Interplay between nitric oxide and VIP in CCK-8-induced phasic contractile activity in the rabbit sphincter of Oddi

AIM: The sphincter of Oddi (SO) plays an important role in delivery of bile into the duodenum. To establish whether vasoactive intestinal polypeptide (VIP) and nitric oxide (nO) were involved in phasic contractile activity of the rabbit SO stimulated by cholecystokinin-octapeptide (CCK-8). methODS: Isolated SO muscle rings were cleaned of fat and mounted horizontally on two small L-shaped hooks one of which was connected to a force transducer for the measurement of isometric *** experiments were carried out in a thermostatically controlled (37±0.2℃) organ bath (5 mL) containing Krebs *** organ fluid was gassed with 95% O2 and 50 mL/L CO2 to keep the pH at 7.40±0.05. Contractile responses to CCK-8 (1 μmol/L) were evaluated in the presence and absence of nG-nitro-L-arginine (LnnA), an inhibitor of nO synthase (100 μmol/L), and (p-chloro-D-Phe6-Leu17)-VIP (VlPa, 30 μmol/L), a VIP receptor antagonist. RESULTS: CCK-8 stimulated the phasic activity of the SO. nO synthase inhibition increased the frequency and amplitude of contractions with a slight increase in developed tension. Pre-incubation with VlPa also attenuated this CCK-8 effect. The combined application of LnnA and VlPa abolished the phasic activity of the muscle rings with a marked increase in tension in response to CCK-8. COnCLUSIOn: VIP and nO together contribute to an increase in phasic activity of SO.

RESULTS:In the diabetic-pancreatitic rats treatment with insulin and CCK-8 significantly elevated pw/bm and the pancreatic contents of protein,amylase and lipase vs the rats receiving only CCK-8 ***-8 administered in combination with insulin also elevated the number of acinar cells with mitotic activities,whereas CCK-8 alone had no effect on laboratory parameters or the mitotic activities in diabetic-pancreatitic rats. Wettability,reactivity,and interface structure in Mg/ni system

The sessile drop method was applied to the experimental investigation of the wetting and spreading behaviors of liquid Mg drops on pure ni *** comparison,the experiments were performed in two variants:(1)using the Capillary Purification(CP)procedure,which allows the non-contact heating and squeezing of a pure oxide-free Mg drop;(2)by classical Contact Heating(CH)*** high-temperature tests were performed under isothermal conditions(CP:760℃for 30 s;CH:715℃for 300 s)using Ar+5 wt%H_(2) *** the sessile drop tests,images of the Mg/ni couples were recorded by CCD cameras(57 fps),which were then applied to calculate the contact angles of metal/substrate *** and transmission electron microscopy analyses,both coupled with energy-dispersive X-ray spectroscopy,were used for detailed structural characterization of the solidified *** was found that an oxide-free Mg drop obtained by the CP procedure showed a wetting phenomenon on the ni substrate(an average contact angleθnng and good wetting over the ni substrate(θ_((CP))~20°in 5 s)to form a final contact angle ofθ_(f(CP))~18°.In contrast,a different wetting behavior was observed for the CH procedure,where the unavoidable primary oxide film on the Mg surface blocked the spreading of liquid Mg showing apparently non-wetting behavior after 300 s contact at the test ***,in both cases,the deep craters formed in the ni substrates under the Mg drops and significant change in the structure of initially pure Mg drops to Mg-ni alloys suggest a strong dissolution of ni in liquid Mg and apparent values of the final contact angles measured for the Mg/ni system.

