Color Doppler sonography and angioscintigraphy in hepatic Hodgkin's lymphoma

AIM： To estimate the characteristics of Color Doppler findings and the results of hepatic radionuclide angiography （HRA） in secondary Hodgkin＇s hepatic lymphoma. METHODS： The research included patients with a diagnosis of Hodgkin＇s lymphoma with metastatic focal lesions in the liver and controls. Morphologic characteristics of focal liver lesions and hemodynamic parameters were examined by pulsed and Color Doppler in the portal, hepatic and splenic veins were examined. Hepatic perfusion index （HPI） estimated by HRA was calculated. was observed. Lesions were mostly hypoechoic and mixed, solitary or multiple. Some of the patients presented with dilated splenic veins and hepatofugal blood flow. A pulse wave was registered in the centre and at the margins of lymphoma. The average velocity of the pulse wave was higher at the margins （P 〉 0.05）. A continuous venous wave was found only at the margins of lymphoma. There was no linear correlation between lymphoma size and velocity of pulse and continuous wave （r = 390, P 〈 0.01）. HPI was significantly lower in patients with lymphomas than in controls （P 〈 0.05）, pointing out increased arterial perfusion in comparison to portal perfusion. CONCLUSION： Color Doppler ultrasonography is a sensitive method for the detection of neovascularization in Hodgkin＇s hepatic lymphoma and estimation of its intensity. Hepatic radionuclide angiography can additionally help in the assesment of vascularisation of liver lesions.

Cytotoxicity of new Ho(Ⅲ) and Pr(Ⅲ) complexes

New complexes of coumarin-3-carboxylic acid(HCCA) with Ho(Ⅲ) and Pr(Ⅲ) were synthesized and their structures and spectral properties were investigated by elemental analysis,IR,Raman and NMR *** to the experimental data the complexes’ formula and geometries were *** frequencies,IR intensities and Raman activities as well as 1H NMR chemical shifts of HCCA and its Ho(Ⅲ) and Pr(Ⅲ) complexes were *** comparative experimental vibrational and NMR analyses of both the ligand and the Ln(Ⅲ) complexes predicted the bidentate binding to Ho(Ⅲ) and Pr(Ⅲ) through the deprotonated carboxylic oxygen and the carbonylic oxygen of the *** cytotoxic/cytostatic properties of the ligand and the newly synthesized complexes of coumarin-3-carboxylic acid with Ho(Ⅲ) and Pr(Ⅲ) were tested by MTT reduction assay against two mouse tumor cell lines:melanoma b16 and fibrosarcoma *** were also tested for cytotoxicity against normal mouse peritoneal *** proliferation inhibitory effect of the complexes compared to that of the ligand proved their cytotoxic/cytostatic properties against both the tumor cell *** addition,the complexes were less cytotoxic against normal mouse macrophages and were able to modulate NO release by activated *** obtained results were in accordance with our previously published data concerning the activity of lanthanide(Ⅲ) complexes with other coumarin derivatives.

In vitro studies of polyethyleneimine coated miRNA microspheres as anticancer agents

RNA plays important roles as a gene-silencing agent, a therapeutic agent for clinical treatment, and in the differentiation, proliferation, and development of cells. However, RNA is very difficult to work with due to its sensitivity and fragility. Another obstacle in using RNA for gene delivery/silencing is its negative charge, which causes its repulsion by cell membranes, which are also negatively charged. Our recent study showed that miR-125b is upregulated in glioblastoma （GMb） and plays an oncogenic role in GMb cells by promoting cell proliferation and inhibiting apoptosis. Endogenous miR-125b can be blocked by transfection of its antisense RNA molecule, miR-125b antisense （miR-125b-AS）. Thus, miR- 125b-AS can be developed as an RNA-based agent for cancer treatment. However, instability during storage and difficulty in delivery into cells has limited the use of RNA-based therapies thus far. In the current work, we demonstrate a short and simple one-step technique for the preparation of positively charged RNA nanospheres （miR-125b-RNS and miR-125b-AS-RNS） coated with a bioavailable polymer, polyethylenimine （PEI）. These RNA nanospheres are able to penetrate the cell directly without the use of liposomes. Our study confirmed that converting miR-125b and miR-125b-AS into nanospheres is a viable approach for storing RNA. In addition, this study provides evidence that PEI-coated RNA nanospheres have the potential to be used as a novel class of anticancer a^ents.

