RNA methyltransferase is responsible for transferring methyl and resulting in methylation on the bases or ribose ring of RNA, which existed widely but mostly remains an open question. A recombinant protein PH1948 predicting RNA methyl- transferase from Pyrococcus horikoshii OT3 has been crystallized. The crystals of selenomethionyl PH1948 belong to space group C2, with unit-cell parameters a=207.0 ?, b=43.1 ?, c=118.2 ?, β=92.1°, and diffract X-rays to 2.2 ? resolution. The VM value was determined to be 2.8 ?3/Da, indicating the presence of four protein molecules in the asymmetric unit.

目的探讨滤波反投影(FBP)、自适应统计迭代重建技术(ASiR)和基于模型的迭代重建算法(MBIR,商品名"VEO")三种重建技术对常规剂量胸部薄层CT图像质量的影响。方法应用能谱CT对15例成年患者行胸部增强CT扫描,扫描条件:100kVp,自动毫安,噪声指数15,螺距0.984∶1,球管转速0.4秒/圈。分别用FBP、50%ASiR(50%比例ASiR和FBP混合以降低噪声)和VEO三种重建算法对原始数据行0.625mm薄层重建,测量图像噪声及胸主动脉与背部肌肉的对比噪声比(CNR),并对3组图像分别进行质量评分,然后行对比分析。结果FBP、50%ASiR和VEO三组图像的噪声分别为24.30±3.55、17.11±2.55及11.69±1.74,50%ASiR和VEO组图像噪声分别较FBP组降低29.59%和51.89%(P均<0.01);胸主动脉与背部肌肉的CNR FBP、50%ASiR和VEO三组图像分别为10.56±3.05、15.15±3.88及21.69±5.62,50%ASiR和VEO组图像CNR较FBP组分别提高43.47%和105.40%(P均<0.01);图像质量主观评分FBP、50%ASiR和VEO三组图像分别为4.03±0.72、4.63±0.41及5.75±0.25,50%ASiR和VEO组图像较FBP组分别提高14.89%和42.68%(P均<0.01)。结论与FBP重建算法比较,在相同剂量条件下,50%ASiR和VEO能显著降低胸部CT图像噪声并提高图像质量;其中VEO重建算法降噪及提高图像质量效果更为显著。

目的探讨能谱CT(GSI)提高腹部增强效果的可行性。方法收集因病情需要先后接受腹部双期增强CT扫描的27例患者,于第1次均采用传统120kV单电压CT进行扫描(对照组),复查时采用GSI扫描(试验组),两次检查对比剂总量均为600mgI/kg体质量。于增强图像上腹主动脉、肝脏、胰腺、肾脏、下腔静脉、门静脉主干选取ROI进行分析,记录GSI图像上各器官相对于120kV单电压扫描的等价CT值时的平均单能量值(等价亚组),以及CT值提高50%(150%亚组)、100%(200%亚组)时的MEI,并记录所对应的噪声值,比较各亚组间单能量值、CT值与噪声值的差异,评估2名医师对MEI各亚组图像质量的主观评分一致性。结果等价亚组能量值(71.92±3.54)keV,图像噪声为(23.07±3.99)HU,与对照组比较,各器官CT值的差异均无统计学意义(P均>0.05);150%亚组能量值(57.39±2.77)keV,图像噪声为(35.30±7.56)HU,较等价亚组增高了53.01%;200%亚组能量值(49.94±2.72)keV,图像噪声为(44.01±8.59)HU,较等价亚组增高了90.77%。与对照组比较,150%亚组、200%亚组间单能量值、CT值与噪声值差异均有统计学意义(P均<0.01)。2名医师对试验组3个亚组的主观评分一致性均很好(P均<0.01)。结论采用GSI的单能量技术于72keV时可以达到常规120kV的腹部增强CT效果,57keV、50keV时增强效果可分别提高50%、100%。

目的探讨腹部CT单源瞬时kVp切换模式与常规CT120kVp扫描的射线剂量和图像噪声的关系。方法采用质量评价标准体模，对其中的水模部分分别进行能谱成像和常规螺旋扫描，获得常规扫描模式与能谱扫描模式mAs的回归方程，并计算出不同能谱成像（GSI）扫描参数所对应的常规120kVp扫描mAs。回顾性分析22例全腹部CT增强扫描患者资料。根据平扫常规扫描所使用的mAs选择相应的能谱模式进行增强门静脉期扫描，分别测量常规和能谱CT扫描在肝实质、膀胱区的图像噪声。采用配对t检验比较常规扫描与能谱扫描的射线剂量和图像噪声。结果22例患者CT能谱成像和常规螺旋扫描的CT剂量指数分别为（13．8±4．8）和（14．0±7．0）mGy，剂量长度乘积分别为（691．1±274．2）和（678．8±385．0）mGy·cm，差异均无统计学意义（t值分别为0．177和-0．260，P值均〉0．05）。在肝脏，65keV单能量图像的噪声为（7．6±1．4）HU，低于常规扫描滤波反投影（FBP）图像和20％自适应统计迭代重建（ASIR）图像[分别为（10．4±1．0）和（9．1±0．9）HU]，与40％ASIR图像相当[（7．8±0．7）HU]，高于60％ASIR图像噪声[（6．7±0．7）HU]；在膀胱，65keV单能量图像的噪声[（9．1±1．3）HU]低于常规扫描FBP图像[（10．5±1．3）HU]，与20％ASIR图像相当[（9．2±1．2）HU]，高于40％和60％ASIR图像的噪声[分别为（8．0±1．1）和（6．8±0．9）HU]。结论通过选择合适的参数，腹部CT能谱扫描与常规扫描的射线剂量相当；65keV单能量图像的噪声低于常规扫描的FBP重建图像、与20％～40％ASIR图像噪声相当，可在临床中常规应用。
