Flammulina velutipes is a very important edible fungus as it is rich in essential nutrients for ***,current molecular studies on this fungus rely on,knockdown and over expression of genes that provide limited information on gene function under specific conditions such as specific time point,growth and *** this case,inducible promoters would be ***,in current study,we selected a well established Tetracycline inducible promoter（Tet） system to use in *** developing of suitable inducible promoter system for ***,a plasmid was constructed by introducing reverse tetracycline transactivator（rt TA2-M2） gene behind Agaricus bisporus specific GPD promoter along with *** specific ***,Teto7 binding site was fused with the *** specific minimal GPD promoter（pmin）.Two reporter genes i.e Enhanced Green Florescence Protein（EGFP） and red florescent protein（d TOMATO）,were selected and cloned behind the teto7 fused with ***,a *** specific intron was introduced to stabilize the expression levels of Tet ***,a range of doxycycline concentrations（50,75,100,150,200,400ug/ml） were tested to establish an effective induction of the Tet system,in L11 strain without interference its growth.

比较分析草菇与其他真菌直系同源基因的关系

担子菌类的食用菌种类多、价值高、产量大,然而其产业的升级发展需要对食用菌生长发育相关生物学问题进行深入解析。目前多种食用菌完成了全基因组测序,然而作为非模式种其与模式丝状真菌间的直系同源基因目前尚缺乏全基因组水平的系统研究,在一定程度上限制了其分子生物学研究的深入。本研究以草菇为参照物种,将其与几种食用菌和模式丝状真菌进行两两直系同源基因分析,并对多物种间不同类型的直系同源基因进行功能富集。结果显示:一对一直系同源基因较多富集于基因复制、转录、翻译、修饰、加工等保守的基本功能类别;非一对一直系同源基因多属于基因家族,且包含了65%的转录因子,功能上富集在碳水化合物、脂质、氨基酸、次生代谢物及外源物质的代谢通路。无直系同源基因则较多富集在与基因重组、修复、信号转导相关的功能类别、特导性转录因子以及未知的预测基因。结果为食用菌分子生物学的深入研究提供有价值的参考。

磁场作为一种无法避免的自然因子,对生物的生长发育有重要影响。然而,目前磁场对大型真菌影响的研究还鲜有报道。本研究采用铷磁铁对金针菇菌丝进行处理,检测金针菇菌丝在不同磁极刺激下的生长和基因表达规律。结果显示:磁场是铷磁铁抑制金针菇菌丝生长的主要原因,且在磁感应强度0.003T以上对菌丝生长的抑制作用较强。通过同源比对获得一个金针菇磁受体和一个铁离子转运蛋白的编码基因,分别命名为ff-magr和ff-fief。定量PCR结果显示ff-magr在N极和S极均出现显著下调表达,N极和S极磁场对ff-fief的表达均无显著影响。结果表明:磁受体蛋白参与了金针菇菌丝受磁场抑制的应答过程,而磁受体不仅仅是通过调控铁的含量来调控菌丝的生长发育。以上结果为合理利用磁场调控真菌生长发育奠定基础,同时也为深入研究磁场对真菌生长发育的分子机理提供参考。

The goal of this study was to isolate and molecular identification of basidiomycetes species with laccase activity from soil and vegetative mycelial aggregations in different forest *** species were isolated from soil and mycelia,using different techniques,samples collected from five different geographic regions with dominant vegetation of Pinusspp.,Abiesspp.,and Quercus spp.,in state of Michoacan,*** activity of fungal isolates was analyzed by guaiacol *** number of basidiomycetes with laccase activity were isolated from mycelialagrigations,collected from Atecuaro（pinus and oak forest）,Michoacán,*** of laccase positive fungi was carried out by sequence comparison and phylogenetic analysis of internal transcribed region（ITS1F-5.8S-ITS4B） of r DNA genes with reference taxa.A total of fourteen strains（14） with high laccase activity of basidiomycetes were identified in thirteen genera i.e.,Bjerkandera,Corticiaceae,Coriolopsis,Echinodontium,Ganoderma,Hexagonia,Irpex,Limonomyces,Psathyrella,Peniophora,Phlebiopsis,Trametes and Trichaptum.

