Purification of enzymatically inactive peptidylarginine deiminase type 6 from mouse ovary that reveals hexameric structure different from other dimeric isoforms

The murine peptidylarginine deiminase (PAD) has five isoforms encoded by different genes and participates in a variety of cellular functions through the citrullination of target proteins. The crystal structure of human PAD4 with a dimeric form was previously solved because of the enzyme’s relevance to rheumatoid arthritis. PAD6, abundant in mouse oocytes and eggs, is believed to take part in early events of embryogenesis, but its biochemical properties are little understood. Here we have purified and characterized a recombinant PAD6. A PAD6 cDNA was cloned from mouse ovary RNA and expressed in Escherichia coli through pET29 and pGEX vectors. When benzoyl-L-arginine ethyl ester was used as a substrate, no appreciable activity was detected with a cell homogenate under conditions where a human PAD4 cDNA caused significant activity. Both proteins were affinity-purified to near homogeneity. The circular dichroism spectra of PAD6 and human PAD4 were similar in the far ultraviolet region. On molecular sieving, PAD6 was eluted faster than human PAD4. The cross-linking of PAD6 with dimethyl suberimidate clearly showed six bands on an sodium dodecyl sulfate-polyacrylamide gel. These results indicate that PAD6 can constitute a hexameric structure. The purified PAD6 still showed no enzymatic activity. This unique structure and loss in enzymatic activity is strongly suggested to favor the formation of egg cytoplasmic sheets as the architectural protein.

Ab Initio Two-Phase Molecular Dynamics on the Melting Curve of SiO_2

Ab initio two-phase molecular dynamics simulations were performed on silica at pressures of 20-160 GPa and temperatures of 2 500-6 000 K to examine its solid-liquid phase boundary. Results indicate a melting temperature （Tin） of 5 900 K at 135 GPa. This is 1 100 K higher than the temperature considered for the core-mantle boundary （CMB） of about 3 800 K. The calculated melting temperature is fairly consistent with classical MD （molecular dynamics） simulations. For liquid silica, the O-O coordination number is found to be 12 along the Tm and is almost unchanged with increasing pressure. The self-diffusion coefficients of O and Si atoms are determined to be 1.3×10^-9-3.3×10^-9 m2/s, and the viscosity is 0.02-0.03 Pa＇s along the Tin. We find that these transport properties depend less on pressure in the wide range up of more than 135 GPa. The eutectic temperatures in the MgO-SiO2 systems were evaluated and found to be 700 K higher than the CMB temperature, though they would decrease considerably in more realistic mantle compositions.

Counter-Rotating Type Tidal Stream Power Unit Boarded on Pillar(Performances and Flow Conditions of Tandem Propellers)

The authors have invented the unique counter-rotating type tidal stream power unit composed of the tandem pro- peUers and the double rotational armature type peculiar generator without the traditional stator. The front and the rear propellers counter-drive the inner and the outer armatures of the peculiar generator, respectively. The unit has the fixftful advantages that not only the output is sufficiently higher without supplementary equipment such as a gearbox, but also the rotational moment hardly act on the pillar because the rotational torque of both propel- lers/armatures are counter-balanced in the unit. This paper discusses experimentally the performances of the power unit and the effects of the propeller rotation on the sea surface. The axial force acting on the pillar in- creases naturally with the increase of not only the stream velocity but also the drag of the tandem propellers. Be- sides, the force vertical to the stream also acts on the pillar, which is induced from the Karman vortex street and the dominant frequencies appear owing to the front and the rear propeller rotations. The propeller rotating in close to the sea surface brings the abnormal wave and the amplitude increases as the stream velocity is faster and/or the drag is stronger.

Degradation mechanism of tin phosphide as Na-ion battery negative electrode

The degradation mechanism of an Sn_4P_3 electrode as Na-ion battery anode was investigated by using a transmission electron microscopic observation. At the first desodiation, we confirmed that Sn nanoparticles with 6 nm in size were dispersed in an amorphous-like P *** to this, we observed aggregated Sn particles with sizes exceeding 50 nm after the drastic capacity fading. The capacity fading mechanism was for the first time confirmed to be Sn aggregation. To improve the capacity decay, we carried out the two kinds of chargeàdischarge cycling tests under the reduced volume changes of Sn particles and P matrix by limiting desodiation reactions of Nae Sn and Na3P, respectively. The Sn_4P_3 electrode exhibited an excellent cyclability with the discharge capacity of 500 mA hg^(-1) for 420 cycles under the limited desodiation, whereas the capacity decay was accelerated under the limited sodiation. The results suggest that the Sn aggregation can be improved by the reduced volume change of the P matrix, and that it is very effective for improving anode performance of Sn_4P_3 electrode.

Characteristics of obese or overweight dogs visiting private Japanese veterinary clinics

Objective:To characterize obese or overweight dogs that visited private Japanese veterinary clinics located in humid subtropical climate ***:Dogs were categorized into four body condition score groups and five body size groups based on their *** logistic regression models were applied to the data.A Chi-squared test was used to examine whether the percentage of obese or overweight dogs differed between ***:There were 15.1% obese dogs and 39.8% overweight *** dogs were characterized by increased age and female sex,whereas overweight dogs were characterized by increased age and neuter status(P < 0.05).Peak probabilities of dogs being either obese or overweight were between 7 and 9 years of age,with the probabilities then declining as the dogs got *** example,in toy sized dogs,the probability of dogs being overweight increased from 33.4% to a peak of 55.1% as dog age rose from 1 to 8years ***,in medium,small and toy sized dogs,neutered dogs were more likely to be overweight than intact dogs,whereas neutered small sized dogs were more likely to be obese than intact small sized dogs(P < 0.05).Additionally,the percentages of obese or overweight dogs differed between the 10 selected breeds with the highest percentage of obese or overweight ***:By taking age,body size,sex and neuter status into account,veterinarians can advise owners about maintaining their dogs in ideal body condition.

