Weather manifests in spatiotemporally coherent structures.Weather forecasts hence are affected by both positional and structural or amplitude errors.This has been long recognized by practicing forecasters(cf.,e.g.,Tropical Cyclone track and intensity errors).Despite the emergence in recent decades of various objective methods for the diagnosis of positional forecast errors,most routine verification or statistical post-processing methods implicitly assume that forecasts have no positional error.The Forecast Error Decomposition(FED)method proposed in this study uses the Field Alignment technique which aligns a gridded forecast with its verifying analysis field.The total error is then partitioned into three orthogonal components:(a)large scale positional,(b)large scale structural,and(c)small scale error variance.The use of FED is demonstrated over a month-long MSLP data set.As expected,positional errors are often characterized by dipole patterns related to the displacement of features,while structural errors appear with single extrema,indicative of magnitude problems.The most important result of this study is that over the test period,more than 50%of the total mean sea level pressure forecast error variance is associated with large scale positional error.The importance of positional error in forecasts of other variables and over different time periods remain to be explored.

Changes in organellar gene expression （OGE） trigger retrograde signaling. The molecular dissection of OGE-dependent retrograde signaling based on analyses of mutants with altered OGE is complicated by compensatory responses that mask the primary signaling defect and by secondary effects that influence other retrograde signaling pathways. Therefore, to identify the earliest effects of altered OGE on nuclear transcript accumulation, we have induced OGE defects in adult plants by ethanol-dependent repression of PRORS1, which encodes a prolyl-tRNA synthetase located in chloroplasts and mitochondria. After 32 h of PRORS1 repression, the translational capacity of chloroplasts was reduced, and this effect subsequently intensified, while basic photosynthetic parameters were still unchanged at 51 h. Analysis of changes in whole-genome transcriptomes during exposure to ethanol revealed that induced PRORS1 silencing affects the expression of 1020 genes in all. Some of these encode photosynthesis-related proteins, including several down-regulated light-harvesting chlorophyll a/b binding （LHC） proteins. interestingly, genes for presumptive endoplasmic reticulum pro- teins are transiently up-regulated. Furthermore, several NAC-domain-containing proteins are among the transcription factors regulated. Candidate cis-acting elements which may coordinate the transcriptional co-regulation of genes sets include both G-box variants and sequence motifs with no similarity to known plant c/s-elements.

Genetic and molecular heterogeneity,together with intrinsic and acquired resistance to therapy,represent the major obstacles to the successful treatment of different types of breast carcinoma.increasing evidence demonstrates that SOX transcription factors in breast carcinomas could act both as oncogenes and tumor suppressors and have been associated with tumor stage and grade,poor prognosis,and therapy resistance.Both SOX2 and SOX18 overexpression has been correlated with poor prognosis in breast carcinomas,and these genes are recognized as potential antitumor targets.Our aim was to evaluate the effect of retinoic acid(RA),a well-known cyto-differentiating agent,on breast carcinoma cells in vitro and to investigate the potential of RA treatment to modify the expression of SOX2 and SOX18 genes.By applying various experimental approaches,we evaluated the effect of RA on basic cellular processes in SK-BR-3 and MCF7 breast carcinoma cell lines.We have shown that RA inhibits cell growth,reduces the number of Ki-67 positive cells,and causes cell-cycle arrest.RA effect was more prominent in SK-BR-3 cell line that lacks SOX2 expression,including a higher decrease in cell viability,reduction in colony formation,and significant remodeling of cellular structure.We have shown that RA treatment led to the downregulation of SOX2 expression in MCF7 cells and to the reduction of SOX18 expression in both cell lines.By functional analysis,we showed that the anti-proliferative effect of RA in both cell lines was not based on the activity of stemness marker SOX2,pointing to a SOX2-independent mechanism of action.The ability of RA to reduce SOX2/SOX18 expression raises the possibility that these genes can be used as biomarkers to distinguish RA-responders from non-responders.Together,our study shows that the response of breast carcinoma cell lines to RA treatment may vary,highlighting that the development of RA-based therapy should consider differences in breast carcinoma subtypes.

