Levels of Genotoxic.and c.rc.nogenic.Ingredients in Plant Food Supplem.nts and Assoc.ated Risk Assessm.nt

The present study desc.ibes the selec.ion, analysis and risk assessm.nt of genotoxic.and c.rc.nogenic.ingredients of botanic.ls and botanic.l preparations whic. c.n be found in food and plant food supplem.nts (PFS). First an inventory was m.de of botanic.l ingredients that are of possible c.nc.rn for hum.n health bec.use of their genotoxic.and/or c.rc.nogenic.properties. In total, 30 botanic.l ingredients were selec.ed and subsequently judged for their ac.ual genotoxic.and/or c.rc.nogenic.potential. Am.ng the 30 c.m.ounds c.nsidered, 18 c.m.ounds were judged to be both genotoxic.and c.rc.nogenic. Interestingly, the m.jority of these c.m.ounds belong to the group of alkenylbenzenes or unsaturated pyrrolizidine alkaloids. Subsequently, based on available c.rc.nogenic.ty data and estim.ted daily hum.n exposure that was determ.ned foc.sing on the intake from.PFS, the m.rgin of Exposure (m.E) was c.lc.lated for the alkenylbenzenes estragole, m.thyleugenol, safrole and β-asarone. c.lc.lating the m.Es for intake estim.tes of these alkenylbenzenes from.PFS resulted in m.E values that were generally lower than 10,000 and often lower than 100. In som. c.ses the m.E was even below 10 m.aning that the estim.ted daily intake is in the range of dose levels c.using m.lignant tum.rs in experim.ntal anim.ls. This result indic.tes that the use of PFS c.ntaining the genotoxic.c.rc.nogens estragole, m.thyleugenol, safrole or β-asarone m.ght raise a potential c.nc.rn for hum.n health and would be of high priority for risk m.nagem.nt.

目的吡咯西里啶生物碱(PyrrolizidineAlkodias,PAs)是天然存在于植物中的次级代谢产物,对人体具有肝毒性。调查发现,PAs广泛存在于植物源性农产品中,对人体健康造成潜在的危害。目前,已有600多种PAs从超6000种植物中鉴定出来,但因缺乏毒理学数据,制约了膳食暴露评估工作的开展。本研究以毛果天芥菜碱和瑞德林为研究对象,建立生理药代动力学模型(physiologic.lly based kinetic.m.delling,PBK)将毛果天芥菜碱和瑞德林的体外肝细胞毒性数据转化为体内肝毒性数据,并预测出其体内肝毒性阈值。材料和方法针对毛果天芥菜碱和瑞德林分别构建总体积为100μL的肝脏/小肠微粒体反应体系。经过对反应时间、底物浓度和微粒体蛋白浓度的优化,毛果天芥菜碱体外动力学反应体系包括:0.1 m.K2HPO4(pH 7.4),混合大鼠肝脏/小肠微粒体(0.04 m.protein/m.),毛果天芥菜碱(0-200μm.;瑞德林的体外动力学反应体系包括:0.1m.K2HPO4(pH 7.4),混合大鼠肝脏/小肠微粒体(0.5m.protein/m.),瑞德林(0-400μm.。加入20 m. NADPH启动整个反应,利用Ac.结束反应。UPLc.测毛果天芥菜碱和瑞德林的浓度变化,再根据米氏方程计算出两者的体外动力学参数。此外,通过文献查阅和基于定量构效(QSAR)的方法,获取大鼠的生理学参数以及毛果天芥菜碱和瑞德林的理化参数。基于获取的动力学参数、理化参数和大鼠的生理学参数建立PBK模型。结果通过与瑞德林的动物毒性实验进行对比,本项目建立的PBK模型预测出的瑞德林在小鼠体内的c.0}x值与动物实验所得到的c.0}x值只相差3倍。利用所建立的PBK模型,将毛果天芥菜碱和瑞德林的体外肝毒性数据进行外推,预测出的毛果天芥菜碱的Bm.L5值与实际动物实验只有1.9-2.9倍的差距,说明本研究所建立的PBK模型能准确的预测出PAs的体内肝毒性。结论和意义动物毒性实验具有耗时长、花销大等缺点,而传统的体外毒性实验存在高-低剂量外推、种间差异和无法模拟化合物动力学过程等诸多不确定因素,难以准确预测化合物的体内毒性。通过构建化合物的PBK模型,能将体外毒性数据进行外推,实现对化合物体内毒性的准确预测,对减少动物的使用和坚持3R原则具有重要的意义。
