C: A Case-Control Study

The diagnosis of liver cirrhosis in patients with chronic hepatitis C has not always been easy, since the gold standard method is the liver biopsy, which is an invasive procedure with interobserver accuracy problems and there have been reports of complications including records of deaths due to hemoperitoneum. Cirrhosis changes the prognosis of the subject with hepatitis C and requires a different clinical management. This study aimed to identify clinical and laboratory variables associated with the diagnosis of cirrhosis in the ultrasonography of patients infected with hepatitis C. In a case-control study, we evaluated 70 cirrhotic patients with chronic hepatitis C compared to a control group of 70 non-cirrhotic people with positive HCV. The results showed, through logistic regression analysis, that the variables blood donor and professional athlete, adjusted for alcohol consumption, showed OR 0.24 and 0.18, with p values of 0.044 and 0.035, respectively. We conclude that the diagnosis of cirrhosis in patients with chronic hepatitis C remains challenging, but the patients with the condition of blood donor or professional athlete prove to be less likely to cirrhosis in ultrasonography in the initial consultation.

Effect of Growth Regulators in Production and Rooting of Coffea arabica ***

The plants of C. arabica do not naturally produce a large number of orthotropic sprouts, hindering clonal multiplication by cutting, which would be the simplest technique with a lower cost of production compared to other methods of vegetative propagation. An alternative form, used successfully in the propagation of eucalyptus, is propagation by minicutting, which consists of pruning the shoot apex of the plant, forming the ministumps, which in variable time interval emits the shoots that will be used to make minicutting. In C. arabica plants the number of shoots can be increased with the application of growth regulators and fertilizers. So, the purpose of this work was to verify the effect of growth regulators and foliar fertilizers on the emission and development of orthotropic shoots of C. arabica. The experiment was installed in a randomized complete block design, with ten treatments composed by growth regulators and fertilizers: T1 (Control);T2 (Stimulate + Sturdy + Enervig);T3 (Stimulate + Vitakelp + Biozyme);T4 (Tiba + Sturdy + Enervig);T5 (Tiba + Vitakelp + Biozyme);T6 (Brs + Sturdy + Enervig) T7 (Brs + Vitakelp + Biozyme);T8 (Stimulate);T9 (TIBA);T10 (Brs-Brassinoesteroid), each plot consisting of two plants. Biometric and physiological parameters were evaluated for the ministumps, as well as the biometric parameters of the shoots and plant survival. The results showed that there was no influence of the treatments on the biometric parameters of the ministumps. A greater number of orthotopic sprouts were obtained in treatments with the TIBA growth regulator, as well as a greater plant survival of the minicuttings, after 25 days of transplanting in a greenhouse. It was concluded that TIBA treatments influenced the production and quality of clonal minicuttings of C. arabica.

Technological insights:combined impedance manometry for esophageal motility testing-current results and further implications

This review focuses on current aspects of the novel technology of combined impedance manometry for esophageal motility testing. It presents methodological features, summarizes current results and discusses implications for further research. The combined technique assesses simultaneously bolus transport and associated peristalsis, thus allowing detailed analysis of the relationships between bolus transit and esophageal motility. Recent studies demonstrate that combined impedance manometry provides important additional information about esophageal motility as compared to conventional manometry: (1) monitoring of bolus transport patterns, (2) calculation of bolus transit parameters, (3) evaluation of bolus clearance, (4) monitoring of swallow associated events such as air movement and reflux, and (5) investigation of the relationships between bolus transit and LES relaxation. Studies with healthy subjects have identified several useful parameters for comprehensive assessment of eosphageal function. These parameters were found to be pathological in patients with classical achalasia, mild GERD, and ineffective esophageal motility. The technology of combined impedance manometry provides an important new tool for esophageal function testing, advancing both clinical and basic research. However, several important issues remain to be standardized to make the technique suitable for widely clinical use.