Relation of the infrasound characteristics and the continuous steel bridge vibration modes generated by the vibration of moving heavy trucks

As heavy trucks pass over highway bridges, bridge vibration occurs and generates infrasound. General trucks in Japan with rear leaf suspension have whole body vibration （suspension spring vibration） frequencies of about 3 Hz. Also, the frequencies of the wheel vibration （tire spring vibration） are about 10-20 Hz. The continuous steel highway bridges with middle span length have vibration modes with the same phase in each span at the frequencies of about 3 Hz and also have those with the secondary mode shape at the frequencies of about 10-20 Hz. Truck vibrations and bridge vibrations are closely related. In this work, vibration tests are conducted using a heavy test truck for two cases of infrasound complaints in order to investigate the relation between the continuous steel bridge vibration modes generated by the vibration of moving heavy trucks and its infrasound characteristics. As a result of the examination, two types of bridge vibration modes are caused by the vibrations of a moving heavy truck. Moreover, the bending vi- bration modes with the same phase in each span have the most powerful infrasound pressure, since each span vibrates with the same phase. Two countermeasures, including viscoelastic damper at the end of the girders and extended deck method, are proposed to reduce the amplitude of bridge vibration and its infrasound.

Quantitative evaluation of deep convolutional neural network-based image denoising for low-dose computed tomography

To minimize radiation risk,dose reduction is important in the diagnostic and therapeutic applications of computed tomography(CT).However,image noise degrades image quality owing to the reduced X-ray dose and a possible unacceptably reduced diagnostic *** learning approaches with convolutional neural networks(CNNs)have been proposed for natural image denoising;however,these approaches might introduce image blurring or loss of original *** aim of this study was to compare the dose-dependent properties of a CNN-based denoising method for low-dose CT with those of other noise-reduction methods on unique CT noise-simulation *** simulate a low-dose CT image,a Poisson noise distribution was introduced to normal-dose images while convoluting the CT unit-specific modulation transfer *** abdominal CT of 100 images obtained from a public database was adopted,and simulated dose-reduction images were created from the original dose at equal 10-step dose-reduction intervals with a final dose of 1/*** images were denoised using the denoising network structure of CNN(DnCNN)as the general CNN model and for transfer *** evaluate the image quality,image similarities determined by the structural similarity index(SSIM)and peak signal-to-noise ratio(PSNR)were calculated for the denoised *** better denoising,in terms of SSIM and PSNR,was achieved by the DnCNN than by other image denoising methods,especially at the ultra-low-dose levels used to generate the 10%and 5%dose-equivalent ***,the developed CNN model can eliminate noise and maintain image sharpness at these dose levels and improve SSIM by approximately 10%from that of the original *** contrast,under small dose-reduction conditions,this model also led to excessive smoothing of the *** quantitative evaluations,the CNN denoising method improved the low-dose CT and prevented over-smoothing by tailoring the CNN model.

Successful laparoscopic splenectomy after living-donor liver transplantation for thrombocytopenia caused by antiviral therapy

Although interferon (IFN) based therapy for recurrent hepatitis C virus (HCV) infection after liver transplantation has been widely accepted, it induces various adverse effects such as thrombocytopenia, resulting in its interruption. Recently, concomitant splenectomy at the time of living donor liver transplantation (LDLT) has been tried to overcome this problem, but this procedure leads to several complications such as excessive intraoperative bleeding and serious infection. A 60-year-old female received LDLT using a left lobe graft from her second son for liver failure caused by hepatitis C-related cirrhosis. Six months after LDLT, she was diagnosed as recurrent HCV infection by liver biopsy. IFN monotherapy was started from 7 mo after LDLT and her platelet count decreased to less than 50 000/μL, which thus made it necessary to discontinue the treatment. We decided to attempt laparoscopic splenectomy (LS) under general anesthesia. Since intra-abdominal findings did not show any adhesion formations around the spleen, LS could be successfully performed. After LS, since her platelet count immediately increased to 225 000/μL 14 d after operation, IFN therapy was restarted and we could convert the combination therapy of IFN and ribavirin, resulting in no detectable viral marker. Inconclusion, LS can be performed safely even after LDLT, and LS after LDLT is a feasible and less invasive modality for thrombocytopenia caused by antiviral therapy.

Environmental Conditions Determining the Timing of the Lifetime Maximum Intensity of Tropical Cyclones over the Western North Pacific and Their Frequency of Occurrence

Environmental conditions determining the timing of the lifetime maximum intensities of tropical cyclones(TCs)are investigated for the TCs over the western North Pacific during the period *** results show that the land controls the timings of the lifetime maximum intensities in 42% of the TCs over this basin,indicating that accurate track forecasts are beneficial for TC intensity *** respect to other TCs that are not affected by the land(i.e.,Ocean-TCs),the timings of their lifetime maximum intensities are determined by multiple oceanic *** particular,interactions between TCs and cold-core eddies occur in a large proportion(nearly 60%)of Ocean-TCs at or shortly after the times of their lifetime maximum intensities,especially in strong TCs(categories 4 and 5),suggesting that a consideration of the above interactions is necessary for improving TC intensity forecasting *** addition,unfavorable oceanic heat content conditions become common as the latitude increases over 25°N,influencing half of the *** vertical wind shear contributes detrimentally to the atmospheric environment in 17% of the TCs over this basin,especially in moderate and weak *** contrast,neither the maximum potential intensity nor the humidity in the middle level of the atmosphere plays dominant roles when TCs turn from their peak intensities to weakening.
