Rapid HPLC Method for Determination of Rebaudioside D in Leaves of Stevia rebaudiana Bertoni Grown in the Southeast of Mexico

Stevia leaves contain glycosides on which biological activity and sweetening capacity has been reported. Besides the main glycosides—stevioside and rebaudioside A—there are minor glycosides that may contribute to the activity and thus it is important to quantify them. Rebaudioside D is one of the minor glycoside present in S. rebaudiana leaves and there are no reports of a validated method to quantify it. Therefore a simple and sensitive high performance liquid chromatography (HPLC) method was validated for the determination of rebaudioside D in leaves of Stevia rebaudiana B. grown in the southeast of México. HPLC method was performed using a C18 column (250 mm × 4.6 mm, 5 μm) and UV detector set at 210 nm. The mobile phase consisted of 32:68 (v/v) mixture of acetonitrile and sodium phosphate buffer (10 mmol/L, pH 2.6), set to a flow rate of 1.0 ml/min. The calculated parameters were: sensitivity, linearity, limit of detection (LOD), limit of quantification (LOQ), accuracy and precision. The retention time of rebaudioside D was found to be 3.47 min ± 0.04 (S.D.). The calibration curves were linear over the working range (25 - 150 μg/ml), with correlation coefficient ≥0.99 and determination coefficient ≥0.98. The calculated limit of detection (LOD) was 8.53 μg/ml, while the limit of quantitation (LOQ) was 25.85 μg/ml. The percent recoveries of fortified samples were 100% ± 10% and precision relative standard deviation was ≤2.79%. The criteria of validation showed accuracy, linearity, and precision;therefore the method is suitable for quantitative analysis of rebaudioside D in Stevia rebaudiana leaves. Rebaudioside D content (g/100g) in Morita II and Criolla varieties grown in the southeast of Mexico were 0.43 and 0.46, respectively with no significant differences (p > 0.05) between them.

Soil Carbon Storage in Experimental Forest Plantations with Tropical Species

Forest ecosystems represent a third of the total organic carbon (TOC) stored in the soil. However, studies still need to be carried out to generate reliable information on its soil carbon storage potential in an experimental plantation. Therefore, the objective of this research was to determine the storage of total organic carbon (TOC) in the soil in two experimental forest plantations with tropical species, located in the state of Nayarit, México. The research was carried out in two experimental plantations with tropical species, located at the Santiago Ixcuintla Experimental Field (CESIX) and at the El Verdine?o Experimental Site (SEEV). The established species are Swietenia macrophylla King (mahogany), Cedrela odorata L. (cedar) and Tabebuia rosea Bertoil (pink trumpet-tree). In addition, data from dasometric variables were used in order to define if any of these variables had a similar variation to the soil TOC stock by species. The sampling was done under a randomized block experimental design in each experimental forest plantation (CESIX and SEEV). Nine composite samples were collected in the soil under the tree species at a depth of 0 - 30 cm, and 1.5 m distance from the tree. To determine the soil TOC, an elemental analyzer with a thermal conductivity detector Thermo Scientific (Flash 2000) was used. The results showed that TOC presented mean values of 103.60 and 219.98 t·ha-1 at a depth of 0 - 30 cm in CESIX and SEEV respectively. The highest TOC was obtained in soil under mahogany;142.95 t·ha-1 in CESIX and 284.41 t·ha-1 in SEEV. The analysis of the differences in TOC between the two plantations was highly significant. Methamphetamine and male suicide in the US-Mexico border region

Introduction: Studying the use of psychoactive substances in completed suicide is essential in order to understand its role in the suicide generating stimuli. The most commonly reported substances are alcohol and opioids. Method: This is a retrospective study of completed suicide database of the Forensics Medical office in the city of Mexicali from 1999 to 2005. This is the capital of the Mexican state of Baja California in the US-Mexico border region. Results: Out of 288 suicides, 260 were men, and the most frequent drug found on autopsy was methamphetamine (p < 0.001). The modal victim was a young male (20 - 39 years old), employed in blue-collar jobs, lived in an urban area, found in their homes, and died by hanging. Discussion: Although no causal effect can be drawn, our study results suggest that methamphetamine use may be a risk factor for suicide in this sample.

Downregulation of Nrf2 and HO-1 expression contributes to oxidative stress in type 2 diabetes mellitus: A study in Juana Koslay City, San Luis, Argentina

