Background: Thalassemia is a prevalent condition characterized by decreased production or absence of hemoglobin globin chains. There are 2 types of thalassemia, alpha thalassemia and beta thalassemia (β-thalassemia), classified according to defects in the alpha or beta protein chain. β-Thalassemia results from a diverse range of mutations in the β subunit of the hemoglobin gene (HBB). This results in severe reductions in erythrocyte and hemoglobin levels. Worldwide, more than 40,000 babies are born with β-thalassemia annually. There are approximately 80 million β-thalassemia carriers. In Pakistan, the annual birth rate of β-thalassemia major carriers is approximately 6,000, and 50,000 to 100,000 patients suffer from this common inherited disorder. IVSI-1, IVSI-5 (G>C), Fr 8/9 (+G), Fr 41/42 (-TTCT), and del 619 are the 5 most common mutations that account for 90% of β-thalassemia cases in Pakistan. This study aimed to investigate the prevalence of β-thalassemia mutations (IVSI-5, Fr 41/42, and Fr 8/9) in the Swat district and to analyze the blood morphology of different mutant thalassemia blood ***: A cross-sectional study was performed at the Forensic Research Lab University of Swat from December 2021 to March 2022. The study included a total of 150 blood samples. Comparative analysis of blood morphology through microscopy was carried out on samples from patients with different β-thalassemia mutations and normal individuals. Genomic DNA extraction was performed according to the Sambrook protocol, and DNA was quantified via agarose gel electrophoresis. Thalassemia mutations were detected by amplification refractory mutation system (ARMS)-polymerase chain reaction (PCR). 胶原蛋白是一种纤维状蛋白质,存在于结缔组织的肌腱、骨骼、皮肤和软骨等细胞外基质中,为组织提供结构支持、弹性和强度。由于其独特的生物特性,胶原蛋白及其衍生物在食品、化妆品和生物医学行业有许多应用。2018年,全球胶原蛋白市场估计价值约42.7亿美元,预计到2025年将增长到66.3亿美元。然而,由于脯氨酸、甘氨酸和羟脯氨酸(其主要氨基酸)等疏水性氨基酸的存在,胶原蛋白不溶于水。由于其强疏水性,胶原蛋白的水解存在困难,因而迫切需求一种获得高产量胶原蛋白的方法。因此,本研究探讨了在不同的处理时间和底物浓度下,使用碱性蛋白酶提取、鉴定和水解牛白踝肌腱(BWAT)中胶原蛋白的方法。为了提取胶原蛋白,BWAT被切成较小的方块,在60℃下干燥并研磨。在这个过程中,将100 g磨碎的样品加入到1L的(1M)氯化钠溶液中,在40℃下连续搅拌4小时,以去除其他蛋白质和脂肪。然后将混合物过滤、冻干并储存,使用凯氏定氮法评估BWAT样品中的蛋白质浓度,通过SDS-PAGE凝胶电泳进行蛋白质鉴定。为了最大限度地提高肽的产量,样品在55℃下用不同浓度(1%-4%)的碱性蛋白酶处理不同的时间段(2-8h),以及底物浓度(1-4g)。在这项研究中,使用了生物化学以及生物物理技术来分析水解程度、肽谱和水解物的分子量分布,用分光光度计测量了340nm处的光密度。结果显示,BWAT含有大量的胶原蛋白。经氯化钠处理后,粗胶原蛋白产量为32.09%。此外,用凯氏定氮法测得到样品中蛋白质含量为87.11%。SDS-PAGE分析结果显示,获得粗蛋白的分子量在130-150 KDa之间,与文献中报道的一致,证实了BWAT样品中胶原蛋白的存在。水解度(DH)分析显示,在使用4%的酶浓度和处理4h的情况下,与1g(13.74±0.41%)、2g(14.97±0.45%)和3g(18.71±0.39%)处理相比,4g底物的DH最高(57.87±0.51%,P<0.001);并且不同的处理时间也会对DH产生影响,同样以1%的酶和2克底物处理时,2h与4h(41.63±0.31%),6h(16.46±0.44%)和8h(15.07±0.21%)处理相比。观察到最低的DH(7.34±0.37%),当底物浓度为1克,使用1%的酶处理4h,蛋白质回收率最低(32.96±0.46%);在2g底物和4%酶的恒定浓度下,与4h(39.73±0.32%)、6h(73.54±0.37%)和8h(75.23±0.21%)处理相比,2h处理的蛋白质回收率最高(89.63±0.63%)。总之,研究了从BWAT生成胶原蛋白肽的最佳酶促水解条件,并确定为酶浓度为4%,底物浓度为4g,水解时间为4h。