PCSK9 and triglyceride-rich lipoprotein metabolism

Pro-protein convertase subtilisin-kexin 9 （PCSK9） is known to affect low-density lipoprotein （LDL） metabolism, but there are indications from several lines of research that it may also influence the metabolism of other lipopro- teins, especially triglyceride-rich lipoproteins （TRL）. This review summarizes the current data on this possible role of PCSK9. A link between PCSK9 and TRL has been suggested through the demonstration of （1） a correlation between plasma PCSK9 and triglyceride （TG） levels in health and disease, （2） a correlation between plasma PCSK9 and markers of carbohydrate metabolism, which is closely related to TG metabolism, （3） an effect of TG-lowering fibrate therapy on plasma PCSK9 levels, （4） an effect of PCSK9 on postprandial lipemia, （5） an effect of PCSK9 on adipose tissue biology, （6） an effect of PCSK9 on apolipoprotein b production from the liver and intestines, （7） an effect of PCSK9 on receptors other than low density lipoprotein receptor （LDLR） that are involved in TRL metabolism, and （8） an effect of anti-PCSK9 therapy on serum TG levels. The underlying mechanisms are unclear but starting to emerge.

Social parasitic Hymenopterans have evolved morphological, chemical, and behavioral adaptations to overcome the sophisticated recognition and defense systems of their social host to invade host nests and exploit their worker force. In bumblebees, so- cial parasitism appeared in at least 3 subgenera independently： in the subgenus Psithyrus consisting entirely of parasitic species, in the subgenus Alpinobombus with bombus hy- perboreus, and in the subgenus Thoracobombus with b. inexspectatus. Cuckoo bumblebee males utilize species-specific cephalic labial gland secretions for mating purposes that can impact their inquiline strategy. We performed cephalic labial gland secretions in b. hyperboreus, b. inexspectatus and their hosts. Males of both parasitic species exhibited high species specific levels of cephalic gland secretions, including different main com- pounds. Our results showed no chemical mimicry in the cephalic gland secretions between inquilines and their host and we did not identify the repellent compounds already known in other cuckoo bumblebees.

The current bumblebee decline leads to inbreeding in populations that fosters a loss of allelic diversity and diploid male production. As diploid males are viable and their offspring are sterile, bumblebee populations can quickly fall in a vortex of extinction. In this article, we investigate for the first time a potential premating mechanism through a major chemical reproductive trait （male cephalic labial gland secretions） that could prevent monandrous virgin queens from mating with diploid males. We focus our study on the cephalic labial gland secretions of diploid and haploid males ofbombus terrestris （L.）. Contrary to initial expectations, our results do not show any significant differentiation of cephalic labial gland secretions between diploid and haploid specimens. Queens seem therefore to be unable to avoid mating with diploid males based on their compositions of cephalic labial gland secretions. This suggests that the vortex of extinction of diploid males could not be stopped through premating avoidance based on the cephalic labial gland secretions but other mechanisms could avoid mating between diploid males and queens.

Intrahepatic expression of genes related to metabotropic receptors in chronic hepatitis

AIM： To screen for genes related to metabotropic re- ceptors that might be involved in the development of chronic hepatitis. METHODS： Assessment of 20 genes associated with metabotropic receptors was performed in liver speci- mens obtained by punch biopsy from 12 patients with autoimmune and chronic hepatitis type b and C. For this purpose, a microarray with low integrity grade and with oligonucleotide DNA probes complementary to target transcripts was used. Evaluation of gene expression was performed in relation to transcript level, correlation between samples and grouping of clinical parameters used in chronic hepatitis assessment. Clini- cal markers of chronic hepatitis included alanine and aspartate aminotransferase, ~,-glutamyltranspeptidase, alkaline phosphatase and cholinesterase activity, levels of iron ions, total cholesterol, triglycerides, albumin, glucose, hemoglobin, platelets, histological analysis of inflammatory and necrotic status, fibrosis according to METAVIR score, steatosis, as well as anthropometric body mass index, waist/hip index, percentage of adi- pose tissue and liver size in ultrasound examination. Gender, age, concomitant diseases and drugs were also taken into account. Validation of oligonucleotide microarray gene expression results was done with the use of quantitative real-time polymerase chain reaction （qRT-PCR）. RESULTS： The highest （0.002 〈 P 〈 0.046） expres- sion among genes encoding main components of metabotropic receptor pathways, such as the a subunit of G-coupled protein, phosphoinositol-dependent pro- tein kinase or arrestin was comparable to that of an- giotensinogen synthesized in the liver. Carcinogenesis suppressor genes, such as chemokine ligand 4, tran- scription factor early growth response protein 1 and lysophosphatidic acid receptor, were characterized by the lowest expression （0.002 〈 P 〈 0.046）, while the factor potentially triggering hepatic cancer, transcrip- tion factor JUN-b, had a 20-fold higher expression. Analysis of Sheep Lymphocyte Chromosomal Aberrations after Exposition to Chlortetracycline

