CYp24A1 inhibition facilitates the anti-tumor effect of vitamin D3 on colorectal cancer cells

AIM:The effects of vitamin D3 have been investigated on various tumors, including colorectal cancer (CRC). 25-hydroxyvitamin-D3-24-hydroxylase (CYp24A1), the enzyme that inactivates the active vitamin D3 metabolite 1,25-dihydroxyvitamin D3 (1,25-D3), is considered to be the main enzyme determining the biological halflife of 1,25-D3. During colorectal carcinogenesis, the expression and concentration of CYp24A1 increases significantly, suggesting that this phenomenon could be responsible for the proposed efficacy of 1,25-D3 in the treatment of CRC. The aim of this study was to investigate the anti-tumor effects of vitamin D3 on the human CRC cell line Caco-2 after inhibition of the cytochrome p450 component of CYp24A1 activity. METHODS:We examined the expression of CYp24A1 mRNA and the effects of 1,25-D3 on the cell line Caco-2 after inhibition of CYp24A1. Cell viability and proliferation were determined by means of sulforhodamine-B staining and bromodeoxyuridine incorporation, respectively, while cytotoxicity was estimated via the lactate dehydrogenase content of the cell culture supernatant. CYp24A1 expression was measured by realtime reverse transcription polymerase chain reaction. A number of tetralone compounds were synthesized to investigate their Cp24A1 inhibitory activity. RESULTS:In response to 1,25-D3, CYp24A1 mRNA expression was enhanced significantly, in a time- and dose-dependent manner. Caco-2 cell viability and proliferation were not influenced by the administration of 1,25-D3 alone, but were markedly reduced by coadministration of 1,25-D3 and kD-35, a CYp24A1-inhibiting tetralone. Our data suggest that the mechanism of action of co-administered kD-35 and 1,25-D3 does not involve a direct cytotoxic effect, but rather the inhibition of cell proliferation. CONCLUSION:These findings demonstrate that the selective inhibition of CYp24A1 by compounds such as kD-35 may be a new approach for enhancement of the anti-tumor effect of 1,25-D3 on CRC.

Start the Earth System Governance that Integrates “Climate-Altering Technologies”

Addressing climate emergencies requires a radical social change, and an “earth system” governance approach that combines different factors(including technologies that affect climate). As the paris Agreement has been reached for four years and came into force for three years, there is a growing recognition that the global average temperature rise cannot be limited to 1.5–2°C only by emissions reduction or existing carbon removal measures. The reason is that the world has not taken enough actions to deal with the crisis. As reported by IpCC(2018), hundreds of millions of people worldwide are already experiencing the harsh consequences of climate change, from storms to floods, to heatwaves and droughts [IpCC. 2018. “International panel on Climate Change Special Report on Global Warming of 1.5 Degrees.” https://***/sr15(accessed November 5, 2019)]. According to Spratt and Dunlop(2019), as the climate change intensifies,all sectors of society have realized the need to avoid the risks brought by climate change and to deal with the severe disasters that already exist [Spratt, David, and Ian Dunlop. 2019.“Existential Climate-related Security Risk: A Scenario Approach.” https://***/ugd/***(accessed November 5, 2019)]. This raises some profound questions. For instances, should people consider a new responsive measure that is compatible with the natural system which sustains life on the earth, and within the limits of the earth’s tolerance? What forms of decision-making might we need, to help us make the smart collective choices needed for a world where no risk-free options remain? Are familiar governance and decision-making processes still suitable for the goals? Who will make the decisions to promote this transformation? If people really want to change the way they make decisions, they may need to create new forms of governance and decide how this transformation begins and which authority is subject to.

Relapsing polychondritis with p-ANCA associated vasculitis: Which triggers the other?

Relapsing polychondritis(Rp) is a rare autoimmune disease with chronic inflammatory/destructive lesions of the cartilaginous tissues. In one third of the cases it is associated with other autoimmune disorders, mostly with anti-neutrophil cytoplasmic antibody(ANCA) associated vasculitis(AAV). We report three cases of Rp with p-ANCA positive AAV. In the first patient Rp developed 1.5 years after the onset of AAV. In the others the signs of Rp were present before the onset of severe crescent glomerulonephritis. patients responded well on steroid and cyclophosphamide. In dialysis dependent cases plasmapheresis was also used successfully. During the 2 and 1.5 years of follow up, they were symptom-free, and had stable glomerular filtration rate. The first patient died after four years of follow-up due to the complications of sudden unset pancytopenia,which raises the possibility of associated hemophagocytic syndrome. In the setting of Rp or AAV physicians should always be aware of the possibility of sudden or insidious appearance of the other disease.

Microbial Biomass C,N and p in Disturbed Dry Tropical Forest Soils,India

Variations in microbial biomass C (MB-C),N (MB-N) and p (MB-p) along a gradient of different dominant vegeta- tion covers (natural forest,mixed deciduous forest,disturbed savanna and grassland ecosystems) in dry tropical soils of Vindhyan plateau,India were studied from january 2005 to December *** water holding capacity,organic C,total N,total p and soil moisture content were comparatively higher in forest soils than in the savanna and grassland *** different study sites the mean annual MB-C,MB-N and MB-p at 0-15 cm soil depth varied from 312.05 ± 4.22 to 653.40 ± 3.17,32.16 ± 6.25 to 75.66 ± 7.21 and 18.94 ± 2.94 to 30.83 ± 23.08 μg g ?1 dry soil,*** all the investigated sites,the maximum MB-C,MB-N and MB-p occurred during the dry period (summer season) and the minimum in wet period (rainy season).In the present study,soil MB-C,MB-N and MB-p were higher at the forest sites compared to savanna and grassland *** differences in MB-C,MB-N and MB-p were significant (p p p p (p positively correlated with organic C,while the relationship between soil moisture and MB-C,MB-N and MB-p (p p p pectively) was *** decreasing order of MB-C,MB-N and MB-p along study ecosystems was natural forest > mixed deciduous forest > savanna > *** results suggested that deforestation and land use practices (conversion of forest into savanna and grassland) caused the alterations in soil properties,which as a consequence,led to reduction in soil nutrients and MB-C,MB-N and MB-p in the soil of disturbed sites (grassland and savanna) compared to undisturbed forest ecosystems.

