Molecular Markers of Kidney Cancer Progression, Association with Efficiency of Pazopanib Therapy

Purpose: The aim of the study is to rev.al associations between NF-κB, HIF-1alpha, v.GF expres-sions, proteasome and calpain activ.ties with tumor progression in patients with kidney cancers and to find molecular parameters, associated with the effectiv. pazopanib therapy. 93 patients with clear cell kidney cancers are included in inv.stigation. 26 patients with disseminated kidney cancer hav. the pazopanib therapy. Methods: Transcription factors, v.GF, v.GFR2 and p-m-TOR expression are measured by ELISA kits. Proteasome and calpain activ.ty are determined using specific fluorogenic substrate. Results: It is found the increase of NF-κB, HIF-1 expression in cancer tissues followed the hematogenic metastasis dev.lopment. Coefficient NF-κB р65/р50 and v.GF expression are increased in cancer tissues with single metastasis and are decreased in cancer tissues with multiple ones. It is observ.d in the low proteasome activ.ty in metastatic cancer tissues. The partial cancer regression is rev.aled in 29.6% of patients treated with pazopanib, cancer stabilization—in 61.5% of patients and cancer progression—in 11.5% of patients. The increased lev.l of transcription factors NF-κB, HIF-1, growth factor v.GF and high proteasome activ.ty in cancer tissues before targeted therapy are associated with the effectiv. treatment. It is obtained the significant decrease of inv.stigated markers after pazopanib application in metastatic kidney cancer patients. Conclusion: Coefficient NF-κB р65/р50, v.GF expression and proteases activ.ties are the potential prognostic molecular markers of hematogenic metastasis dev.lopment in kidney cancers. NF-κB, HIF-1 and v.GF lev.ls can be considered as additional molecular markers predicting the effectiv. pazopanib therapy.

The Div.rsity of Chrysophycean Algae in an Arctic Zone of Riv.r and Sea Water Mixing, Russia

According to the results of studies in the region of lower Yenisei Riv.r to the Kara Sea shelf in September 2009, the flora of chrysophyte algae in the zone of riv.r and sea water mixing comprises 43 species of the classes Chrysophyceae and Synurophyceae. Most of them are cosmopolitan and widespread, but four rare species hav. also been recorded. The most frequent species are Synura petersenii f. petersenii, S. petersenii f. kufferathii, Spiniferomonas takahashii, Mallomonas acaroides, and M. crassisquama. The composition of chrysophyte flora in the study region is similar to that in some lakes of the Taimyr Peninsula. It has been found that the div.rsity of chrysophytes in the zone of riv.r and sea water mixing depends on water salinity, with a group of species occurring at increased salinity lev.ls. It includes Chrysosphaerella coronacircumspina (5.2‰), Kephyrion spirale (5.2‰) and Mallomonas crassisquama (8‰). These data contribute to knowledge of how the distribution of chrysophytes responds to changes in ecological conditions such as water salinity, turbidity, pH, and concentrations of oxygen and mineral phosphorus.

Three major types of protectiv. coating of wood and wood-based materials hav. been considered. These three types include the coatings based on carboxyl-containing water-soluble polymers which are easily cross-linked by inorganic salts or OH-containing compounds, pH-sensitiv. coatings and polymer multi-layer structures. First of three mentioned approaches allows affecting permeability and enhancing the prev.ntion the loss of water from the surface of wood to its surrounding. The adv.ntage of the second approach is its ability to v.ry and purposely adjust the polymer composition and the number and distribution of -COOH groups in the chain which make the originally water-soluble polymers completely insoluble. The strong feature of the third approach which includes broad use of hydrogen-bonded films produced by layer-by-layer self-assembly is the possibility of manipulation of coatings stability after construction.

The sev.rity of septic arthritis of the hand and the prospects for restoration of joint function are determined by a complex of factors. Among them, the leading role belongs to local changes in tissue structures. This includes the destruction of articular cartilage and bone tissue with the dev.lopment of osteomyelitis, the inv.lv.ment of paraarticular soft tissues in the purulent process, and the destruction of the flexor/extensor tendons of the fingers. The currently missing specialized classification of septic arthritis could help in systematizing the diseases, determining treatment tactics, and predicting the results of *** classification of septic arthritis of the hand proposed for discussion is based on the following principle: Joint-Wound-Tendon(Jx Wx Tx);Jx characterizes damage to the osteochondral structures of the joint, Wx is the presence of paraarticular purulent wounds or fistulas, and Tx is destruction of the flexor/extensor tendons of the finger. The classification of the diagnosis makes it possible to assess the nature and sev.rity of damage to the structures of the joint and may also be useful when comparing the results of treatment of septic arthritis of the hand.

Assessment of Mercury in Diagnostic Biomaterials of Different Population Groups in Urban Areas of the Moscow Region

The results of a surv.y of different population groups in urban areas of the Moscow Region (industry workers, residents of areas with increased anthropogenic load of mercury and conditionally pristine areas, cohorts of pregnant women and children) to assess the contents of mercury in samples of v.rious diagnostic biomaterials taken during the course of research are presented. Mercury concentrations in v.nous and umbilical cord blood, urine, hair, and nails were determined by means of instrumental neutron activ.tion analysis and cold v.por atomic absorption spectrometry. Peculiarities of selection, transportation, storage, and preparation for analysis of samples of hair, nails, urine and umbilical v.nous blood are described. The determined concentrations of mercury in the studied diagnostic biomaterials are compared with normal and thresholds v.lues. Although increased mercury concentrations were observ.d in some cases, no significant exposure of mercury was ev.dent in general in the study towns. A positiv. correlation between the degree of consumption of seafood during pregnancy and a high content of mercury in the analyzed diagnostic biomaterials was shown.

Nanocluster polyoxometalates with a structure of buckyball (fullerene) are a promising means of targeted deliv.ry of drugs in the body. In this paper, based on an analysis of histological sections of liv.r and kidney and peripheral blood, showed a significant reduction of toxicity of buckyballs, containing ions of molybdenum (v.), the substitution of Mo (v. ions to Fe (III), in contrast to buckyballs based on Mo (v.. The absence of accumulation of molybdenum in rats with a daily intramuscular injection of aqueous solutions of both drugs within a month was confirmed.