This paper investigates the effect of the circular economy on CO_(2)emissions growth by considering the role of energy transition,climate policy stringency,industrialization,and supply chain pressure from 1997 to 2020 using panel quantile Autoregressive Distributed Lags(QARDL)and the panel *** employ cointegration association in the long run among the variables,and the results of the two models confirm *** reveal that circular economy and climate policy stringency significantly negatively impact carbon *** the other hand,the energy transition,industrialization,and supply chain pressures are crucial to determining CO_(2)emissions in the short and long *** finding further explores that municipal waste generation recycling is considerable at the mean and upper 90th quantiles than the lower ***,the empirical results of the current study provide acumens for policymakers of advanced economies and emerging markets to maintain the balance among circular economy,energy transition,environmental policy stringency,and supply chain pressure for reducing CO_(2)emissions without halting economic growth and sustainable ***,practical implications are reported through the lens of carbon neutrality and structural changes.

This study aims to demystify the role of green energy and green technology in establishing the nexus between behavioural intentions of tourists,technologies,and digital payments by using Perceived value(PV),Compatibility(CO),Perceived Enjoyment(PE),and Social Influence(SI)as a predictor variables,Trust(TR)and Satisfaction(SA)as a mediating variables and Behavioural Intentions(BI)as an outcome *** the empirical estimation,we employ smart PLS-SEM,TAM(Technology Acceptance Model)and SPSS and Tested the LCC *** findings suggest that green energy and perceived value have the highest positive impact on tourists’trust towards digital payments followed by compatibility,social influence and perceived ***,tourists’satisfaction and green technology is one of the important determinants of choosing any digital mode of payment,is mostly influence by perceived value,perceived enjoyment,compatibility and social ***,if we choose between trust and satisfaction,trust plays a significant role in exploring the behavioural intentions of tourists about green energy and green technology followed by tourists’*** addition,Tourists’trust and satisfaction are highly correlated and influence each *** study offers novel policy implications in terms of use of green technology and green energy in enhancing trust and satisfaction of tourists in order to deeper understanding of different dimensions of digital payments and M-wallets,and allowing them to explore the long-term value inherent of digital payments and M-wallets.

Software testing is a critical phase due to misconceptions about ambiguities in the requirements during specification,which affect the testing ***,it is difficult to identify all faults in *** requirement changes continuously,it increases the irrelevancy and redundancy during *** to these challenges;fault detection capability decreases and there arises a need to improve the testing process,which is based on changes in requirements *** this research,we have developed a model to resolve testing challenges through requirement prioritization and prediction in an agile-based *** research objective is to identify the most relevant and meaningful requirements through semantic analysis for correct change *** compute the similarity of requirements through case-based reasoning,which predicted the requirements for reuse and restricted to error-based ***,the apriori algorithm mapped out requirement frequency to select relevant test cases based on frequently reused or not reused test cases to increase the fault detection ***,the proposed model was evaluated by conducting *** results showed that requirement redundancy and irrelevancy improved due to semantic analysis,which correctly predicted the requirements,increasing the fault detection rate and resulting in high user *** predicted requirements are mapped into test cases,increasing the fault detection rate after changes to achieve higher user ***,the model improves the redundancy and irrelevancy of requirements by more than 90%compared to other clustering methods and the analytical hierarchical process,achieving an 80%fault detection rate at an earlier ***,it provides guidelines for practitioners and researchers in the modern *** the future,we will provide the working prototype of this model for proof of concept.

Mating involvesthe transition from a self-sterileinto a fertile *** a prerequisite for the production of fruiting bodies,this is a fundamental step in the life cycle of mushroom forming *** mating,two genetically distinct monokaryons called mating types,will fuse and establish a *** compatibilityof mating types as well as the development of a dikaryonare controlled by independent gene clusters atthe MAT-A and the MAT-B *** MAT loci have been studied poorly for the majority of mushroom forming *** deepen our understanding of mating,we identified and mapped the structures of a series of MAT loci in the important edible mushroom *** results showed that the MAT-A and the MAT-B loci of *** each contained two separate,functionally redundantsubloci（Aa/Ab and Ba/Bb）.Depending on the interacting mating types,different MAT-A and MAT-Bsublociare predicted to control the mating *** analysis demonstrated that new combinations of MAT-Aa and MAT-Absubloci could be generated at a low frequency during ***,we demonstrated different combinations of MAT-Ba and *** means that *** can generate new mating types not only by recombining MAT-A with MAT-B loci（i.e.a typical tetrapolar mating type system）,but also can occasionally create new MAT-A and MAT-B mating *** resultsof our study provide important directions for further research on *** type genes of loci that were indicated to control mating in specific combinations are currently subjected to transformation assays that will confirm（or reject） their control over their respective A or B *** resulting mutants will be used to explore thedownstreamgenetic mechanisms of mating in ***.
