<i>In Vitro</i>Priming of Sugarcane Varieties (RB966928 and RB867515)

Sugarcane, as a glycophyte, shows sensitivity to saline soils at various stages of its growth. The aim of this study was to evaluate the in vitro priming response in two sugarcane varieties (RB966928 and RB867515). Micropropagated plants, from meristems, received priming treatments by adding the salts (NaCl and KCl) in different concentrations (0.0;12.5;25.0 and 50.0 mM), in the MS medium. Subsequently, the plants were cultivated in rooting medium without addition of salts, acclimatized and submitted to gradient ex vitro saline stress with 20 → 40 and 60 mM, of each salt, for 30 days. The analyzed variables were dry matter of shoot and root, number of tillers and estimation of chlorophyll content. The experiment was carried out in a 2 × 2 × 4 factorial arrangement, in a completely randomized experimental design. Twenty replicates were used throughout the experiment. data were submitted to analysis of variance and regression and the means were compared by Tukey’s test, at a 5% probability level. The priming treatments presented a significant effect, with triple interaction, in the chlorophyll index. In the treatment with NaCl, the variety RB966928 showed an increase in the chlorophyll index with the increase of treatment levels, up to an optimal limit of 31.47 mM. On the other hand, the variety RB867515 showed decreasing in chlorophyll index. In contrast, in KCl treatment, the variety RB867515 presented the increase at the chlorophyll index with the maximum point of 25 mM. For the variables, shoot dry matter (SDM) and root dry matter (RDM) there was a significant difference (p 0.05 and p 0.01, respectively) only between the varieties. The variety RB966928 presented higher SDM and RDM in relation to the variety RB867515. Studies are recommended with increasing the duration of the priming treatments and more detailed study of the culture throughout its productive cycle.

Formation of DNA Strand Breaks in Peripheral Lymphocytes of Rats After Exposure to Natural Sunlight

Objective This paper aims to evaluate the genotoxicity in peripheral blood lymphocytes of rats after exposure to sunlight at different time points of day in a tropical region of Brazil（5oS,42oW）.Materials and Methods Thirty Wistar-Hannover rats,three months old,were randomly divided into three groups of 10 animals each：Group I [control,without exposure to ultraviolet（UV） radiation],Group II（exposed to sunlight during 08：00 *** 10：00 a.m.）,and Group III（exposed to sunlight during 10：00 *** 12：00 a.m.）.After a week of exposure,peripheral blood samples were taken from the tail of these animals to prepare smears on two slides per *** 24 h after exposure to sunlight in Group III,a new collection was obtained to observe the repair *** alkaline comet assay was used in this study to evaluate the genotoxic activity of sunlight（P0.05）.Results There was no statistical difference between Group I and II（P=0.672）.On the other hand,the exposure to sunlight in Group III showed genotoxic action in comparison to the other groups（P0.0001）.Also,there was no significant repair in Group III R（P=0.407）.Conclusion This study has shown a genotoxic potential of sunlight（UVA-B） in lymphocytes of mammals from 10：00 *** 12：00 a.m.,due to a higher intensity of UV in this tropical region.

BACKGROUND Toll-like receptor-2(TLR2) is responsible for recognizing Helicobacter pylori(***) and activating the immune response. Polymorphisms in TLR2 may modulate gastric *** To evaluate whether the TLR2 19216 T/C(rs3804099) and TLR2-196 to-174 ins/del(rs111200466) polymorphisms contribute to gastric carcinogenesis in the Brazilian population, and to determine the influence of both polymorphisms and H. pylori infection on TLR2 mRNA *** DNA was extracted from 854 peripheral blood leukocyte or gastric tissue samples[202 gastric cancer(GC), 269 chronic gastritis(CG), and 383 control/healthy(C)]and genotyped by allele-specific PCR or restriction fragment length polymorphism(RFLP)-PCR. Quantitative polymerase chain reaction by Taq Man■ assay was used to quantify TLR2 mRNA levels in fresh gastric tissues(48 GC, 36 CG, and 14 C).RESULTS Regarding the TLR2-196 to -174 polymorphism, the ins/del and del/del genotypes were associated with a higher risk of GC by comparison with the C in all of the analyzed inheritance models(codominant, dominant, recessive, overdominant and log-additive;P < 0.0001). Similarly, an increased risk was observed when comparing the GC and CG groups [codominant(P < 0.0001), dominant(P <0.0001), recessive(P = 0.0260), overdominant(P < 0.0001) and log-additive(P <0.0001)]. In contrast, TLR2 19216 T/C was associated with a protective effect in the GC group compared to the C group [dominant(P = 0.0420) and log-additive(P =0.0300)]. Regarding the association of polymorphisms with H. pylori infection,individuals infected with H. pylori and harboring the TLR2-196 to-174 ins/del polymorphism had an increased risk of gastric carcinogenesis [codominant(P =0.0120), dominant(P = 0.0051), overdominant(P = 0.0240) and log-additive(P =0.0030)], while TLR2 19216 T/C was associated with a protective effect[codominant(P = 0.0039), dominant(P < 0.0001), overdominant(P = 0.0097) and log-additive(P = 0.0021)]. The knowledge of the epidemiology of parasitic infections in stray and domestic animals, especially of its incidence and prevalence, is fundamental to adopting effective prophylactic measures. Stray dogs play an important role in environmental contamination favoring the transmission cycle of zoonotic agents. Among the parasitic infections that affect humans, Giardia duodenalis is the most common intestinal protozoa and was designated as a re-emerging infectious disease. This study aimed to determine the prevalence of G. duodenalis in dogs siezed by the Center for Control of Zoonoses (CCZ) of the city of Lages, Santa Catarina, Brazil using two diagnostic techniques. In 357 stool samples analysed, the prevalence of G. duodenalis cysts was 5.3% (19/357) and 4.8% (17/357) detected by floatation and sedimentation techniques, respecttively. No correspondence between gender and age was found among the methods used for analyzing the infected dogs in this study. Our data suggested that two diagnostic techniques should be used in a complementary way to ensure that false negatives are not neglected.