Severity of gastroesophageal reflux disease influences daytime somnolence: A clinical study of 134 patients underwent upper panendoscopy

AIM: To asses the relationship between severity of gastroesophageal refluxe disease and Epworth sleepiness scale as an indicator of daytime somnolence. METHODS: One hundred and thirty-four patients underwent an upper panendoscopy as indicated by the typical reflux symptoms and were also investigated with regard to somnolence. Sleepiness was evaluated by Epworth Sleepiness Scale, which was compared to the severity of endoscopic findings (savary-Miller/modified by Siewert). patients with psychiatric disorders or being on sedato-hypnotics as well as shift workers were excluded from the study. The relationship between the severity of the reflux disease and daytime somnolence was analyzed with the help of multivariate regression analysis. RESULTS: A positive tendency was found between the severity of the reflux disease and the corresponding Epworth Sleepiness Scale. In the case of the more severe type-savary-Miller Ⅲ- at least a mild hypersomnia was found. For this group daytime somnolence was significantly higher than in the case of the non-erosive type of Gastroesophageal Reflux Disease representing the mildest stage of reflux disease. CONCLUSION: The severity of Gastroesophageal Reflux Disease influences daytime somnolence.

Autoimmune pancreatitis: Functional and morphological recovery after steroid therapy

Autoimmune pancreatitis, a recently recognized type of chronic pancreatitis, is not rare in japan, but reports of it elsewhere are relatively uncommon. We report the first preoperatively diagnosed case of autoimmune pancreatitis in Hungary, which responded well to steroid treatment and provided radiographic and functional evidence of this improvement. A 62-year-old female presented with a 4-month history of recurrent epigastric pain and a 5-kg weight loss. The oral glucose tolerance test （OGTT） indicated diabetes mellitus and the result of the fecal elastase test was abnormal. Ultrasonography （US） and the CT scan demonstrated a diffusely enlarged pancreas, and endoscopic retrograde cholangiopancreatography （ERCp） an irregular main pancreatic duct with long strictures in the head and tail. Autoimmune pancreatitis was diagnosed. The patient was started on 32 mg prednisolone daily, After 4 wk, the OGTT and faecal elastase test results had normalized. The repeated US and CT scan revealed a marked improvement of the diffuse pancreatic swelling, while on repeated ERCp, the main pancreatic duct narrowing was seen to be ameliorated. It is important to be aware of this disease and its diagnosis, because AIp can clinically resemble pancreatobiliary malignancies, or chronic or acute pancreatitis, However, in contrast with chronic pancreatitis, its symptoms and morphologic and laboratory alterations are completely reversed by oral steroid therapy.

Objective:To investigate antidiabetic efficacy of the extract of field grown and in vitro raised leaves of Solanum xanthocarpum(***) against alloxan induced diabetic rats. Methods:The antidiabetic activity of the crude methanol extracts of the field grown and in vitro raised leaves of *** at different concentrations(100-200 mg/kg bw) was tested against alloxan induced diabetic *** antidiabetic efficacy was validated through various biochemical parameters and the antioxidant effect was also *** phytochemical analyses of field grown *** and in vitro rasied *** leaves were done by estimating their chlorophyll,carotenoids,total sugar,protein,amino acid and minerals contents. Results:The results revealed that the methanol extracts of both the leaves(field grown and in vitro raised) of *** was efficient anti hyperglycemic agents at a concentration of 200 mg/kg bw and posses potent antioxidant ***,the extracts of in vitro rasied *** raised leaves exhibit higher efficacy than the field grown leaves in all tested *** composition and mineral analysis of *** revealed higher concentration of contents in in vitro rasied *** than field grown ***. Conclusions:From the results it can be concluded that the leaves extracts of *** can be a potential candidate in treating the hyperglycemic conditions and suits to be an agent to reduce oxidative stress.

Behavior of transporting pipeline sections without and with hydrogen exposure based on full-scale tests

pipeline transport of hydrogen is one of today’s economic and environmental *** order to find safe and reliable application of both existing gas and build new pipelines,it is essential to carry out tests on full-scale pipeline section,including the potentially more dangerous places than the main pipe,the girth *** the investigations,pipeline sections of p355NH steel with girth welds were prepared and exposed to pure hydrogen at twice the maximum allowable operating pressure for 41 ***,full-scale burst tests were carried out and specimens were cut and prepared from the typical locations of the failed pipeline sections for mechanical,and macro-and microstructural *** results obtained were evaluated and compared with data from previous full-scale tests on pipeline sections without hydrogen *** results showed differences in the behavior of pipeline sections loaded in different ways,with different characteristics of the materials and the welded joints,both in the cases without hydrogen exposure and in the cases exposed to hydrogen.
