Brazilian vaccinia virus strains show a classical orthopoxvirus infection course and cross-protection

objectives:The purpose of this work was to study the infection course and cross-protection in mice after intradermal injection of Vaccinia virus(VACV) strain Western Reserve and three Brazilian VACV strains: Aracatuba,Muriae and BeAn58058 isolated from cow,human and rodent,respectively.Methods:Balb/c mice were inoculated by footpad and back scarification and daily monitored regarding lesion development and weight loss.To check cross protection after intradermal VACV inoculation,mice were subsequendy infected with different VACV strains and monitored to check lesion development.Serum neutralization assays were performed to check for the presence of antibodies against orthopoxvirus.Results:After VACV intradermal inoculation the lesion development pattern was similar in mice infected with the different virus strains.By using the footpad scarification model,cross-protection among VACV strains was observed.Moreover,neutralizing antibodies against orthopoxvirus were detected in sera from mice infected with all VACV strains.Conclusion:Although it was not possible to observe virulence differences among VACV strains isolated from cow,rodent and human using the murine model,this inoculation route showed to be an appropriated model to study lesions development since it mimics natural infections by VACV in nature.

Domain adversarial training for classification of cracking in images of concrete surfaces

The development of automatic methods to recognize cracks in surfaces of concrete has been under focus in recent years,firstly through computer vision methods and more recently focusing on convolutional neural networks that are delivering promising results.Challenges are still persisting in crack recognition,namely due to the confusion added by the myriad of elements commonly found on concrete surfaces.The robustness of these methods would deal with these elements if access to correspondingly heterogeneous datasets was possible.Even so,this would be a cumbersome methodology,since training would be needed for each particular case and models would be case dependent.Thus,efforts from the scientific community are focusing on generalizing neural network models to achieve high per-formance in images from different domains,slightly different from those in which they were effectively trained.The generalization of networks can be achieved by domain adaptation techniques at the training stage.Domain adapta-tion enables finding a feature space in which features from both domains are invariant,and thus,classes become separable.The work presented here proposes the DA-Crack method,which is a domain adversarial training method,to generalize a neural network for recognizing cracks in images of concrete surfaces.The domain adversarial method uses a convolutional extractor followed by a classifier and a discriminator,and relies on two datasets:a source labeled dataset and a target unlabeled small dataset.The classifier is responsible for the classification of images randomly chosen,while the discriminator is dedicated to uncovering to which dataset each image belongs.Backpropagation from the discriminator reverses the gradient used to update the extractor.This enables fighting the convergence promoted by the updating backpropagated from the classifier,and thus generalizing the extractor enabling it for crack recognition of images from both source and target datasets.Results show that the DA-Crack training method improved accuracy i

AIM:To compare the performance of three commercially available anti-human epidermalgrowth factor receptor 2(HER2)antibodies in whole-tissue sections and tissue microarrays(TMAs)of a series of gastric tumors.METHoDS:We present a comparative analysis of three anti-HER2 antibodies(HercepTest,4B5 and SP3)using TMA and whole-tissue sections prepared from the same paraffin blocks of 199 gastric adenocarcinomas operated upon between january 2004 and December2008 at a Brazilian cancer hospital.The data on the patients’age,sex,the anatomical location of the tumor and the Lauren’s histological classification were collected from clinical and pathological records.The immunohistochemical(IHC)results were examined by two pathologists and the cases were classified as positive(3+),equivocal(2+)and negative(0 or 1+),according to the criteria of the IHC scoring system of gastric cancer.TMAs and whole-tissue sections were evaluated separately and independently.All cases yielding discordant IHC results and/or scored as 2+were subjected to dual-color in situ hybridization in order to determine the final HER2 status.Besides determining the sensitivity and predictive value for HER2-positive status,we measured the accuracy of each antibody by calculating the area under the receiver operating characteristic(RoC)curve.The agreement between the results obtained using the TMAs and those obtained using the whole-tissue sections was assessed by means of Kappa coefficient.RESULTS:Intratumoral heterogeneity of HER2 expression was observed with all antibodies.HER2-positive expression(3+)in the whole-tissue sections was observed in 23 cases(11.6%)using the 4B5 antibody,in 18 cases(9.1%)using the SP3 antibody and in 10 cases(5.1%)using the HercepTest antibody.In the TMAs,11 positive cases(5.6%)were identified using SP3 antibody,9(4.6%)using the 4B5 antibody and 6(3%)using the HercepTest antibody.The sensitivity using whole-tissue sections and TMA,respectively,was 95.2%and 42.9%with 4B5,90.5%and 66.7%with SP3 and 47.6%and42.9%with HercepTest.The ac

Background: In GERD patients, ineffective esophageal motility(IEM), a hypocont ractile disorder, is the most common motor abnormality. IEM has been associated with reflux in both the supine and upright position, prolonged esophageal cleara nce,and delayed of bolus transport. IEM has been equally present in erosive and in nonerosive GERD. Goal: Considering that reflux has been found to be more seve re in erosive GERD than in nonerosive GERD patients and that IEM delays esophage al clearance, our hypothesis is that patients with erosive GERD have more severe IEM than those with nonerosive disease.Study: A retrospective review of consecu tive manometries of patients with the chief complaint of heartburn and a diagnos is of IEM were performed, and patients with both erosive and nonerosive GERD wer e selected. According to the number of ineffective contractions, IEM was stratif ied into three groups:30%to 40%, mild; 50%to 60%, moderate; and greater than 60%, severe. We also registered the number of low amplitude,failed, and normal waves in each manometry of both groups.We evaluated 110 patients: 70 (64%) wit h erosive GERD and 40 (36%)with nonerosive GERD. The percentage of mild,moderat e and severe IEM was similar in erosive and in nonerosive GERD patients, as well the number of low amplitude, failed or normal waves (Ponclusion: There were no differences between the severity of IEM in erosive and in nonerosive GE RD patients.