Electrochemical Evaluation of Aminoguanidine Hydrazone Derivative with Potential Anticancer Activity:Studies of Glassy Carbon/CNT and Gold Electrodes Both Modified with PAMAM

Aminoguanidine hydrazones (AGHs) are a class of compounds that have interesting pharmacological activities. They are derived from the same chemical group as aminoguanidine, so it has mixed properties (receptor and donor) in the formation of hydrogen bonds. Its anticancer agent properties were recently highlighted, but the molecules of this class have solubility in aqueous solutions that can be considered low. The identification of this class, by a simple, sensitive and low-cost technique, such as electrochemistry, which also allows the evaluation of its solubilization process through agents such as PAMAM dendrimer is the main objective of the work described here. The electrochemical response of the LQM10 (AGH derivative) was evaluated, as well as its behavior in different electrochemical sensors. Electrochemical experiments were performed in buffered (phosphate at pH 7.02 and acetate at 4.5). LQM10 has a reversible oxidation peak with a potential of +0.22 V. It was efficiently detected in different electrodes tested (glass carbon/CNT, glass carbon/CNT/PAMAM), which proves the viability of the electrodes for various analyses and has the determination of the apparent constant association, indicating its interaction with the analysis that is higher in the presence of the PAMAM encapsulating agent. This was corroborated by the results for the modified gold electrode with MUA and PAMAM. The sum of the results shows the possibility of electrochemically evaluating the Aminoguanidine hydrazone derivative, the viability of electrodes employed and the greater solubilization of LQM10 in the presence of the PAMAM dendrimer.

Comparison of Three Experimental Models for Rat Osteoarthritis Induction

Background: Osteoarthritis is a slowly progressive and debilitating disease with high prevalence in adult population. Knee is one of the joints most affected by this disorder. There are several models for animals’ osteoarthritis induction, however it is not identified any paper that compares these techniques. The present study was aimed to define the most appropriate model for rats osteoarthritis induction. Material and Methods: 40 Wistar rats were distributed into 4 groups of 10 animals each: normality group (NG);meniscectomy group (MG);quinolone group (QG) and iodoacetate group (IG). Radiographic images of the rat’s knees were analyzed as well as the amount of chondrocytes in the epiphyseal and articular cartilage. Results: In the radiographic analysis, there was a low correlation between the raters. Regarding the amount of chondrocytes in the epiphyseal cartilage, it was noticed that the IG and QG groups had fewer chondrocytes than NG, in contrast to MG that reported similar results to normality (p > 0.05). There was no significant difference between IG and QG groups (p > 0.05). Regarding the amount of chondrocytes in articular cartilage, it was noticed that the IG group showed fewer chondrocytes than NG (p 0.05). There was no significant difference between QG and MG groups (p > 0.05). Conclusion: Intraarticular injection of iodoacetate in rats is the model with greatest effect on reduction of chondrocytes amount.

Alterations in epigenetic control of gene expression play an important role in many diseases, including gastric cancer. Many studies have identified a large number of upregulated oncogenic mi RNAs and downregulated tumour-suppressor mi RNAs in this type of cancer. In this review, we provide an overview of the role of mi RNAs, pointing to their potential to be useful as diagnostic and/or prognostic biomarkers in gastric cancer. Moreover, we discuss the influence of polymorphisms and epigenetic modifications on mi RNA activity.