Tetracyclines are broad spectrum antibiotics and one type of various their compounds—chlortetracycline has been successfully used for our cytogenetic studies. We have selected for these studies the chromosomal assay because it is wellknown cytogenetic biomarker, which has been used to assess DNA damage at the chromosomal level. We analyzed the chromosomal aberrations and mitotic index in peripheral lymphocytes of sheep exposed by chlortetracycline chloride in pure tap water. To the food of the experimental group of animals (à n = 6) were added also 0.35 gof preparation Aureovit12 C80 plv. a. u. v. per kg b. w. and day (i.e. 168 mg of chlortetracycline hydrochloride/kg b. w.) daily in food. The frequencies of aberrant cells (AbC) in the experimental and control groups of sheep were stated to differ significantly (P = 0.001). In experimental group, chromatid breaks were the dominant type of chromosomal aberrations. No statistical differences in mitotic index values were found in the both groups (P > 0.05). Increased frequencies of chromosome aberrations in peripheral lymphocytes of sheep exposed by chlortetracycline in food, suggested a potential hazard which needs the attention from the viewpoint of human and animal health. This knowledge is useful for all, for the human and veterinary medicine, as well as for the aquaculture. Many of these chlortetracycline substances for their environmental persistence and chemical unstability are present in the environment for various periods, and as a result, they are responsible for several effects on human and animal health through food chain.

CRP, but not TNF-α or IL-6, decreases after weight loss in patients with morbid obesity exposed to intensive weight reduction and balneological treatment

Objective：The aim of this study was to evaluate the concentrations of C-reactive protein （CRP）, tumor necrosis factor-α （TNF-α）, interleukin-6 （IL-6）, and the degree of homeostasis model assessment-insulin resistance （HOMA-IR） in patients with morbid obesity exposed to a three-week low-calorie diet and balneotherapy. Methods：The study included 33 patients （25 females and 8 males; mean age 46 years） with body mass index （bMI） values of〉40 kg/m2. Evaluations of CRP, IL-6, TNF-α, lipid profile, HOMA-IR, and fasting glucose were carried out before （baseline data） and three weeks after the treatment. The control group consisted of 20 healthy volunteers （15 females and 5 males） with a mean age of 39 years and bMI values of≤24.9 kg/m2. Results：In the blood of patients with morbid obesity we found significantly elevated levels of CRP, TNF-α, triglycerides, HOMA-IR and fasting glucose, but a de-creased level of high density lipoprotein （HDL）-cholesterol, compared with the healthy individuals. The treatment resulted in about a 9.4%reduction in body weight from 122.5 to 111.0 kg and a significant decrease in the concen-tration of CRP, but no change in TNF-αor IL-6. HOMA-IR was significantly reduced. Conclusions：The decrease in CRP level without changes in TNF-α or IL-6 concentrations after the low-calorie diet and balneological treatment, suggests that an essential amount of adipose tissue must be removed before proper adipocyte function is restored. The decrease in HOMA-IR indicates an improvement in insulin sensitivity, which is beneficial in obese patients.

Random lasing was experimentally investigated in pyrromethene 597-doped strongly disordered chiral liquid crystals (CLCs) composed of the nematic liquid crystal SLC1717 and the chiral agent Cb15. The concentration of the chiral agent tuned the bandgap, and disordered CLC microdomains were achieved by fast quenching of the mixture from the isotropic to the cholesteric phase. Random lasing and band edge lasing were observed synchronously, and their behavior changed with the spectral location of the bandgap. The emission band for band edge lasing shifted with the change of the bandgap, while the emission band for random lasing remained practically constant. The results show that the threshold for random lasing sharply decreases if the CLC selective reflection band overlaps with the fluorescence peak of the dye molecules and if the band edge coincides at the same time with the excitation wavelength.