Using AdS/CFT correspondence, we analyze the holographic Einstein images via the response function of the complex scalar field as a probe wave on an AdS Schwarzschild scalar-tensor-vector gravity (STVG) black hole (BH). We find that the amplitude of the response function decreases with increasing values of coupling parameter alpha and increases with decreasing temperature T. The frequency omega of the wave source also plays a significant role in wave periods; as we increase the values of omega, the periods of waves decrease, indicating that the total response function closely depends on the wave source. Further, we investigate the optical appearance of the holographic images of the BH in bulk. We found that the holographic ring always appears with surrounding concentric stripes when the observer is located at the north pole, and an extremely bright ring appears when the observer is at the position of the photon sphere of the BH. This ring changes into a luminosity-deformed ring or a bright light spot as the observational angle changes. The corresponding brightness profiles show that the luminosity of the ring decreases and the shadow radius increases with increasing values of alpha. The relation between temperature T and the inverse of the horizon is discussed; T is small at the beginning of the horizon and then increases as the horizon radius increases. This effect can be used to distinguish the STVG BH solution from other BH solutions. Moreover, these significant features are also reflected in the Einstein ring and corresponding brightness profiles. In addition, we compare the results obtained by wave optics and geometric optics, which align well, implying that the holographic scheme adopted in this study is valid.

Climate and land use changes have a significant impact on the runoff generation process in urban environments, and these effects could get worse in the future. The combined contributions of these changes have increased the risk of ***, there is a need for integrated modeling to better understand the runoff variability, especially in small urban catchments. To quantify and separate the effects of land-use changes and climate change on the hydrological response of urban catchments with a distributed hydrological model(Storm Water Management Model, SWMM), this study introduces a new integrated approach based on the Machine Learning based land use change modeler and climate change scenarios under *** on supervised classification and land use change model analysis, accumulated impervious area increase from 22%(in2023) to 33%(in 2060) was observed in the study area. Furthermore, integrating this projected increase in imperviousness with future climate change into SWMM by considering three different scenarios i.e., S1(Climate Change), S2(Combined Land Use and Climate Change), and S3(Land use Change) resulted that climate change could cause an increase in runoff from 13.2% to18.3% in peak runoff and the contribution of land use could range from 9.1% to 18.6%. Similarly, in response to the coupled effects of climate and land-use change, the runoff would likely change from 24.53% to 39.66%. Conclusively, the study showed that despite climate change, intensive urban development by the substitution of impervious surfaces could also have a severe impact on the microclimate and hydrology of small catchments. Lastly, this study could provide a way forward for the future planning and management of water resources in small catchments which could be extended to larger catchments.

Malware is an ever-present and dynamic threat to networks and computer systems in cybersecurity,and because of its complexity and evasiveness,it is challenging to identify using traditional signature-based detection *** study article discusses the growing danger to cybersecurity that malware hidden in PDF files poses,highlighting the shortcomings of conventional detection techniques and the difficulties presented by adversarial *** article presents a new method that improves PDF virus detection by using document analysis and a Logistic Model *** a dataset from the Canadian Institute for Cybersecurity,a comparative analysis is carried out with well-known machine learning models,such as Credal Decision Tree,Naïve Bayes,Average One Dependency Estimator,Locally Weighted Learning,and Stochastic Gradient *** traditional structural and JavaScript-centric PDF analysis,the research makes a substantial contribution to the area by boosting precision and resilience in malware *** use of Logistic Model Tree,a thorough feature selection approach,and increased focus on PDF file attributes all contribute to the efficiency of PDF virus *** paper emphasizes Logistic Model Tree’s critical role in tackling increasing cybersecurity threats and proposes a viable answer to practical issues in the *** results reveal that the Logistic Model Tree is superior,with improved accuracy of 97.46%when compared to benchmark models,demonstrating its usefulness in addressing the ever-changing threat landscape.