Directed x-rays produced in the interaction of sub-picosecond laser pulses of moderate relativistic intensity with plasma of near-critical density are investigated. Synchrotron-like (betatron) radiation occurs in the process of direct laser acceleration (DLA) of electrons in a relativisticlaser channel when the electrons undergo transverse betatron oscillations in self-generated quasi-static electric and magnetic fields. In anexperiment at the PHELIX laser system, high-current directed beams of DLA electrons with a mean energy ten times higher than the ponderomotive potential and maximum energy up to 100 MeV were measured at 10^(19) W/cm^(2)laser intensity. The spectrum of directed x-raysin the range of 5–60 keV was evaluated using two sets of Ross filters placed at 0°and 10°to the laser pulse propagation axis. The differential x-ray absorption method allowed for absolute measurements of the angular-dependent photon fluence. We report 10^(13) photons/sr withenergies >5 keV measured at 0°to the laser axis and a brilliance of 10^(21) photons s^(−1) mm^(−2) mrad−2(0.1%BW)−1. The angular distributionof the emission has an FWHM of 14°–16°. Thanks to the ultra-high photon fluence, point-like radiation source, and ultra-short emissiontime, DLA-based keV backlighters are promising for various applications in high-energy-density research with kilojoule petawatt-class laserfacilities.

Machine fault diagnostics are essential for industrial operations, and advancements in machine learning have significantly advanced these systems by providing accurate predictions and expedited solutions. Machine learning models, especially those utilizing complex algorithms like deep learning, have demonstrated major potential in extracting important information from large operational datasets. Despite their efficiency, machine learning models face challenges, making Explainable AI (XAI) crucial for improving their understandability and fine-tuning. The importance of feature contribution and selection using XAI in the diagnosis of machine faults is examined in this study. The technique is applied to evaluate different machine-learning algorithms. Extreme Gradient Boosting, Support Vector Machine, Gaussian naive Bayes, and Random Forest classifiers are used alongside Logistic Regression (LR) as a baseline model because their efficacy and simplicity are evaluated thoroughly with empirical analysis. The XAI is used as a targeted feature selection technique to select among 29 features of the time and frequency domain. The XAI approach is lightweight, trained with only targeted features, and achieved similar results as the traditional approach. The accuracy without XAI on baseline LR is 79.57%, whereas the approach with XAI on LR is 80.28%. Copyright © 2024 The Authors. Published by Tech Science Press.

A chemo-mechanical model is developed to investigate the effects on the stress development of the coating of polycrystalline ni-rich LinixMnyCo_(z)O_(2)(x≥0.8)(nMC)particles with poly(3,4-ethylenedioxythiophene)(PEDOT).The simulation results show that the coating of primary nMC particles significantly reduces the stress generation by efficiently accommodating the volume change associated with the lithium diffusion,and the coating layer plays roles both as a cushion against the volume change and a channel for the lithium transport,promoting the lithium distribution across the secondary particles more ***,the lower stiffness,higher ionic conductivity,and larger thickness of the coating layer improve the stress *** paper provides a mathematical framework for calculating the chemo-mechanical responses of anisotropic electrode materials and fundamental insights into how the coating of nMC active particles mitigates stress levels.

Development and characterization of 3D-printed electroconductive pHEMA-co-MAA nP-laden hydrogels for tissue engineering

Tissue engineering(TE)continues to be widely explored as a potential solution to meet critical clinical needs for diseased tissue replacement and tissue *** this study,we developed a poly(2-hydroxyethyl methacrylate-co-methacrylic acid)(pHEMA-co-MAA)based hydrogel loaded with newly synthesized conductive poly(3,4-ethylene-dioxythiophene)(PEDOT)and polypyrrole(PPy)nanoparticles(nPs),and subsequently processed these hydrogels into tissue engineered constructs via three-dimensional(3D)*** presence of the nPs was critical as they altered the rheological properties during ***,all samples exhibited suitable shear thinning properties,allowing for the development of an optimized processing window for 3D *** were 3D printed into pre-determined disk-shaped configurations of 2 and 10 mm in height and diameter,*** observed that the nPs disrupted the gel crosslinking efficiencies,leading to shorter degradation times and compressive mechanical properties ranging between 450 and 550 *** conductivity of the printed hydrogels increased along with the nP concentration to(5.10±0.37)×10^(−7)S/*** vitro studies with cortical astrocyte cell cultures demonstrated that exposure to the pHEMA-co-MAA nP hydrogels yielded high cellular viability and proliferation ***,hydrogel antimicrobial studies with staphylococcus epidermidis bacteria revealed that the developed hydrogels affected bacterial *** together,these materials show promise for various TE strategies.