Oxidative stress is associated with diabetes mellitus, a condition characterized by increased prevalence and progression rate of cardiovascular disease. NFE2-related factor 2 (Nrf2) is a master regulator of cellular detoxification responses and redox status. The aim of this study was to examine associations between type 2 Diabetes Mellitus (T2DM), oxidative stress and the expression of NFE2-related factor 2 (Nrf2) in a population of diabetic patients living in Juana Koslay City, San Luis, Argentina. In addition, we evaluated the functional relevance of Nrf2 by measuring the HO-1 expression among persons with type 2 diabetes. We measured clinical and biochemical parameters related to lipid metabolism and oxidative stress in a population of Type 2 Diabetes Mellitus patients (T2DM, n = 40) and controls (Co, n = 30). Compared to Co, T2DM patients had higher fasting serum glucose, glycated hemoglobin, triglycerides, total cholesterol, low-density lipoprotein cholesterol, and thiobarbituric acid reactive substances and lower high-density lipoprotein cholesterol. T2DM individuals had also higher atherogenic index and body mass index than controls. We also founded that HO-1 mRNA in whole blood was lower in T2DM than controls, suggesting that T2DM may have an altered antioxidant response to oxidative stress. Interestingly, we found reduced Nrf2 mRNA in whole blood from T2DM compared to Co. The results from this study provide novel evidence that genes associated to antioxidant defense mechanisms are markedly reduced in patients with type 2 diabetes, and that the reduction in the expression of these genes could be associated to hyperglycemia and increased levels of MDA. Linear regression analysis revealed that there was a strong and positive correlation between the changes of Nrf2 and HO-1 expression levels.

Hyperglycemia promotes overexpression of SR-BII isoform of the scavenger receptor class B type I in type 2 diabetes mellitus: A study in Juana Koslay City, San Luis, Argentina

The scavenger receptor class B type I (SR-BI) is a high-density lipoprotein (HDL) receptor involved in reverse cholesterol transport. Some studies reported the association to be stronger in the presence of diabetes. The full length gene encoding SR-BI is comprised in 13 exons that are alternatively spliced to produce two major transcripts: the full length SR-BI and the splice variant SR-BII, in which exon 12 is skipped. Considering that type 2 diabetes status is characterized by changes in the concentration of plasma lipids, modifications in lipoprotein size and composition, which may be important modulators of the SR-BI expression;the aims of the study were to examine the influence of SR-BI polymorphism (rs838895) on lipid profile and SR-BI mRNA expression in a population of diabetic patients living in Juana Koslay City. Blood samples were drawn from controls (n = 40) and Type 2 diabetic patients (n = 66) and DNA and total RNA were obtained. SR-BI mRNA expression was measured by RT-PCR and SR-BI polymorphism was detected by Tetra Primer ARMSPCR. Compared to controls, diabetic patients had higher fasting serum glucose, glycated hemoglobin, triglycerides, total cholesterol, lowdensity lipoprotein cholesterol, and lower highdensity lipoprotein cholesterol. SR-BI mRNA expression was lower in T2DM when compared to controls, suggesting that the hyperglycemia presents in T2DM patients down-regulates SR-BI mRNA expression. Interestingly, we found that decreased SR-BI expression resulted in markedly increased plasma LDL concentrations in T2DM subjects, and the overexpression of SRBII isoform is responsible for the markedly increased plasma LDL-c concentrations. The polymorphism (rs838895) did not modify the mRNA level of SR-BI in leucocytes from control and diabetic patients. This study provides novel evidence suggesting that hyperglycemia may affect reverse cholesterol transport by controlling SRBI expression in diabetic patients. Phenotypical Aspects of a Familial Syndromic Retinitis Pigmentosa

Aim: To report a familial case of syndromic retinitis pigmentosa identified at Aristide Le Dantec Hospital in Dakar and to describe their clinical characteristics ophthalmic. Observation: We report a sibling group of nine children, four died at a young age from unknown causes. Three children were affected by retinitis pigmentosa, two cases were syndromic. A history of nyctalopia was found in all three affected children. The mean age of onset of decreased visual acuity was 6.6 years. Patient 1 affected by syndromic retinitis pigmentosa had an extraocular sign of cystic dilation of the main bile duct. Patient 2 had myoclonic epilepsy, psychomotor retardation, and the molar tooth sign on cerebral MRI (highly suggestive of Joubert syndrome). The third child had isolated retinitis pigmentosa. Ophthalmological examinations (fundus examination, electroretinogram, and visual evoked potentials) and pediatric examinations in the remaining two children were normal. Discussion and Conclusion: Retinitis pigmentosa is a rare degenerative disease that can be associated with several other malformations, highlighting the importance of screening for associated conditions. It presents a grim functional prognosis and a life prognosis dependent on extraocular manifestations. Molecular biology (karyotyping, next-generation sequencing) could have identified the implicated genes and allowed for a formal diagnosis and genetic counseling.

Vertical deformation model on postseismic phase using exponential and logarithmic function based on InSAR

The Palu MW7.4 earthquake occurred on September 28, 2018, with the epicenter at 119.86°, 0.72°. The severe shaking caused severe damage in Central Sulawesi, especially in Palu. We conducted a postseismic deformation study to determine the deformation pattern and reduce future earthquakes’ *** Synthetic Aperture Radar(In SAR) data were processed using Li CSBAS to get the time series. The time series data were fitted to exponential and logarithmic functions to determine the mechanism of postseismic deformation. The exponential model identified the influence of the viscoelastic mechanism, and the logarithm identified the afterslip mechanism. The Palu earthquake was fitted to logarithmic and exponential, but the logarithmic was more significant than an exponential *** mechanism predominates, and viscoelastic mechanisms play a minor role in this postseismic deformation.
