Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. Evaluating methods to quantify sediment volumes trapped behind check dams,Saldana badlands(Spain)

The determination of sediment yield in catchments based on the sediment trapped by check dams is becoming a subject of *** fact,several methods have been developed in recent years to estimate the sediment retained by check *** complexity,precision and accuracy of each method vary *** this study,we evaluate the sediment trapped by check dams comparing the Sections method respect to the Prism,Pyramid,DTMs and Trapezoid *** analyzed a sample of 25 check dams(α-1=90%;ε=10%) in the Saldana badlands(Spain).The results showed that the Sections method offered a volume of retained sediment between the others,which gave an absolute variation from 22%to40%.The high variability of the check dam and sediment wedge sizes made necessary to compare methods in groups combining both *** significant differences in sediment volumes could be found between the methods for very small(height(h):1.8-2.3 m;trapped volume(V):6-102 m3) or large check dams(h > 2.3 m;V:165-387 m3),while significant differences are found for small(h nevertheless,volume differences between groups ranged up to + 25%.For these reason,the size of the check dam,the shape of the sediment wedge and the accuracy of the measure methods must considered when selecting an appropriate method to obtain the volume of retained sediment by check dams.A correct estimation of the sediment retention is needed to evaluate the role and efficiency of check dams in restoration projects or to estimate sediment yields.

Quantification of the Mixture of Hydrothermal and Fresh Water in Tectonic Valleys

This study was conducted to identify the origin, hydrogeochemical processes and evolution of groundwater in a tectonic valley. This study was carried out with the aim of quantifying the proportions of groundwater flows contributing to the water chemistry abstracted in a zone of convergence favored by the presence of active faults. The study area is located in the Trans-Mexican Volcanic Belt. End members methodology was applied to identify the mixing of hydrothermal with fresh groundwater, where changes in the aquifer geology result in distinct groundwater chemical signatures. Ternary mixing was quantified using conservative elements. Moreover, other evolutionary processes, such as ion exchange and silicate weathering occur due to changes in the geology of the area. In ternary mixing, each of the end members is associated with the lithology through which it circulates. The local flow contributes 70% of the water to the system, the intermediate flow contributes 14%, and the regional flow contributes 16%. Three types of water are produced: na-HCO3, due to the interaction of water with volcanic rocks of rhyolitic composition, na-Mg-HCO3, due to the interaction of water with volcanic rocks of basaltic-andesite composition, and Ca-HCO3, due to the interaction of water with sedimentary calcareous rocks.

Lagrangian Model PETROMAR-3D to Describe Complex Processes in Marine Oil Spills

The version 2.1 of PETROMAR-3D model, created in the Center for Marine Meteorology of the Meteorology Institute of Cuba, is presented. This Lagrangian model has been designed to describe the physical processes of marine oil spills in the face of multiple scenarios of the marine environment. Although it is applicable to any part of the world, it is mainly designed for inter-American seas. The novelty has been to integrate the processes of drift and weathering into a model, with updated methods that incorporate 3D phenomena, a very favorable situation to achieve an operating system in Cuba and the region for the immediate and medium term. Python was chosen as the programming language because it has advanced libraries for numerical modeling, automation work and other useful tools for pre-and post-processing. By means of adapters, an important number of atmospheric, hydrodynamic and wave models have been considered to create the scenarios efficiently. The modular distribution in which the code has been created facilitates its use for other dispersion analysis and biophysical applications. Finally, a set of simple images are presented, aimed at informing decision-makers in order to mitigate the effects of the spill on the environment.

Stratified Cox Regression Analysis of Survival under CIMAvax^(■)EGF Vaccine

Background: The Center of Molecular Immunology (CIM) is a center in Cuba devoted to the research, development and manufacturing of biotechnological products. CIMAvax?EGF is a vaccine for the treatment of non-small cell lung cancer patients (nSCL). Purpose: The aim of this work is to evaluate the effects of some potential prognostic factors on the overall survival of patients treated with CIMAvax?EGF vaccine, based on data collected in a phase II and a phase III clinical trials. Methods: The stratified Cox regression model is used to evaluate the effects of these prognostic factors, based on separate analysis for each trial, and on the combined data from both trials. Results: Patients with Performance status 0 or 1, with iv stage of tumor and male under 60 years obtain more benefit in terms of overall survival if they receive CIMAvax?EGF. Conclusions: Vaccinated group has a better performance if patients have a performance status 0 or 1, stage iv and age under 60 years. These prognostic factors influence overall survival in a positive way for those patients that received CIMAvax?EGF.

Background: 70% of ovarian cancer cases are diagnosed at an advanced stage (III or iv) of the disease and, in turn, with a high prevalence of peritoneal carcinosis and ascites, which leads to progressive malnutrition in patients, with the consequent deterioration of their general condition. There is a very important relationship between nutritional status, quality of life, survival, and the ability to tolerate multidisciplinary treatment of peritoneal carcinosis. Methods: A phase II, open-label, single-center, non-randomised clinical trial was conducted that included 36 patients with advanced disease who were administered the nutritional supplement Ocoxin, 30 ml twice a day, beginning one week before chemotherapy (CT) based on carboplatin/paclitaxel, of which they receive three cycles with neoadjuvant intent. Ocoxin treatment was continued during chemotherapy and for three weeks after completion of the last cycle, as well as during any periods for which this treatment was discontinued due to toxicity. The effect of Ocoxin on the quality of life was assessed through the QLQ C30 and QLQ OV28 questionnaires from the start of treatment until the end of the follow-up period. In addition, the Karnofsky Index and nutritional parameters were assessed. Results: There were no significant differences between adverse events versus baseline values, except in leukocytes, lymphocytes, neutrophils, ALT, and AST. There was no deterioration of the QoL scales, except for those related to the effects of chemotherapy and alopecia. Conclusions: Ocoxin as an adjuvant to chemotherapy appears to improve better tolerance to chemotherapy, showed a good safety profile, and improved quality of life. For further information on Ocoxin neoadjuvant therapy benefits, a phase III clinical trial will be needed.