AiM:To evaluate the association between 25-hydroxyvitamin D[25(OH)D]and sustained virological response(SVR)in hepatitis C virus(HCV)infected individuals.METHODS:Relevant studies were identified by systematically searching MEDLiNE databases up to March2012 and abstracts of the European and American Congress of Hepatology conducted in 2011.Studies must provide information on SVR and the levels of 25(OH)D3and/or 25(OH)D2[henceforth referred to as 25(OH)D]in sera samples from HCV infected individuals.The inclusion criteria were:clinical studies that included HCV infected patients aged older than 18 years regardless of HCV genotype or ethnic group;provided information on SVR rates;and were reported in the English languageas full papers.Due to the heterogeneity of studies in categorizing serum vitamin D levels,a cut-off value of30 ng/mL of serum 25(OH)D was used.Heterogeneity was assessed using i2statistics.The summary odds ratios with their corresponding 95%Ci were calculated based on a random-effects model.RESULTS:Overall,11 studies(8 observational and 3interventional)involving 1575 individuals were included and 1117 HCV infected individuals(71%)showed low vitamin D levels.Most of the studies included monoinfected HCV individuals with the mean age ranging from 38 to 56 years.Four studies were conducted in human immunodeficiency virus/HCV infected individuals.Regarding vitamin D measurement,most of the studies employed radioimmunoassays(n=5)followed by chemiluminescence(n=4)and just one study employed high performance/pressure liquid chromatography(HPLC).Basal vitamin D levels varied from 17 to43 ng/mL in the studies selected,and most of the HCV infected individuals had genotype 1(1068/1575)with mean viral load varying from log 4.5-5.9 Ui/mL.With regard to HCV treatment,most of the studies(n=8)included HCV individuals without previous treatment,where the pooled SVR rate was 46.4%.High rates of SVR were observed in HCV individuals with vitamin D levels above 30 ng/mL(OR=1.57;95%Ci:1.12-2.2)and those supplemented with v

Artificial intelligence(Ai)is becoming popular for the Rate of Penetration(ROP)estimation,hence,the need to study the best techniques and their advantages over empirical models.Various literatures were analysed to determine the prevalence of Ai in ROP computation and compare the computation accuracies with empirical models.Artificial Neural Network(ANN)accounted for over 92%of the Ai techniques used for ROP computation and Weight on Bit(WOB)mostly influenced the computation accuracy.The accuracy of Ai algorithms is better than the empirical models thus,will improve the drilling efficiency,reduce cost and enhance the development of pad wells.

To address the issues of investment appeal in the Russian electric power industry,this study analyzes the dynamics of corporate governance,including permanent redistribution of property and compliance with the Russian Corporate Governance Code,in wholesale and territorial-generating companies.The increasing concentration of property in the hands of the state and its implications for investors are also noted.This study reveals the violations of essential principles,and the substantial differences in corporate governance practices,in the best-and worst-performing companies.Additional standards for better corporate governance practices to benefit the investors in the context of the current Environmental,Social,and corporate Governance(ESG)agenda are proposed.This study provides a new insight at the development of corporate governance in Russian power generating companies through property redistribution and compliance with corporate governance principles.

A novel implementation of negative stiffness elements(NSEs)is proposed,utilizing industrial grade nitrogen gas springs as pre-stressed stiffness elements in a configuration with lever arms.This NSE is combined with an inerter to form a stiff dynamic absorber(SDA)for vertical seismic protection of structures with base isolation.The SDA is optimized to minimize vertical accelerations while ensuring static structural integrity,excellent damping performance and containment of relative displacements.The introduction of gas springs in place of conventional linear springs addresses important practical limitations through features of non-linearity and industrial grade manufacturing.The proposed implementation is dimensioned for a 50-ton structure and evaluated numerically for 25 actual earthquake records,in comparison with a linear SDA model and an equivalent conventional damper(CD).individual and averaged results of acceleration and displacement time histories demonstrate vastly superior response compared to CD regarding induced accelerations for similar displacements.Performance equivalency with the linear SDA model indicates the stability of the gas spring implementation while guaranteeing predictability,tested endurance,proper tolerances,and off-axis motion resistance without requiring additional guiding components,as opposed to conventional springs.These features render the proposed implementation a promising solution for the realization of NSEs in seismic protection.
